The black bean (Phaseolus vulgaris L.) is one of the staples in the venezuelan diet. Productivity is seriously affected by the common bacterial blight caused by Xanthomonas phaseoli. Incorporating resistance genes through breeding programs is presented as an alternative control. In order to evaluate the resistance to common bacterial blight, black beans families F2:4 were studied, from crossing XAN-154 and MEM-0301013. The plants were inoculated with Tucutunemo strain, using the multiple needle method on protophylls. The variables evaluated, spot size (mm) and percentage of leaf area affected (% FAA) allowed us to study the disease advance. For the moment of maximum development of disease, 25 days after plants inoculation (DDI), the frequency distribution of the different degrees of reaction (scale from 1 to 9) was determined. The goodness of fit test (χ2) was performed, yielding that segregation observed corresponded to a single dominant gene in F2. Two families F2:4 (16 and 36) were found completely resistant, the father XAN-154 and genotype XAN-149 showed high resistance. Most families F2:4 showed high to moderate resistance. The resistance provided by the parent XAN-154, could be used as a source of monogenic resistance for the conformation of other populations in genetic breeding. There was evidence of the presence of polygenes for resistance possibly provided by both parents; these genes of smaller effect, could be used in future breeding programs.
The Delphacidae family is a group of insects that cause significant damage in cereals. Genetic diversity among six genotypes of rice (Oryza sativa L.), with different behaviors for resistance and susceptibility to mechanical damage from sogata (Tagosodes orizicolus) was studied by molecular SSR microsatellite markers, distributed in the twelve rice chromosomes.151 SSR were used for UPGMA clustering analysis and Dice distance. In the discriminant analysis, three groups were formed with cophenetic correlation of 0.87, the first constituted by Bluebonnet 50; in the second group were D-Sativa, Cimarron, Venezuela 21 and Zeta 15; and in the last group was Makalioka. Similarity coefficients of the groups were 0.55, 0.74 and 0.66 for each of the clusters. Clear separation was observed in the formation of groups between Makalioka and Bluebonnet 50 as resistant and susceptible genotypes, respectively. Molecular analysis showed a close genetic variability for genotypes. In determining the polymorphism between contrasting parents for mechanical sogata damage, the highest percentage 23.18% was observed between Zeta 15 and Bluebonnet 50 cultivars, followed by Cimarron and Bluebonnet 50 with 20.53%, and the lowest percentage for D-Sativa and Venezuela 21, and Venezuela 21 and Zeta 15, with 12.58% and 4.64%, respectively. These results will be used in the identification and association between SSR and characteristics related to mechanical sogata damage.
Black Sigatoka is one of the most damaging disease that attacks plantain cultivation. In order to evaluate the in vitro susceptibility of the fungus to horn plantain rachis ferments, a trial was conducted at the Universidad Nacional Experimental Sur del Lago (UNESUR) and the Centro Internacional del Plátano (CIPLAT), Venezuela. Media agar-water (16%) were prepared with rachis ferments, 2 and 4 months old, and two fungicides (propiconazole and benzimidazole), using six Petri dishes per treatment and five leaf samples of 2 cm² per dish, placed in a humid chamber for 48 hours, and incubated for 1 hour at 26 °C in darkness. Number of germinated and not germinated spores were counted, as well as, the number of short, distorted, long hyphae and length of long hyphae. It was determined the chemical and mineral composition of each extract. Data were analyzed using descriptive statistics making a comparison test of means (ANOVA). The results showed a lack of sensitivity of the fungus to different chemical fungicides used, while in the presence of ferments there was greater inhibition of germination and growth of the pathogen. Chemical analysis of the extracts showed the presence of most of nutrients, such as potassium, and saponins that can inhibit growth of the pathogen. These results demonstrated that the ferments of plantain rachis can control (Mycosphaerella fijensis Morelet) in vitro conditions, therefore their effectiveness should be evaluated under field conditions.
The rice (Oryza sativa L.), can be grown in waterlogged soil or not. The anatomical characterization of rice under two water management conditions was the main objective of this research in order to observe changes associated with adaptation to water stress. The cultivars 'Fundarroz PN-1', 'Cimarron', 'Fonaiap 2000' and 4 experimental lines were evaluated, transplanted in the field under two conditions, without stress (flooding) and stress (intermittent irrigation). Samples of leaves and roots of five plants at 42, 63, 88 and 95 days after transplantation were studied. Samples were dehydrated in tertiary butyl alcohol, embedded in paraffin blocks, cut and stained with Safranin-Fatsgreen technique. Observations and measurements were made with the program MOTIC 2.0. Collected data were: leaf and leaf central rib sizes, number of spaces, and numbers of vascular bundles (NVB) in the rib and blade, bulliformes cell numbers; in roots, number of xylem bundles, aerenchyma arrangement, and thickening of endodermis and thickness of the layer surrounding endodermis were measured. To identify the anatomical characters of greater variation among genotypes and irrigation conditions descriptive statistic was used. The NVB and thickening of the endodermis were the characters that showed greater variation, it was observed an increase of the NVB in first-and second-order stress conditions. Fonaiap 2000 and Somaclone 2 were the only genotypes that showed third-order vascular bundles. Changes in the thickening of the endodermis could be associated with tolerance to water deficit.
In participatory evaluations, farmers play a key role in cultivar selection because of their local knowledge and systemic perception of the crop. They actively integrate in the research process and synergy is achived with researchers to obtain technologies that really suit their conditions and needs. This study aimed to facilitate the participation of farmers in the evaluation and selection of cultivars of black beans (Phaseolus vulgaris L.), allowing the incorporation of new cultivars to local production systems. The study was conducted in the community of Santa Rosa in Ceiba Mocha, Guarico State. The methodology for open and absolute evaluation was applied and allowed the identification of criteria of local selection or preference of farmers and the acceptance or rejection of the 18 cultivars evaluated. The number of pods/plant, number of grains/pod and yield (Y) in kg ha-1 were determined. The cultivars with higher percentages of acceptance by farmers, corresponded to: Tacarigua (88%), Sesentera (75%), Local Guárico (75%) and Linea 13 (74%), using as important selection criteria, those related to Y, mainly the number of pods per plant, in addition to biotic resistance to pests and diseases. The results showed similarities with the evaluation of the technical team, hence the need for breeders to take into account the criteria and opinions of local farmers in the selection, as they are the potential users of such cultivars.
The andean blackberry (Rubus glaucus Benth), as well as, several wild species in Venezuela are found on hillsides located above 1.500 m. Despite of being an important fruit crop in the country, because of the nutraceutical properties and the great economical value of the activity for the producers, too little is known about its genetic variability and its vulnerability due to the loss of their natural habitats and pest and disease damages. The objective of this work was to characterize the genetic variability of a sample composed by 65 cultivated and wild Rubus genotypes collected in the states of Táchira, Mérida and Aragua. Microsatellite markers were used to characterize the genetic variability. A number of 16 microsatellite primers were evaluated, 6 of which were polymorphic, generating 18 polymorphic fragments with an average of 2.83 alleles per locus. The molecular analysis revealed a great similarity among the R. glaucus genotypes possible due to the use of few especific primers, the vegetative propagation of this crop and that genotypes were collected in nearby geographical regions. Participatory evaluation and selection of cultivars of Vigna unguiculata (L.) The participation of farmers along with breeders led to the evaluation and selection of cultivars, which can be more easily adopted as new varieties.
Sunflower (Helianthus annuus L.) cultivation in Venezuela is a suitable alternative to national oil production, which would contribute to overcome a long-time deficit in the country. Seed suply is a critical step in Venezuelan sunflower production, as it is imported; therefore, it is necessary to begin a national seed production program including sunflower breeding programs. The objective of this research was to characterize morphological and molecularly three segregating populations coming from three commercial hybrids, in order to determine its suitability as basic population in future selection schemes. Eighteen morphological variables were recorded on 120 plants coming from three segregating populations, and five RAPD primers were applied using DNA from 24 plants per population. Frequency distribution, mean tests, and principal components analysis revealed a wide morphological variability within each population. Plant height, number of ray flowers, and bract length were the most variable morphological attributes. Principal coordinates analysis based on 22 RAPD fragments also displayed a wide variability within populations. Genetic variability identified by morphological and molecular characterization made segregating populations, coming from commercial hybrids, good candidates to be used as basic population for breeding programs aiming to obtain national cultivars adapted to venezuelan production conditions.
Where prolonged drought is a major constraint for the success of food crops, cassava (Manihot esculenta Crantz) is an alternative under these conditions. However, despite its hardiness this crop is affected by the environmental extremes. Therefore, the aim of this study was to evaluate the physiological and agronomic potential of four promising clones of cassava in the Experimental Field of the National Agricultural Research Center (INIA-CENIAP), Maracay, Venezuela. The cultivars Bolívar 32, Concha rosada, CM 6740-7 y Per 183 were evaluated using a completely randomized design with factorial arrangement (4*9), corresponding to four cultivars of cassava and nine months evaluation, respectively, with three replications. The results indicated cultivars differences (P≤0.05) for number of leaves/plant, leaf area, plant height and petiole length. Leaf area (cm²), petiole length (cm), photosynthesis (μmol CO2 m-2 s-1), stomatal conductance (mmol CO2 m-2 s-1) and transpiration (mmol H2O m-2 s-1) showed a significant effect (P≤0.05) between month and clone interaction. With the principal component multivariate analysis, the four clones showed divergence among them. Finally, stomatal conductance, transpiration, photosynthesis and leaf number did not showed significant differences due to the small number of clones evaluated. Photosynthetic rate obtained for this crop presumed a C3-C4 metabolism.
The phenotypic stability analysis provides detailed information on the behavior of each genotype against environmental variations, in this sense, the objective of this research was to detect the presence of genotype by environment interaction as well as to know the adaptability and phenotypic stability of eighteen maize hybrids (Zea mays L.), by using the models index of superiority and bilinear regression. Twelve experimental hybrids and six commercial controls were planted in seven locations of Venezuela, during 2012 rainy season, in a randomized complete block design with three replications. The trait of interest was grain yield (t ha-1). Combined analyses of variance showed that hybrids performance were inconsistent because of environmental variations. With the model of index of superiority the hybrids 'DK 1596' and 'Exp A-1' were identifi ed as stable, while the model of bilinear regression allowed to identify the hybrids: 'Exp A-1', 'Exp A-2', 'Exp A-3', 'Exp A -5', 'Exp B-1', 'Exp B-4', 'Pioneer 30F35' and 'DK-370' as stable. With both models the hybrid Exp A-1 was identifi ed as stable whereas Exp B-4 as adaptable to favorable conditions. The model of bilinear regression was more discriminatory than the index of superiority; therefore, the fi rst one was more effi cient and suitable for the analysis of both adaptability and stability. The hybrids Exp A-1 and Exp B-4 showed a good productive performance in the different localities of evaluation, which should be considered for their release as commercial cultivars.
