The parasite infects naturally the snail, Pyrgophorus cf. spiralis (Guppy 1864), and the definitive host, Oreochromis mossambicus (Peters, 1852). Both molluscs and fish were collected from the Parque Litoral Laguna de los Patos, Cumaná, Estado Sucre, Venezuela. The cercariae were developed in redia in the digestive gland of molluscs and encysted on the water surface and more frequently on the bottom and walls of dishes. The 2-4 days old cysts of C. tilapiae were fed to laboratory reared O. mossambicus, and the trematode reaches sexual maturity in 22-32 days after the experimental infection. All developmental stages of C. tilapiae are described and illustrated. This species was compared with other members of Culuwiya and with Saccocoelioides tarpazensis Díaz and Gonzalez, 1990, for being the only recorded haploporids species in Venezuela and its life cycle had been elucidated and for the similarity with C. tilapiae. This species is redescribed herein represents the first record of Culuwiya Overstreet and Curran, 2005 in Venezuela.
Diseases transmitted through food products associated to fish consumption, actually constitute a serious sanitary problem in Venezuela, because gastrointestinal pathologies are more frequent, maybe originated directly from members of freshwater fish bacterioflora. For this, the objective of this research was focused in the enteropathogenicity determination, specifically citotoxicity and enterotoxicity of strains isolated from tilapias, rainbow trouts and cachamas, captured from farms and natural environments. In that sense, 12 strains were used, two of each of the following species: Aeromonas hydrophila, Escherichia coli, Klebsiella pneumoniae, Pseudomonas fluorescens, Plesiomonas shigelloides and Vibrio cholerae, using a Vero cellular line and the suckling mouse model. The enterotoxin production resulted positive for 42% of the strains, observing abdominal distension with a whitish intestinal fluid in mice inoculated with Aeromonas hydrophila, Klebsiella pneumoniae and Vibrio cholerae, while in those inoculated with E. coli the fluid was hemorraghic. All the strains resulted citotoxic in the trial with Vero cells, producing intracellular (toxic granulations) and extra cellular alterations (desegregation and changes in the cellular morphology). These results allow the inference that there is a relation between fish consumption and the appearance of disease in humans.
Helicobacter, Gram negative bacterias are curved or helicoidal microorganisms, widely spread in the stomach of humans and several animals, found as normal microbiota or causing of gastrointestinal pathologies. The objective of the work consisted of determining the relation between the presence and loads of helicobacterias in canine, with the degree of contact with its positive owners proprietors to these bacteria using rapid urease test. A sample of 31 healthy dogs of the city of Chillán, Chile were used, from which gastric fungus biopsies were obtained by endoscopy. These biopsies were put under the Rapid Urease Test. The owner of these canines were all positive for helicobacteria, determined by means of the same test. Ninety sevent percent of the canine biopsies were positive for helicobacteria. Thirty nine percent of the biopsies with the fastest positive reaction to the urease test belonged to canines with a high contact grade to their owners. The results suggest that the high charges of helicobacteria found in canines could be because of the higher contact grade of these animals with their owner. It would be important then to determine in future studies, whether if these pets could be a reserve or a transmission source for people, or viceverse, and the bacterial species involved.
A Cross-sectional and descriptive study was carried out in countryside of Puerto Boyacá town for identification of Cochliomyia hominivorax myiasis risk factors on cattle. Fourty four cattle farms were selected with a not-random sampling. Personal interviews were carried out to livestock farmers. Land use, livestock production system, and products for treatment of screwworm myiasis were investigated on February 6-15 of 2007. Stuff and instructions were supplied for taking larvae from animal wounds afterwards register survey application. Those materials were collect since last week on April of 2007 until first week on May of 2007. Frequency analysis, cross-tabs measures and non parametric tests were used. 49 screwworm myiasis cases (0.4%) out of 12,325 registered cattle were found, in 18 (41%) out of 44 selected cattle farms. The main cause of wounds recorded was injury by barbed wire with 16 cases (32.7%). Body sites more frequently infested were navel and scrotum with 9 cases (18.4%) each one. Association between age group in cattle and screwworm myiasis cases was founded (P=0.000; α=0.05). Association between cattle production system and screwworm myiasis cases was founded (P=0.044; α=0.05). Usual handling of non appropriate products for screwworm myiasis treatment like “creolina” (31 farms), gasoline (4 farms), used motor lubricant oil (2 farms), Baygon® (1 farm) and Vanodine® (1 farm) was registered. It may be concluded that selected cattle farms shared risk factors, depending on cattle production system. It demands epidemiological intervention to avoid damages to animals and consumers for using of inappropriate products for treatment of traumatic myiasis.
The performance of the following diagnostic tests: Milk Ring Test (MRT), Indirect Enzyme-linked Immunosorbent Assay (iELISA), Rose Bengal Test (RBT) and Competitive ELISA (cELISA) used for 5 years (2003-2007) in the epidemiological surveillance system for bovine brucellosis (Brucella abortus) in Machiques de Perijá County was evaluated. For this purpose, milk and sera samples previously assessed through the abovementioned tests were reassessed to define their true condition (true positive or true negative). Samples were considered as true positive through epidemiological findings and a positive result in Buffered Plate Antigen Test, which was confirmed through Fluorescence Polarization Assay (FPA); samples were considered as true negative only when they were taken from FPA-negative animals that did not seroconvert during the study. Results from tests in milk showed a higher performance of iELISA versus MRT, in sensibility: 98.55 and 56.52%; in specificity: 99.41 and 98.82%; in positive predictive value: 98.58 and 95.12%; in negative predictive value: 99.49 and 84.84%; and in coefficients of probability, respectively. The level of agreement between test and true status of samples was higher in iELISA: 97.8 versus 55.3%. Results from true positive and true negative samples warrant a reappreciation of the contribution of RBT in diagnostics, control and eradication of brucellosis in the studied area, taking into account its medium values of validity (169.92 versus 197.21) and accuracy (171.74 versus 197.02), as well as its low coefficients of probability. By contrast, cELISA showed an excellent diagnostic performance. Characterization of the immune response of bovines with mycobacterim bovis field infection in an area with high incidence of bovine tuberculosis in Venezuela http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S0798-22592009000500007&lng=en&nrm=iso&tlng=en La tuberculosis bovina es una enfermedad crónica, zoonótica, infecciosa y contagiosa teniendo como agente causal al Mycobacterium bovis inductor de una respuesta inmunitaria diversa.
The bovine tuberculosis is a chronic, zoonótic infectious disease and contagious having as causal agent to the Mycobacterium bovis inductive of a diverse immune response. This sandal, from an answer cellular, able to control the infection, happening through powerful, but a nonefficient one, humoral response to a state of not response or anergia, facilitating the dissemination of mycobacteria. In this study animals selected by the Venezuelan National Institute of Integral Agricultural Health (INSAI), like reactors to the simple test of the Tuberculina in a property with antecedents of tuberculosis by but of 20 years. To these animals the following tests were applied: Comparative of the tuberculina (PPD-B and PPD-A), test for Gamma Interferon (INF-y) and a Immun-enzimatic Test for TBC (ELISA-TBC), next was made the inspection post morten in the refrigerator where the compatible macrocospic injuries with tuberculosis were evaluated and classified. The selected weaves were used for bacteriological studies, DNA extraction and later amplification specific sequence with specific boots IS6110 for tuberculoso complex M., applying the test of Chain Reaction of Polimerasa (PCR-SSP) for the definitive identification of the pathogen. The result of these tests allowed to observe different patterns from immune response: cellular (PPD+, PPD+ - INF-y + or INF-y +), mixed (PPD+ and/or INF-y, ELISA-TBC+) and humoral (ELISA-TBC). Also anérgics animals detected themselves, negatives to all the immunological tests, positive in bacteriology and PCR-SSP. It was possible to be established the progression of the disease from the severity of the injuries and the age of the animals. These different immune responses may explain the persistence of infection in farm notwithstanding the implementation of the resolution of official control and eradication of Bovine Tuberculosis in the region.
The primary target of this study was to compare the use of an analog of sGnRH (Ovaprim®) with the carp pituitary extract (CPE) in the induced reproduction of Colossoma macropomum. Three treatments, one control, one with Ovaprim® (0.5 mL/kg) and one with CPE were evaluated (5mg/kg). The reproductive performance was evaluated in males, by means of the production and concentration of sperm, in the females, spawning ratio, absolute fecundity, relative fecundity, rate of fertilization and hatching. The Ovaprim® had an effect similar to the CPE in the production and concentration of semen of the injected males. The spawning ratio was similar (100%) in both treatments. A statistically significant (P<0.05) difference between the number of eggs obtained with Ovaprim® (155.6 x10³) and CPE (67.5 x10³). The best rates of fertilization and hatching were obtained in the group treated with CPE (63.26 and 68.48%) that with Ovaprim® (11.12 and 19.75%), respectively. These results indicate that the treatment with Ovaprim® is effective to induce the spawning, but produces eggs of low quality.
To evaluate the deep bedding system for the swine production in Cuba at small scale, it was carried out this experiment. Seventy two pigs (castrated males and females), YL (Camborough) cross, of approximately 21 Kg live weight and 75 days average age were used. The pigs were distributed into a random block design with two treatments (T1, deep bedding based on gramineous hay and T2, concrete floor). The animal performance traits (up to approximately 100 kg live weight) were studied. The animals of both treatments consumed a balanced feed based on soybean and corn meal and 30% of the wheat Cuban byproduct (byproduct of the wheat milling in the Cuban feeding industry), with enriched sugar cane molasses. There were significant differences for the consumption and the alimentary conversion (P<0.05) to the pigs housed in deep bedding system respecting to the pigs housed in concrete floor: 2.53, 2.74; 3.42 and 3.63, respectively. Significant differences were obtained (P<0.01) for the nitrogen and the phosphorous in the bedding material at the end of the experience: 1.79 and 0.58, respectively. There was not presence of Salmonella spp. neither helmints eggs. The sanitary quality of the bedding material, at the end of the study guaranteed to continue with a second production cycle. It was saved 177 m³ of water with this system. It was conclude that the evaluated deep bedding system for the swine production generates an important economic and environmental impact respecting to the conventional housing system and offers the main aspects to generalize this technology to the small and medium scale in Cuba.
The goal of this study was to evaluate the acrosomal integrity, biochemical functionality of the sperm membrane, and the relationship between them in boars with and without persistence of cytoplasmic droplets (PGC). It were used 254 ejaculates from 48 boars between eight and 48 months of age that were classified as young (<1 year), mature (1 to 3 years), and old (>3 years). Data were analyzed using analysis of variance for repeated measurements in the same individual (random effect). The effect of source (block effect) and age group on PGC, acrosomal integrity, membrane function, and proportion of live and with cytoplasmic droplets sperm cells were evaluated. The effect of PGC on acrosomal integrity and membrane function was also tested. Presence of PGC was lower in young (37 ± 6%) than mature (52 ± 5%; P=0.05) and old (85 ± 13%; P<0.01) boars. The difference in PGC between mature and old boars was significant (P=0.05). Old boars showed a higher proportion of cells with GC than young ones (34.6 ± 4.8 vs. 19.7 ± 2.3%, respectively; P=0.01), while mature boars were no different from the other two groups (23.5 ± 2.1%). Boars that did not show PGC had a higher proportion of intact acrosomes than those with PGC (86.5 ± 1.2 vs. 80.2 ± 1.1%, respectively; P<0.01). Defects in acrosomal integrity were significantly correlated with sperm defects associated to GC (between -0.20 y -0.36; P<0.05). These results suggest that membrane and morphologic defects tend to occur together in the same ejaculate, pointing out the need to build a multifactor seminal quality index that can be used as decision criterion in the swine industry. Encuesta exploratoria en el mercado mexicano sobre las características de músculos de cerdos originarios de Estados Unidos y México http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S0798-22592009000500011&lng=en&nrm=iso&tlng=en Trends in variation of physical, chemical and sensory traits due to country of origin (COO) were evaluated for samples from United States of America (U.S.A.) and Mexico of three pork muscles, Rectus femoris (RF, n = 20 from each COO), Vastus medialis (VM, n = 20 from each COO), and Longissimus dorsi (LD, n = 10 from each COO) in two separate, small-scale, exploratory surveys conducted at Mexico City. Compositional, physical and chemical properties and consumer acceptability traits of these Mexican and U.S.A. pork samples were quite similar, though Mexican pork samples generally were more variable. LD samples from U.S.A. had greater (P<0.05) water-holding and emulsifying capacities whereas both RF and LD from U.S.A. required lower shear force (P<0.05) compared to Mexican counterparts. Ratings from consumers did not indicate preference for pork from any of the countries. Because of the limited number of observations for the samples surveyed these results are preliminary and may not adequately characterize the populations of each country, but they did reveal important trends for selected traits of Mexican and U.S.A. pork currently available at the local market.
Bull spermatozoa express receptors for platelet-activating factor http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S0798-22592009000500012&lng=en&nrm=iso&tlng=en Platelet-activating factor (PAF; 1-O-Alkyl-2-acetyl-sn-glycero-3-phosphorylcholine) is a ubiquitous phospholipid that is implicated in the mediation of a wide variety of reproductive processes. To better understand the role of PAF in bovine reproduction, it was designed experiments to: (a) determine whether bull spermatozoa express receptors for PAF and (b) study the effect of exogenous PAF on in vitro sperm physiology (i.e., capacitation, acrosome reaction, motility, and fertilizing ability). Bull sperm express PAF receptor as determined by two approaches: RT-PCR and immunofluorescence. However, exposure of spermatozoa to different concentrations of exogenous PAF (10-11-10-6 M) did not affect capacitation, acrosome reaction or motility. Consistent with these findings, coculture of gametes in medium containing increasing concentrations of PAF (1 x 10-8-8 x 10-6 M) did not improve in vitro fertilization outcome as measured by percentage of inseminated oocytes reaching 2-cell stage 48 h after fertilization. In contrast, PAF at 8 x 10-6 M concentration significantly inhibited IVF. In conclusion, although bull sperm have PAF receptors, exposure of bull spermatozoa to exogenous PAF failed to enhance the sperm function parameters measured in this study. Additional studies are warranted to elucidate the biological role of PAF on bull spermatozoa.
Data from 1097 lambs’ birth weights (BW), 543 lambs’ weaning weights (WW) and 543 preweaning daily gain weight (DGW) were analyzed in order to compare preweaning performance of three breeds Pelibuey (P), Dorper (D), Katahdin (K) and two crosses (PxD, PxK). The variables studied were from a sheep farm at Huimanguillo, Tabasco, México and were collected from January 2001 until April 2005. The traits included lambs’ birth weights, lambs’ weaning weights and preweaning daily gain weight. The final model which describes BW included the fixed effects of birth year (BY) (five classifications, 2001 to 2005), breed group (P, D, K, PxD and PxK) within the birth year BG(BY), birth type (BT) (single and twin), interaction BYxBT, lambs sex (female and male) (SX), birth season (S) (dry, rainfall and rainfall and winds). Weaning age (WA) and BW were as considered as a continuous independent variable. With the exception of S, all the main effects affected (P<0.01) BW, WW and DGW. The interaction BYxBT affected only BW (P<0.01). Lambs PxD showed higher WW (P<0.01) than Pelibuey bred in years 2001 and 2002. DGW was higher in this group than P group in year 2001. The rainfall season was the worst in WW, with 15.4 kg. Single lambs and male lambs had better performance than twin lambs and female lambs. Weaning age and birth weight affecting WW. Regression coefficients were 0.023 ± 0.005 and 1.023 ± 0.315, respectively. In conclusión that under the present conditions, in the preweaning period, the hybrid breed group seems to be less important than others factors (for example: the weaning age, birth season and birth weight) as determinants of the weight at weaning. The Dorper and Katahdin breeds offer a viable alternative, considering the weight of the lambs at weaning in order to include them in systems of production in a humid, hot climate, such as pure breds or a Dorper x Pelibuey crossbreed.
ABSTRACT The consumption of shellfish has been associated with microbial infections even in cases where shellfish complied with the current regulation, which is based on bacterial analysis. Depuration processes try to eliminate microorganisms using seawater to allow living, filter-feeding shellfish to naturally purge themselves from agents they accumulated from the environment. In this study, depuration rates of potential indicators were estimated. Four experiments, with naturally-contaminated shellfish (Polymesoda solida), were performed. For evaluating the shellfish depuration process, total coliforms (TC), fecal coliforms (FC), fecal streptococcus (FS), enterococcus (EN) and mesophilic aerobic bacteria (MAB) were evaluated as bacterial indicators. Total inorganic content was used as physical indicator. Artificial prepared seawater of the depuration tank was disinfected by UV irradiation. Depuration removal rates of experiments running for 120 hours (five days) at 28°C, 5 psu, in a 150 L tanks were effective (80%) and more efficient during the first 72 hours, allowing an adequate bacterial and physical quality for consumption after this time, no matter the clams were collected in contaminated areas which do not complain with maximal allowable level.
