The begomovirus-Bemisia tabaci (Gennadius) complex is the main sanitary problem of tomatoes in Venezuela. Genotypes resistant to begomoviruses are a valuable alternative for integrated management of this problem. Consequently, it was assessed the performance of the cultivars Rio Grande (CRG), Alba El cid and Shan TY, the first one susceptible and the other three promoted as resistant to TYLCV. This virus, along with ToVEV, are two common tomato begomoviruses in Venezuela. Transmission assays mediated by B. tabaci were conducted to assess time for symptoms appearance and transmission efficiency. CRG showed TYLCV symptoms after 12.6 ± 1.2 days post inoculation (DPI) in 100 % of the plants, while for the remaining cultivars it ranged from 14.1 ± 1.3 to 16 ± 1.5 DPI with 18.5-33.3 % of symptomatic plants, generally with mild symptoms. With ToVEV for CRG, symptoms appeared at 12.4 ± 1.2 DPI with 100 % of affected plants, and for the other cultivars more than half of the plants (ranging 66.7 to 81.5 %) showed symptoms at 12 ± 1.1 to 14.9 ± 1.4 DPI. El Cid showed the most severe symptoms. In all plants exposed to begomovirus, viral DNA was detected by PCR. The resistance to a begomovirus can provide some resistance to another but not to all of them. However, those that showed tolerance could be utilized to reduce the effect of these begomoviruses on tomato crops in Venezuela.
Due to apomictic condition, garlic shows a vegetative reproduction, thus genetic breeding is limited to clonal selection and search for variability by procedures such as somaclonal variation or induced mutagenesis. After clone or mutant with desired characteristics is obtained, maintenance or preservation in germoplasm banks should be considered. In this regard, analyses by RAPD technique was made for evaluating genetic stability in garlic microbulbs clone ‘Boconó’ conserved in vitro or to detect presence of genetic variations in materials irradiated with gamma. During conservation, microbulbs were stored in five media: T1 = MS + sucrose 45 g·L-1, T2 = ½ MS + sucrose 45 g·L-1, T3 = ½ MS, T4 = ¼ MS + sucrose 45 g·L-1 and T5 = ¼ MS. Mutagenic treatments consisted in irradiance of microbulbs with gamma at 0, 6, 8 and 10 krad. Out of 12 primers used, 10 and 9 yield 62 and 71 polymorphic bands in treatments related to conservation and mutagenesis, respectively. Results showed that ¼ MS and ½ MS medium, both with sucrose, can be used to conserve microbulbs with genetic stability for 210 days, while the application of 8 or 10 krad gamma rays promoted changes or genetic variations in the garlic microbulbs.
Renewal of cocoa crops must be done with strong seedlings to guarantee field transplant survival, which is highly associated with root development in the first months of plant growth. We aimed to evaluate the effect of substrates inoculated with microorganisms in the production of cocoa plants during nursery stage. Seeds of clon IMC-67 were planted in bags. A completely randomized design was used, consisting in five treatments with sixty replicates each, one bag per plot. Treatments were: (T1) absolute control, 1:1 sand:soil; (T2) bioabono, 2:1:1 soil:bioabono:sand; (T3) Commercial mixed inoculant, 2:1:1 soil:vermicompost:sand; (T4) Cocoa rhizobacteria, 2:1:1 soil:vermicompost:sand; and (T5) chemical control, 2:1:1 soil:vermicompost:sand + chemical fertilization. Plant height, root length, number of leaves, stem diameter, shoot and root fresh and dry weight, as well as the microbial populations in SRS agar were measured. The analysis showed significant differences (P≤0.05) between treatments inoculated with microorganisms (T3 and T4) and those that were not inoculated (T1 and T5) in most of the variables. Therefore, the inoculation of these microorganisms is recommended to improve cocoa plant development in nursery stage, and in turn, ensuring the survival after field transplant. Substrate inoculation also promoted the increase of rhizobacteria populations.
Charcoal rot of soybean, caused by Macrophomina phaseolina (Tassi) Goid, is an economically important disease causing damage of varying magnitude in soybean, and shows high incidence and severity when prolonged periods of hot, dry conditions exist. It is known that Trichoderma spp. is a fungus that can parasitize other fungi. Similarly, the bacterium Rhizobium sp. can control the attack of pathogens. A research was conducted to study the effect of soybean inoculation with strains of Trichoderma sp. and native Rhizobium sp. in presence of M. phaseolina. To do this, experiments under field and shadehouse were performed, and the data were analyzed by univariate and principal components. Simple and combined treatments of Trichoderma and Rhizobium resulted in higher values than the control in terms of plant height and biomass, and nodule production. Inoculation of soybean seeds with strains of these microorganisms at the time of planting is a practice that increases plant vigor and protects from infection by M. phaseolina.
The implementation of plant tissue culture of Jatropha curcas encounters many problems such as the browning caused by phenolic oxidation. Therefore, a rapid methodology for comparison of treatments that can prevent this oxidation of the plant tissue is required. The determination of the enzyme polyphenol oxidase (PPO) activity could be used for this purpose due to the association between browning and the activity of the enzyme in J. curcas. In the present investigation the effect of sodium hypochlorite (NaClO) as a disinfecting agent on PPO enzyme activity in leaf and petiole explants of INI-3 and INI-6 genotypes of the species was evaluated. The results showed that the NaClO caused an increase in the PPO activity of leaves and petioles in both genotypes, thus indicating that the disinfectant is an indirect agent that can promote tissue browning in the plant. In addition, significant differences of PPO activity were found in both explant types and genotypes.
The diversity of plants in tropical regions is very important because this biota helps maintain the stability of the ecosystem, while many of them have economic importance. In this study, floristic and structural aspects of two thorny semiarid ecosystems were characterized in San Francisco, Torres Municipality, Lara State, Venezuela. Sampling was made from April 2010 to January 2011. Five 5 x 10 m-subplots were established to cover a total area of 250 m² in each location. All individuals with diameter at breast height greater or equal to 2.5 cm were sampled at each subplot. Thirty seven species included in 32 genera and 20 families belonging from Magnoliopsida were found. Families with the higher number of species were: Leguminosae sensu lato with 8 species, Cactaceae with 7 species, Capparaceae and Euphorbiaceae with three species each, Malvaceae and Rubiaceae with two species each. Results indicated that in San Francisco the predominating diametric class varied from 10.01 to 15.0 cm (DBH), representing 28 % of the total number of individuals, while in Santa Rosa, varied from 15.01 to 20 cm DHB representing 23.91 % of the total number. The species with higher IVI in San Francisco were Subpilocereus repandus, Mimosa arenosa and Malpighia emarginata, while in Santa Rosa were Croton argyrophyllus, Pilocarpus goudotianus and Aspidosperma cuspa. Results showed that both studied areas have low diversity values. Based on altimetric classes we can state that predominat vegetation cover is a xeric thorny and a low scrub in San Francisco and Santa Rosa, respectively.
Due to the intensive use of pesticides in agriculture, the presence of residues of these substances in food products, as well as the human health risk that they involve, seem to be inevitable. The objective of the present study was to determine the presence of pesticide residues in strawberry (Fragraria x ananassa), cultured in the Municipality of Rivas Dávila of Mérida State, Venezuela. For the analysis, the SPE-HPLC-DAD method was used. Seven pesticides were detected: chlorpyrifos 8 mg∙kg-1, diazinon 12 mg∙kg-1, ethyl parathion 0.036 mg∙kg-1, methamidophos 2.7 mg∙kg-1, mancozeb 23 mg∙kg-1, methomyl 1.2 mg∙kg-1 and metribuzin 0.10 mg∙kg-1; with the exception of ethyl parathion and metribuzin, all were above the maximum residue limit (MRL) established by the Alimentary Code and European Union. Methamidophos, chlorpyriphos and diazinon were found 270, 230 and 120 times above the MRL, with frequencies of detection of 70 %, 90 % and 70 %, respectively; mancozeb was 2.3 times above the MRL, but it was detected in all the samples. The results indicate that the consumption of strawberry, produced in the Municipality of Rivas Dávila, could constitute an important way of pesticide exposure through diet, because the fresh product consumer is exposed to a mixture of pesticides that are noxious to human health, can act additively or synergistically on the organism.
To reduce the excessive growth and malformations of roots during lifting of seedlings in trays, the vegetative growth of eggplants (Solanum melongena L.) ‘Long purple’ treated with chemical root pruning was characterized using 50 % copper hydroxide [Cu (OH)2] compared to untreated plants in Cabudare, Lara State, Venezuela. The seedlings were established in germination trays under greenhouse conditions and plants were evaluated the day of transplant and 23 and 49 days thereafter. The experiment was conducted as a split block design with two treatments (0 and 50% of [Cu (OH)2]), and 25 repetition using one plant per plot. The volume of roots, fresh biomass and number of leaves were significantly higher in plants treated with copper hydroxide 49 days after transplantation. Both 23 and 49 days after transplant, root length of treated plants was higher as compared to untreated ones. By applying copper hydroxide a decrease of radical malformations was observed, and a more fibrous, dense and compact root system was generated.
