Su toxicidad intravenosa y oral ha sido reportada, aunque su prescripción se fundamenta en aplicaciones tópicas. Esta investigación evaluó los efectos cronotrópicos y dromotrópicos de la aplicación tópica de amitraz (12% v/v; 4mL/L) en ratas anestesiadas con pentobarbital sódico y en perros conscientes. Se usaron 16 ratas y 12 perros, los cuales se sometieron a un electrocardiograma (ECG) control. Subsecuentemente, a 16 ratas se les aplicó amitraz en baño de inmersión por 2 min y a 12 perros se les aplicó baños de aspersión. Posteriormente, se registraron dos ECGs consecutivos cada 30 min, durante 1 h. Como indicadores de cronotropismo y dromotropismo, se usaron la frecuencia cardiaca (FC) y el intervalo P-R, respectivamente. La FC en ratas fue reducida (control: 265,7±12,1 lat/min vs amitraz: 234, 4±13,8 lat/min) a los 30 min del baño con amitraz, alcanzando diferencias significativas (P<0,05), a los 60 min (225,0±13,6 lat/min). Se prolongó el intervalo P-R (control: 54,3±2,0 mseg vs amitraz: 67,9±2,4 mseg) (P<0,05) a los 30 min y 60 min (72,1±2,8 mseg), respectivamente. En contraste, no se observaron cambios significativos en la FC e intervalo P-R en perros. Los efectos cronotrópicos y dromotrópicos negativos observados en ratas, podrían reflejar una acción agonista a2-adrenérgica presináptica en el sistema nervioso central, con la consecuente reducción del tono simpático, sin descartar una acción parasimpaticomimética. Posiblemente, la absorción cutánea del amitraz en caninos, sería incapaz de desarrollar estos efectos. En caninos sin premedicación, pudieran desarrollarse respuestas autonómicas reflejas capaces de regular un posible efecto del amitraz sobre las variables estudiadas. Las dosis terapéuticas de amitraz, no produjeron cambios en el ECG de caninos aparentemente sanos. Se requieren experimentos adicionales para esclarecer si dosis superiores en aplicación tópica actúan en forma similar en perros con lesiones de piel producidas por ácaros o garrapatas.<hr/>Amitraz is a diamidine molecule widely used as an ectoparasiticide in Veterinary Medicine. Although amitraz is prescribed as a topical treatment, its toxicity has been reported in oral and intravenous administration. This investigation assessed the potential chronotropic and dromotropic effects of topical amitraz (12% v/v; 4mL/L) in anesthetized rats and conscious dogs. Sixteen rats and twelve dogs were used. Before treatment, body surface was moistened with water and all animals were subjected to a baseline electrocardiogram (ECG). Subsequently, amitraz was applied to rats (n=16) as an immersion bath for 2 min, or as a spray application to dogs (n=12), respectively. Two consecutive ECGs were recorded at 30 min interval for 1 h. The heart rate (HR) and the P-R intervals were calculated to estimate chronotropism and dromotropism, respectively. The HR of rats was reduced (control: 265.7±12.1 beats/min vs amitraz: 234.4±13.8 beats/min) at 30 min after amitraz application, reaching a significant (P<0,05) value at 60 min (225.0±13.6 beats/min). The P-R interval was significantly (P<0.05) prolonged at 30 min (control: 54.3±2.0 msec vs amitraz: 67.9±2.4 msec) and 60 min (72.1±2.8 msec), respectively. In contrast, no significant changes in HR and P-R interval were found in dogs treated with amitraz. The negative chronotropic and dromotropic effects observed in rats may reflect the activation of pre-synaptic a2-adrenoceptors in the central nervous system and a reduction in the sympathetic outflow, without excluding a potential parasympatomimetic effect. The lack of these effects in dogs might be related either to reduced skin absorption or to a potential autonomic regulatory reflex in conscious dogs. The therapeutic dose of amitraz used herein did not modify the ECG in apparently healthy dogs. Further experiments are required to establish the effects of higher topical doses in dogs suffering from skin diseases related to mites or ticks infestations. <![CDATA[<b>Histology of the Spectacled Caiman Kidney <i>(Caiman crocodilus crocodilus)</i></b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S0258-65762009000200002&lng=pt&nrm=iso&tlng=pt Tras haberse reportado similitudes estructurales entre las nefronas de dos especies de crocodílidos poco relacionadas taxonómicamente, pero con hábitat similar, Caiman crocodilus crocodilus y Crocodilus acutus, y con el fin de determinar si estas estructuras ofrecen a C. c. crocodilus cierta ventaja en su capacidad de adaptación al hábitat, se llevó a cabo el estudio histológico del riñón de nueve babas macho adultas y de una hembra adulta, usando las coloraciones Hematoxilina-Eosina y tricrómica de Mallory. Entre otros hallazgos, se demostró la existencia de estructuras histológicas que permiten sugerir la presencia del aparato yuxtaglomerular en esta especie.<hr/>Structural similarities have been reported between the nephrons of two crocodrilian species (Caiman crocrodilus crocrodilus and Crocodilus acutus) not taxonomically related, but with a similar habitat. A histological study of the kidney was conducted to determine if these structures give the C. c crocodiles certain advantages in its capacity for adaptation to the habitat. Nine adult male and one adult female caimans were used in the study. Samples were stained with hematoxilin-eosin and Mallory’s tricromic stains. The results of the present study reveal the presence of microscopic structures that suggest the existence of a juxtaglomerular apparatus in this species. <![CDATA[<i>Characterization of Eastern Equine Encephalitis Virus by Reverse Transcriptase Polymerase Chain Reaction and Analysis of Single</i>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S0258-65762009000200003&lng=pt&nrm=iso&tlng=pt El análisis de polimorfismo en la conformación de cadenas sencillas (SSCP, por sus siglas en inglés) se utilizó para caracterizar regiones del genoma de tres cepas de virus de encefalitis equina del este (VEEE), aisladas de cerebros de equinos muertos. Los segmentos de ARN fueron amplificados mediante la transcriptasa reversa acoplada a la reacción en cadena de la polimerasa (RT-PCR). Se analizó un fragmento de 542 pb de la región 3’no traducible del ARNm 26S y un fragmento de 532 pb del gen de la proteína no estructural P4 (nsP4), encontrándose que la migración de las cadenas sencillas de estos fragmentos de ADNc fueron diferentes en estos aislados virales, lo que indica que presentan polimorfismo genético en estas regiones del genoma, el cual es característico de cepas de VEEE, variantes Sur Americanas.<hr/>The single-strand conformation polymorphism (SSCP) analysis was developed to characterize RNA segments of the genome in three strains of virus eastern equine encephalitis (EEEV), isolated from dead equine brains. The RNA segments were amplified by the reverse transcriptase polymerase chain reaction (RT-PCR). The analyses of the 542 pb fragment from the 3’ untranslated region of the 26S mRNA and the 532 pb fragment of the nonstructural P4 protein (nsP4) gene were performed. The results of the present investigation show that the migration of the single strands of the c-DNA fragments was different in these isolated viruses, which indicate genetic polymorphism in these genome regions, which is characteristic of EEEV strains from South American varieties. <![CDATA[<b>Patrón de citocinas en hígado y bazo de perros con Leishmaniasis visceral natural</b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S0258-65762009000200004&lng=pt&nrm=iso&tlng=pt En el presente estudio se caracterizó el patrón de citocinas del bazo e hígado de perros infectados en forma natural por Leishmania infantum (chagasi) provenientes del estado Nueva Esparta, Venezuela. Los perros fueron clasificados como sintomáticos y asintomáticos considerando tres signos principales de esta patología como hepatoesplenomegalia, caquexia y onicogrifosis. Muestras de hígado y bazo fueron tomadas para evaluar carga parasitaria y la expresión de citocinas y quimiocinas por inmunohistoquímica. En los perros sintomáticos se observó una mayor carga parasitaria tanto en el hígado como en bazo. En el bazo se observó una densidad de células positivas para interferón gamma (IFN-g), interlucinas (IL-12, IL-10, IL-4, IL-8) y Proteína 1 quimioatrayente de monocito (MCP-1), similar en perros asintomáticos y sintomáticos. Mientras que en el hígado la evaluación de las citocinas IFN-g, IL-12, IL-10 IL-8 y MCP-1, mostró una tendencia al aumento en el grupo de perros sintomáticos, con incremento significativo de las células IL-4+ en los perros sintomáticos, con respecto a los asintomáticos. La densidad de células IL-4+ en el hígado se correlacionó positivamente con la hepatomegalia. Al cuantificar las concentraciones séricas de la quimiocina Proteina 1 alfa inflamatoria de macrófago (MIP-1a) se observó una alta producción en los perros asintomáticos. Estos resultados confirman laexistencia de una respuesta órgano-especifica frente a L. infantum (chagasi), del tipo T cooperador 2 (Th2) en el hígado de los perros sintomáticos, caracterizada por un marcado predominio de IL-4 sobre IFN-g e IL-12. La elevada producción de IL-4 fue directamente relacionada con la hepatomegalia.<hr/>In the present study, we characterized the cytokine pattern in the spleen and liver of naturally infected dogs with Leishmania infantum (chagasi) from the Nueva Esparta state of Venezuela. The dogs were classified as symptomatic and asymptomatic considering three main signs of this disease: hepatosplenomegalia, cachexia and onichogriphosis. Tissue samples from liver and spleen were taken to evaluate parasite load, cytokines and chemokines expression by immunohistochemistry. In symptomatic dogs, a higher parasitic load was observed in the liver and spleen as compared to asymptomatic dogs. In the spleen, the density of gamma interferón (IFN-g), interleukins (IL-12, IL-10, IL-4, IL-8) and Monocyte Chemoattractant Protein-1 (MCP-1), it was similar between asymptomatic and symptomatic dogs. Whereas, in the liver IFN-g, IL-12, IL-10 IL-8 and MCP-1 showed a tendency to increase in symptomatic dogs, with a significant increment of IL-4 + cells. In the liver, the density of IL-4+ cells was positively correlated with hepatomegaly. In addition, higher concentrations of Macrophage inflammatory protein 1 alpha (MIP-1a) were observed in the serum of asymptomatic than in symptomatic dogs. These results corroborate the existence of an organ-specific immune respose against L. infantum (chagasi), with a T helper 2 (Th2) immune response in the liver of symptomatic dogs, with prevalence of IL-4 over IFN-g and IL-12. The high production of IL-4 is directly related with hepatomegalia. <![CDATA[Effect of Breed Group and Some Non-genetic Factors on Milk Production  and Calving Interval in Dual Purpose Cows]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S0258-65762009000200005&lng=pt&nrm=iso&tlng=pt Para determinar el efecto del grupo racial y algunos factores no genéticos sobre la producción de leche corregida a 244 días (P244) y el intervalo entre parto (IEP), se analizaron 9469 y 6288 observaciones, respectivamente. Los animales se encontraban a pastoreo y en dos ordeños diarios. El modelo estadístico incluyó los efectos: finca (F:1, 2), año de parto (AP: 1995-2005) para P244 y (AP: 1995-2004) para IEP; mes de parto (MP:1,2,3,……,12), número de lactancia, (NL:1,…, 6 ó más), grupo racial ≥ ¾ Bos indicus, ≥ ¾ Bos taurus, 50%Holstein-50% Cebú, 50% Pardo Suizo-50% Cebú y Mosaico lechero, (GR:>CEBU, >E, H50CB50, PS50CB50, ML) y las interacciones FxAP, FxGR, FxNL, APxMP, APxNL. Todos los efectos afectaron a P244d e IEP, a excepción del mes de parto que no fue significativo para P244. La P244 e IEP promedio fueron de 1744 kg y 467 d, respectivamente. Las diferencias entre el mejor y el peor año en P244 e IEP fueron de 275 kg y 141 d, respectivamente. El GR con mayor producción de leche y con el mayor IEP fue PS50CB50 con 1844 kg y 487 d, respectivamente y el de menor producción y menor IEP fue>CEBU con 1619 kg y 452 d. La diferencia en P244 y IEP entre la primera y la quinta lactancia fue de 181 kg y 20 d, respectivamente. Existieron diferencias entre las dos haciendas de 84 kg y 22 d para P244d e IEP, respectivamente. Es interesante resaltar que la interacción FxGR fue significativa (P<0.01), indicando que los GR se comportan de forma diferente en cada finca. Se puede concluir que los efectos no genéticos y el GR tienen una alta influencia sobre la P244 y el IEP.<hr/>In order to establish the effect of breed group and some non-genetic factors on milk production corrected at 244 days (P244) and also on calving interval (IEP), 9469 and 6288, observations were analyzed, respectively. The animals were at grazing and milked twice daily. The statistical model included effects of: farm (F: 1,2); year of birth (AP:1995-2005) for P244, and (AP: 1995-2004) for IEP; month of birth (MP: 1, … 12), lactation number, (NL: 1...6 or more), breed group: mostly cebu (≥ ¾ Bos indicus); mostly European (≥ ¾ Bos taurus); 50% Holstein-50% Zebu; 50% Brown Swiss-50% Cebu and Dairy Crossbred Mosaic (GR:>CEBU, >E, H50CB50, PS50CB50, ML) and the interactions FxAP, FxGR, FxNL, APxMP, APxNL. All studied factor affected P244d and IEP, except the month of birth which, was not significant for P244. The average P244 and IEP were 1744 kg and 466 d, respectively. The differences between the best and worst year for P244 and IEP were 275 kg and 141 d, respectively. The GR with the higher milk production and highest IEP was PS50CB50 with 1844 kg and 487 d, respectively. As well, GR with the lower production and lowest IEP was > CEBUE with 1619 kg and 452 d, respectively. The difference for P244 and IEP between the first and fifth sixth lactation was 181 kg and 20 d, respectively. There was difference between the two farms of 84 kg and 22 d for P244d and IEP, respectively. It is interesting to note that FxGR interaction was significant (P<0.01), indicating that GR performed differently on each farm. It can be concluded that the non-genetic effects and GR have a high influence on the P244 and IEP. <![CDATA[<b><i>Overfeeding Effects on Beef Carcass Traits and  Postmortem Degradation of Vinculin and Troponin-T in Dual Purpose Steers</i></b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S0258-65762009000200006&lng=pt&nrm=iso&tlng=pt El efecto de sobrealimentación sobre las características de la canal y la degradación de la vinculina y la troponina-T a tres tiempos postmortem (0, 24 y 48 h), fue evaluado en 10 novillos doble propósito de 21 m de edad. Los animales fueron segregados al azar en dos tratamientos durante 12 m, como sigue: Tratamiento 1 (T1): cinco (5) novillos (grupo testigo) alimentados con heno (Cynodon nlemfuensis), más 20% de concentrado (relación heno:concentrado = 80:20, para una ganancia diaria de peso (GDP) de 100 g/d). Tratamiento 2 (T2): cinco (5) novillos alimentados con heno (Cynodon nlemfuensis), más 40% de concentrado (relación heno: concentrado = 60:40, para una GDP de 500 g/d). Las canales fueron evaluadas, para las características cualitativas y cuantitativas a las 24 h postmortem. Para evaluar la degradación de vinculina y troponina-T mediante la técnica de Western Blot, se colectaron 0,5 g del músculo longissimus dorsi de cada media canal al beneficio (0, 24 y 48 h postmortem). El análisis de varianza(ANAVAR) incluyó el peso de los animales al inicio del ensayo como covariable. Los resultados indicaron que las canales de los novillos sobrealimentados mejoraron sus características en canal, presentando mayor área del ojo costal, mayor espesor de grasa y mayor madurez ósea (P<0,05), sin alcanzar niveles adecuados de acabado de grasa. Se observó que la sobrealimentación no varió el patrón de degradación de la vinculina y la troponina-T a ningún tiempo postmortem. La vinculina se degradó a partir de las 24 h postmortem; sin embargo, no se detectó degradación de la troponina-T. Estos resultados deben ser confirmados con un número mayor de animales junto con la evaluación cuantitativa de la expresión de las proteínas.<hr/>The effect of overfeeding on carcass characteristics and on degradation of vinculin and troponin-T at three different postmortem times (0, 24, and 48 h) were evaluated in dual purpose steers. A total of 10 steers, 21 months of age was used. The animals were randomly selected and allocated to two different treatment protocols for 12 months, as follows: Treatment 1 (T1): Five steers (Control Group) fed with a diet consistent of hay (Cynodon nlemfuensis) plus concentrate, with a hay/concentrate ratio of 80:20, to reach a daily weight gain (DWG) of 100 g/d; Treatment 2 (T2); five steers fed with hay (Cynodon nlemfuensis) plus concentrate with a hay/concentrate ratio of 60:40, to obtain a DWG of 500 g/d. Carcasses were assessed for both qualitative and quantitative characteristics at 24 h postmortem. To evaluate the degradation of vinculin and troponin-T at three different times (0, 24, and 48 h postmortem), 0.5 g of the longissimus dorsi muscle was collected from each half carcass and subjected to a Western blot analysis. Samples were analyzed by ANOVA. The analysis included the weight of the animals as a covariance. The results of the present investigation indicate that the carcasses of overfed steers exhibited better carcass traits, showing larger loin eye areas, back fat thickness, and a skeletal maturity (P≤0.05), without reaching adequate levels of fat finish. It was also observed that overfeeding did not change the pattern of degradation of vinculin and troponin-T at any postmortem time. Vinculin showed degradation at 24 h postmortem while troponin-T was not degraded. These results must be confirmed using a greater number of animals and a quantitative evaluation of protein expression.
