A study was conducted to determine if the video-assisted lobectomy performed on a canine model, reduces surgical time, bleeding volume, nerve structures injuries; and improves the cosmetic results of the cervical scar, when compared to conventional lobectomy. Thirty-six adult crossbred dogs of either sex from the animal facility of the Institute of Experimental Surgery, Universidad Central de Venezuela were used. The animals were randomly allocated into two experimental groups: Group A: Eighteen dogs who underwent conventional thyroid lobectomy (CTL); Group B: Eighteen dogs who underwent video-assisted thyroid lobectomy (VATL) without gas insulation. Cosmetic results were assessed by surgeons according to a scale, as follows: excellent, good, average, and bad. The results of the experiment show that dogs operated with the VATL technique exhibited significantly (P<0.05) the highest improvements (9.2±0.5), when compared to the CTL technique (5.8±0.7). Surgical time lasted longer (P<0.001) in dogs under VATL (81±3 min) than in dogs subjected to CTL (62±4 min). Bleeding volume was significantly higher in CTL (34±5 mL) than in VATL (23±5 mL). No statistically significant differences were found regarding paralysis of vocal cords or surgical wound infections. The VATL is a valid alternative in conventional surgery, becoming a feasible and safe experimental model. Comparative and multicentric studies should be carried out in human subjects to determine both safety and efficacy of these techniques.
A study was conducted with the purpose of establishing the morphologic characteristics of the urinary pathway of the male spectacled caiman (Caiman crocodilus crocodilus). A total of nine adult caimans were used. Samples of the urinary tract were collected and stained with Hematoxilyn-Eosin and Mallory’s trichromic stains. The results of the present study reveal that the presence of a muscular layer, departing from the collecting tubules, could be associated to the need of mobilizing small amounts of urine loaded with precipitated urates, by means of peristaltic movements, which may be associated with the need of this reptile of producing reduced volumes of isosmotic urine. This would lead to a reduction of fluid loss at a renal level. This possibly implies the secretion of mucines along the urinary tract, as a facilitator mechanism for the transport of precipitated urates.
A histochemical study of mucins and lipids of the urinary tract of the spectacled caiman was conducted, using PAS stain, PAS-Alcian blue stain (at pH values of 2.5 and 1.0, respectively), “O” red oil stain, and Shultz method for steroids. Nine mature males and one mature female were used. The results of the present study show that the most relevant findings were the presence of neutral mucins in the brush border and of cytoplasmic droplets in all tubular nephrons and in the apical surface of the urothelium, as well as the presence of sialomucins from the distal tubule to the ureter. Lipid droplets composed of free fatty acids were observed in the cytoplasm of the cells of the intermediate segment of the nephron. The importance of mucins as mucosal protectors and urinary tract lubricants is discussed. The results of the study in the Caiman crocodilus crocodilus, appear to demonstrate the absence of portions of the nephron that either participate in the secretion of seminal fluid or are similar to the sexual segment of lizards and snakes.
A study was conducted to obtain and characterize the nutritional, enzymatic, and ocratoxigenic properties of enzyme additives obtained from fermentation of moist residues of commercial production of pasta, of both a monoculture of Aspergillus niger (A1) and a mixed culture (coculture) of Aspergillus niger-Saccharomyces cerevisiae (A2). The effects on productive performance parameters were assessed, during 21 d. A total of 150 broilers (Cobb-500) were used and fed with diets which included the additives A1 and A2 in the amount of 30 and 3000 ppm, respectively, using a completely randomized design. The results show that both α-amylase and glucoamylase activities in A2 were 42 and 69% higher than those of A1, respectively. The physicochemical composition of additives revealed that they are mainly composed of starch and protein. Both A1 and A2 showed a similar amylase activity (15540 and 16020 FAU/g, respectively). However, the A2 exhibited higher glucoamylase and phytase activities (U/g) 58% than the A1. The ocratoxin A levels for A1 (2.10-4 ppb) and A2 (2.10-2 ppb), respectively, were below the median lethal dose (LD50) for chickens and for the limits established by FAO/WHO. Additionally, the viability of Aspergillus niger in A1 and A2 was 3.10³ UFC/mL and of 2.10² CFU/mL in yeast cells, respectively. The inclusion of 30 ppm of A1 and 3000 ppm of A2, respectively significantly (P<0.05) improved feed conversion (11-13%) and feed intake (6-8%), when compared to control animals. The treatments did not significantly affect weight gain.
An experimental research was conducted to describe both macroscopically and microscopically skin pythiosis in the Venezuelan Creole horse, which dwells in the Venezuelan plains of the States of Apure, Barinas, and Portuguesa. Visits were made to different farms between January 2008 and December 2009. Samples of eight horses with a skin disease consistent with skin pythiosis were taken and macroscopically analyzed. For the histopathological description, several tissue fragments were obtained and fixed in 10% buffer formalin. Subsequently, samples were stained with Hematoxylin-Eosin and Grocott’s silver methenamine, and observed under the light microscope. All samples showed histopathological findings consistent with skin pythiosis, which was confirmed through microbiological cultures. The results show that skin pythiosis is a dermatosis commonly seen in the Creole horse that populates the Venezuelan plains. Macroscopically, the lesions were characterized as irregular plaque-shaped, ulcerated, increased in volume, hemorrhagic, with a white-yellowish color, and a granulation tissue appearance, when examined both superficially and profoundly. Eventually, necrotic areas that contained irregular, yellowish, hard, sandy surfaces, which corresponded to kunker structures, were found. Conversely, the microscopic study revealed a diffuse to multifocal eosinophilic, granulomatous dermatitis.
Different experimental murine models have shown that protective immunity against Lesihmania depends upon T cells, cytokines, and antigen presenting cells. However, the role of cytokines in naturally-infected hosts like domestic dogs is controversial. Few studies have evaluated cytokines in dogs naturally-infected with Leishmania infantum/chagasi. Since the domestic dog is the main reservoir of the parasite, a study was conducted to determine cytokines in serum of 33 dogs with Canine Visceral Lesihmaniasis from endemic areas of the State of Nueva Esparta, Venezuela. Dogs were classified as symptomatic (SD) and asymptomatic (AD), according to the expression of three or more clinical signs and levels of antibodies for rK39 and rK26. Ten non-infected, rK39 negative controls were included from an endemic area (EA) and ten dogs from a non-endemic area were used as healthy controls (HC). The following cytokines (pg/mL) were measured in serum by flow cytometry (CBA Hu Th1/Th 2, BD TM kit): IFN-γ, TNF-α, IL-10, IL-6, IL-4 and IL-2. Results show a higher concentration (P<0.05) of IFN-γ (69.93±7.46), IL-4 (7.51±2.68), TNF-α (3.86±1.46), and IL-2 (39.85±3.84) in AD when compared with SD (60.8±10.6; 5.28±0.80; 2.76± 0.72; and 36.04±3.61, respectively); and HC (51±14; 4.65±0.2; 3.21±0.89, and 32.65±5.86, respectively). The AD also showed higher levels (P<0.01) of IL-6 (4.9±0.55) compared with HC (4.02±0.64). Results show that AD exhibit a higher proportion of cellular activation and proinflammatory cytokines. Results indicate that measuring of serum cytokines could reflect the immunological status in dogs in future clinical trials oriented to either vaccination or therapy.
To evaluate the hematology, blood metabolites, and organ weights in growing pigs fed peach-palm meal (PPM) and synthetic lysine (SL), 72 crossbred (Yorkshire x Landrace) barrows (30±0.5 kg) were used. Pigs were allotted into six treatments in a completely randomized design with a 2 x 3 factorial arrangement, with two levels of SL (0 and 0.27%) and three levels of PPM (0, 25 and 50%), with four replications per treatment and three pigs per experimental unit. The study lasted six weeks and blood samples were collected at the onsent of the assay and at third and sixth weeks. The hematological variables evaluated were hemoglobin, erythrocytes and leukocyte count, total leukocyte count, hematocrit, mean corpuscular volume (MCV), mean corpuscular hemoglobin concentration (MCHC). Blood metabolites evaluated were: total plasma proteins, urea, glucose, fructosamine, alanine aminotransferase (ALT), aspartate aminotransferase (AST). At the end of the trial, 24 pigs were slaughtered (four/treatment) and the lungs, heart, stomach, pancreas, small intestine, liver, spleen, and kidneys were weighed. The PPM x SL interaction did not significantly influence the variables studied. The SL did not affect hematology, while PPM (25 and 50%) significantly decreased erythrocyte count (P<0,05), hematocrit (P<0,05) and hemoglobin (P=0.09), without significant variations in MCV, MCHC, and leukocyte count, when compared to pigs that were fed no PPM. Pigs fed PPM had lower total proteins(P<0.01) and the SL reduced urea (P<0.05). The AST was lower (P=0.01) in pigs fed SL than in pigs without SL. Pigs that consumed 25% PPM tended to have (P=0.10) heavier stomachs than those fed basal diet or 50% PPM (P=0.06). Pigs fed 25 and 50% PPM exhibited the lightest spleens (P<0.01), compared to those without PPM. The PPM together with SL in the diet of growing pigs had no detrimental effects on hematological variables, blood metabolites, and organ weights.
