Sus efectos cardiovasculares han sido atribuidos a acciones directas y/o indirectas, actuando sobre sus órganos blancos y/o modulando el tono autonómico, respectivamente. El objetivo de esta investigación fue determinar los efectos de la xilazina in vivo sobre la presión arterial media (PAM) y la frecuencia cardiaca (FC) en conejos albinos Nueva Zelanda, y evaluar sus efectos in vitro en aurícula derecha y en vena safena aisladas. Se anestesiaron e instrumentaron cuatro conejos, para medir la PAM en forma directa y la FC mediante electrocardiografía. Una vez estabilizadas las variables hemodinámicas, se evaluaron dosis crecientes-acumulativas de xilazina intravenosa. Se determinó el efecto de concentraciones crecientes-acumulativas de xilazina sobre la aurícula derecha y vena safena en órganos aislados convencionales. Se evaluaron  los efectos de xilazina sobre la frecuencia cardiaca (lat/min) y la fuerza de contracción (g) auricular, y sobre la vena safena, en ausencia y presencia de prazosín (30nM) y yohimbina (0,1µM). La xilazina in vivo, causó una respuesta bifásica en la PAM, caracterizada por un aumento ligero, seguido de una profunda hipotensión dosis-dependiente, acompañada de bradicardia. En la aurícula derecha, la xilazina no causó modificaciones en su actividad espontánea, aunque en la vena safena produjo contracciones concentración-dependiente sensibles a yohimbina, pero resistentes a prazosín. Los resultados sugieren que la xilazina endovenosa causa reducción en la PAM y bradicardia en el conejo anestesiado, sin modificar las propiedades cronotrópicas e inotrópicas de la aurícula derecha aislada. Sin embargo, en la vena safena aislada, se demuestra que la xilazina causa vasocontracción, primordialmente a través de la acción sobre receptores a2-adrenérgicos. La xilazina ejerce efectos cardiovasculares en el conejo, probablemente a través de la modulación del sistema nervioso autónomo y no por acción directa sobre el corazón<hr/>Xylazine is an a2-adrenoceptor agonist widely employed in Veterinary Medicine as a sedative and analgesic compound. Xylazine cardiovascular effects have been attributed to direct and/or indirect actions, acting on target organs and/or modulating the autonomic outflow, respectively. The aim of this investigation was to determine the in vivo effect of xylazine on mean blood pressure (MBP) and heart rate (HR) in albino New Zealand rabbits and to assess the in vitro effects of xylazine on the isolated  right atrium and saphenous vein. Four rabbits were anesthetized and instrumentalized for directly measuring  MBP; and HR by electrocardiography respectively. After the stabilization of haemodynamic variables, intravenous cumulative increasing doses of xylazine were administered and evaluated.The effects of xylazine cumulative concentration-response curves on  the isolated right atrium and on the saphenous vein in conventional in vitro organ bath studies were assessed. Contraction rate (beats/min) and contraction force (g) were measured in the right atrium. Xylazine-induced contraction (g) in the absence and presence of prazosin (30nM) and yohimbine (0.1µM) was measured in the isolated saphenous vein. The intravenous administration of xylazine caused a biphasic response on the MBP, characterized by slight increase followed by a profound dose-dependent hypotension along with bradycardia. Xylazine did not modify the spontaneous activity of the isolated right atrium, although, yohimbine-sensitive and prazosin-resistant concentration-dependent contractions by xylazine were observed in the isolated saphenous vein. The results suggest that intravenous administration of xylazine causes a reduction in the MBP and bradycardia in the anesthetized rabbit, without a clear modification on the chronotropic and inotropic properties of the isolated right atrium. Nevertheless, xylazine caused vasocontraction of the isolated saphenous vein, primarily by activating a2-adrenoceptors. Xylazine appears to exert its cardiovascular effects in the rabbit by modulating the autonomic nervous system outflow and not by a direct action on the heart <![CDATA[<b>Morphological and Histochemical Characterization of the Liver of the Spectacled Cayman (<i>Caiman crocodilus crocodilus</i>)</b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S0258-65762012000100002&lng=pt&nrm=iso&tlng=pt La morfología, organización y los componentes intracitoplasmáticos del hepatocito de la baba (Caiman crocodilus crocodilus) son aspectos que se han estudiado parcialmente hasta el momento. Con el fin de generar información sobre la morfología microscópica del hígado de esta subespecie, se realizó un estudio descriptivo de las características morfológicas e histoquímicas del hígado de la baba, lo que permitirá la comprensión de algunas funciones hepáticas en estos ejemplares. De esta manera, se podrán realizar en el futuro, investigaciones utilizando a dicho órgano como modelo experimental para el estudio de las rutas metabólicas de los metabolitos procesados a este nivel. Se tomaron ocho muestras de hígado, se fijaron en formol al 10%  y se procesaron por los métodos histológicos de coloraciones de rutina (Hematoxilina y Eosina). Asimismo, se utilizaron las coloraciones especiales de Tricrómico de Gallegos, el método de Wilder y el aceite rojo “O”. Los hallazgos del presente trabajo revelaron la presencia de un hígado bilobulado, ubicado en la cavidad celómica caudal, con los  lóbulos derecho e izquierdo unidos por un istmo. El órgano se encuentra rodeado por una delgada cápsula de tejido conectivo, su parénquima se dispone en cordones tortuosos con un patrón acinar, con presencia de células pigmentadas compatibles con melanóforos, dispersas en el tejido hepático, y con gotas lipídicas intracitoplasmáticas de diversos tamaños, que en algunos casos ocupaban casi la totalidad del citoplasma de los hepatocitos<hr/>The morphology, organization and intracytoplasmic components of the liver of the spectacled cayman (Caiman crocodilus crocodilus) are aspects that have been partially studied so far. A descriptive study of the morphological and histochemical characteristics of the liver of this subspecies was performed. This investigation would enable researchers to understand the liver functions in these animals, thus allowing future research using this organ as an experimental model. Eight liver samples were fixed in 10% v/v formalin and processed by conventional histology using routine hematoxylin and eosin staining. Additionaly, special stains, such as the Trichrome of Gallegos method and the red oil “O” and Wilder, were also used. The findings revealed the presence of a bilobed liver located in the midline of the caudal coelomic cavity, surrounded by a thin capsule of connective tissue; the parenchyma showed the layout of the classic hepatic lobule, the hepatocytes were arranged in tortuous cords and an acinar pattern. The presence of pigment-laden cells compatible with melanophores scattered in liver tissue was evidenced. Intracytoplasmic lipid droplets of various sizes occupying almost the entire cytoplasm of the hepatocytes were observed <![CDATA[<b>Assessment of the Efficacy of the Agar Gel Immunodiffusion Test for the Detection of Antibodies Against the Enzootic Bovine Leukosis Virus</b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S0258-65762012000100003&lng=pt&nrm=iso&tlng=pt contra el virus de la leucosis enzoótica bovina (VLEB), comparándola con el ensayo inmunoenzimático (ELISA). Se procesaron 360 muestras de suero bovino para ambas pruebas, siguiendo las indicaciones del fabricante. El grado de concordancia (GC), valores predictivos (VP) y razones de verosimilitud (RV) fueron determinados y analizados. El GC entre ambas pruebas resultó ser del 86,4%, con un coeficiente Kappa (K) = 0,7. La IDGA detectó una significativa (P<0,001) menor proporción (47,2%) de sueros positivos que el ELISA (60,8%). La sensibilidad de la IDGA resultó ser del 77,6% por debajo de lo deseado, mientras que tuvo un 100% de especificidad. El VP positivo de100% y la razón de verosimilitud positiva RV con valor indeterminado (∞), mostraron que ésta es una prueba segura para la identificación de bovinos infectados con el VLEB. Contrariamente, el VP negativo de 74,2% y la RV negativa de 0,2, indican que existe una baja probabilidad, de que los bovinos con resultados seronegativos alVLEB, estén realmente libres de la infección por el VLEB. Los resultados sugieren que la IDGA es una prueba adecuada para estudios epidemiológicos en poblaciones con alta prevalencia del VLEB, por ser baja la posibilidad de generar resultados falsos positivos. Sin embargo, en poblaciones de baja prevalencia, su uso estaría restringido, debido a la considerable probabilidad de emitir resultados falsos negativos. En consecuencia, el ELISA debería ser la prueba de elección en programas de erradicación. Este trabajo es una contribución para el control y erradicación de la leucosis enzoótica bovina, al aportar información que permite seleccionar la técnica más adecuada, según la situación epidemiológica de esa enfermedad en la zona y el objetivo sanitario que se persigan<hr/>The efficacy of the agar gel immunodifusion test (AGID) for the detection of antibodies against the enzootic bovine leucosis virus (EBLV) was evaluated and compared with the ELISA test. A total of 360 bovine serum samples were tested for both tests, following the manufacturer instructions. The agreement degree (AD), predictive values (PV), and coefficient of probability (CP) were determined and analyzed. There was a good AD between both tests (86.4%; Kappa coefficient =0.7). The AGID test detected a significant (P<0.001) lesser proportion (47.2%) of positive sera than the ELISA test (60.8%). The AGID sensitivity was 77.6% lower than the expected, while there was 100% specificity. The positive PV of a 100% and the positive CP with an undetermined value (∞) showed that AGID is a safe test for the detection of cattle infected with EBLV. In contrast, the negative PV of 74.2% and the negative CP of 0.2 indicate that there is a slight probability that cattle, whose sera were negative to EBLV using the AGID, are really free of EBLV infection. These results suggest that AGID is a suitable test for epidemiological studies, inpopulations with high EBLV prevalence, due to the minor risk of producing false positive results. However in populations with low prevalence of the disease, the use of the AGID test would be restricted due to the high possibility of giving false negative results. Therefore, the ELISA should be the test selected for eradication programs. This work is a contribution to the control and eradication programs of the enzootic bovine leucosis, since it provides information which allows the selection of the most adequate technique, according to the epidemiological status of this disease in the zone and the sanitary target pursued <![CDATA[<b>Use of Oral Disinfectants in Broilers</b>: <b>Effects on Health Status </b>and Production Performance]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S0258-65762012000100004&lng=pt&nrm=iso&tlng=pt Se evaluó el efecto de cinco desinfectantes comerciales suministrados en el agua de bebida, sobre el estado de salud y desempeño productivo de pollos de engorde. Se utilizaron 216 pollitos machos Arbor Acres de un día de nacidos, durante 28 d y distribuidos en seis tratamientos: A: agua sin desinfectante; B:250 ppm de un fenol; C:250 ppm de un aldehído; D:250 ppm de un halógeno; E:250 ppm de ácidos orgánicos y F:250 ppm de un cresol. Cada tratamiento contó con seis réplicas de seis aves cada una. Se evaluaron variables productivas: consumo de alimento y agua, mortalidad y peso. Al final del ensayo, se tomaron muestras de órganos para histopatología y de sangre periférica para hematología y determinación de SGPT y TPP. No hubo diferencias estadísticas para el incremento de peso, el consumo de alimento y la conversión de alimento entre los tratamientos. El consumo de agua fue mayor en el tratamiento C (3.114,57 mL) (p<0,05). No se observó mortalidad en ninguno de los tratamientos. Histopatológicamente, no hubo alteración evidente en duodeno. Se observó leve congestión de los vasos portales y sinusoides hepáticos en todos los tratamientos, excepto en el tratamiento A. También se observó congestión moderada de vasos intersticiales e infiltrado mononuclear en el riñón, para los tratamientos B, E y F; observándose riñones normales en los pollos de los los tratamientos A, C y D. Los niveles séricos de glutamato-piruvato-transaminasa (GPT) y las proteínas plasmáticas fueron similares entre los tratamientos. Se observó leucopenia (p<0,05) en los tratamientos C (4.933 cel/mL), D (2.466 cel/mL), E (3.466 cel/ mL) y F (3.400 cel/ mL), con respecto a los tratamientos A (9.600 cel/ mL) y B (7.266 cel/ mL). El hematocrito fue menor al control (tratamiento A) en todos los tratamientos (p<0,05). A pesar de no observarse lesiones evidentes en duodeno, el consumo de desinfectantes produjo anemia y leucopenia, así como también lesiones moderadas en hígado y riñón; por lo que su uso prolongado, podría ocasionar problemas de salud, sin efectos sobre el desempeño productivo<hr/>The effect of the addition of five commercial disinfectants to the drinking water on health status and on productive performance of broilers was evaluated. Two-hundred and sixteen one-day old male Arbor Acres broilers were allocated into six treatments for 28 days, as follows: A: water without disinfectant; B:250 ppm of a phenol; C:250 ppm of an aldehyde; D:250 ppm of an halogene; E:250 ppm of organic acids; and F: 250 ppm of a cresol. Each treatment included six replicates of six broilers. Productive variables such as feed and water intake, body weight, feed conversion, and mortality, were assessed. At the end of the experiment, tissue samples were taken for histopathology studies; peripheral blood was withdrawn for hematocrit (Ht) and white cells count; and for measuring serum glutamic-pyruvic transaminase (SGPT) and total plasma proteins (TPP). The results showed no statistical differences (p>0.05) among treatments for body weight gain, feed intake, and feed conversion. Water intake was highest (p<0.05) in treatment C (3,114.57 mL). No mortality was seen in all treatments. No evident histopathology changes were observed in duodenum samples. A minor congestion of portal blood vessels, and sinusoids was evidenced in all treatments, except for treatment A. Additionally, a moderate congestion along with a mononuclear infiltrate of interstitial blood vessels was evident in kidneys of treatments B, E, and F, whereas treatments A, C, and D had a normal appearance. The SGPT and TPP levels were similar (p>0,05) among treatments. There was leukopenia (p<0.05) in treatments C (4,933 cells/mL), D (2,466 cells/mL), E (3,466 cells/mL), and F (3,400 cells/ mL), compared to treatments A (9,600 cells/ mL) and B (7,266 cells/ mL). The Ht was lower (p<0.05) than the control in all treatments. Despite the absence of overt duodenal lesions, consumption of disinfectants in the drinking water caused anemia and leucopenia in all treatments, as well as moderate lesions in kidneys and liver. It is concluded that their prolonged use could cause serious health problems, once the disinfectants or their metabolites distribute through the circulatory system. However, the productive variables were not affected <![CDATA[<b>Ovarian Follicular Dynamics During the Estrous Cycle in Brahman Cows</b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S0258-65762012000100005&lng=pt&nrm=iso&tlng=pt Con el objetivo de caracterizar la dinámica folicular durante el ciclo estral (CE) en vacas Brahman, fueron evaluados 12 CE’s con duración promedio de 21,3 ± 0,7 d (rango=19-24 d). Setenta y cinco porciento (9/12) de los CE’s presentaron dos ondas de crecimiento folicular (CF), emergiendo los días 1,6 ± 0,4 y 8,4 ± 0,4 del ciclo, respectivamente. En CE’s de una sola onda (2/12), la emergencia ocurrió el día 14,5 ± 0,7; mientras en el CE de tres ondas (1/12), las mismas emergieron los días 2, 10 y 17. Durante el CE ocurrieron cambios en las clases foliculares (Clase 1 [C1]: ≤5 mm; Clase 2 [C2]: 6-9 mm; Clase 3 [C3]: ≥10 mm). Disminuyó el número de folículos C1 mientras aumentó el número de folículos C2 durante el reclutamiento, y descendió el número de folículos C2 mientras aumentó el número de folículos C3 durante la selección. La tasa de crecimiento (TC) fue mayor (P<0,05) para el folículo dominante (FD1= 0,78 ± 0,1 mm/d; FD2=0,40 ± 0,1 mm/d), que en el folículo subordinado (FS1= -0,01 ± 0,1 mm/d; FS2= 0,22 ± 0,1 mm/d). No hubo diferencia en el máximo diámetro alcanzado por el FD (FD1= 12,2 ± 0,4 mm; FD2= 12,7 ± 0,4 mm). El diámetro del cuerpo lúteo (CL) aumentó (fase de crecimiento) hasta alcanzar su máximo tamaño (fase de meseta), disminuyendo hasta sudesaparición (fase de regresión). Los niveles de P4 mostraron correlación positiva con el diámetro del CL (r=0,54; P<0,01), aumentando los niveles de P4 durante la fase de crecimiento del CL y disminuyendo durante su regresión, con poca variación durante la fase de meseta. Estos resultados permiten señalar que la dinámica folicular en vacas Brahman presenta los mismos patrones observados en ganado Bos indicus, previamente reportados<hr/>To characterize the follicular dynamics during the estrous cycle (EC) in Brahman cows, 12 ECs with a mean duration of 21.3 ± 0.7 d (range= 19-24 d), were evaluated. Seventy five percent (9/12) of the EC’s had two waves of follicular growth (FG; 20.6+0.5 d), emerging on days 1.6+0.4 and 8.4+0.4, respectively. In EC’s of one wave of FG (2/12), emergence occurred on day 14.5+0.7; whereas in EC’s with three waves (1/12), waves emerged on days 2, 10 and 17, respectively. During the EC, the number of follicles (Class 1 [C1]= ≤ 5 mm, Class 2 [C2]= 6-9 mm; Class 3 [C3]= ≥ 10 mm) changed, showing a decrease in C1 follicles and an increase in C2 follicles, during the recruitment phase, and a decrease in C2 follicles during the selection phase, with an increase in C3 follicles. The growth rate of the dominant follicle (DF; DF1=0.78 ± 0.1 mm/d; FD2= 0.40 ± 0.1 mm/d) was greater (P<0.05) than the growth rate for subordinate follicles (SF1=-0.01 ± 0.1 mm/d; SF2=0.22 ± 0.1 mm/d). There was no difference in size between DF1 and DF2 (12.2±0.4 vs. 12.7 ± 0.4 mm). From the beginning the diameter of the corpus luteum (CL) increased (growth phase) until it reached the maximum size (plateau phase), to decrease due to its regression. Plasma progesterone (P4) levels showed a positive correlation (r=0.54; P< 0.01) with the diameter of the CL. These results allowed to conclude that the follicular dynamics during the EC in Brahman cows had the same patterns observed in Bos indicus and Bos taurus cattle, previously reported <![CDATA[<b>Phylogenetic Analysis of Venezuelan Strains of the Eastern </b>Equine Encephalitis Virus]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S0258-65762012000100006&lng=pt&nrm=iso&tlng=pt Se realizó un estudio molecular de cinco cepas de virus de encefalitis equina del este (VEEE) mediante secuenciación de productos obtenidos en el ensayo de transcriptasa reversa acoplada a la reacción en cadena de la polimerasa (RT-PCR, por sus siglas en inglés). Secuencias de ~ 500 pb de la región 3’ no traducible del genoma se compararon con secuencias homólogas variantes Sur Americana (SA) y Norte Americana (NA) del complejo VEEE. La cepa EE VE02 EL PAO, con la cual se desarrolló una vacuna inactivada oleosa tuvo más similitud con la cepa EE VE76 EL DELIRIO, seguida por EE VE75 CATATUMBO,  EE VE96 MLLANO y EE VE84 TUCACAS respectivamente. La alineación de secuencias entre las cepas típicas reveló diferencias de 2,82 a 8,48 %. En tanto que, la cepa EE VE02 EL PAO difirió en un 8,86 % de la de Perú (EE PE-0,0155 del linaje III) y en un 38,08 % de la de EEUU (EE-PE-6 del linaje I, usada en vacunas comerciales). Los resultados indican que las cepas autóctonas tuvieron más similitud con la cepa SA, que con la NA. Se han identificado cuatro principales linajes en el complejo de VEEE (I-IV). El árbol filogenético evidenció dos clado: uno que agrupa los linajes I-III y el otro, el linaje IV. La mayoría de las secuencias están contenidas en el primer clado que, a su vez, se separa en dos grupos que contienen secuencias del linaje I y secuencias de los linajes  II-III y, debido al valor bootstrap alto (99%), los grupos se consideran completamente distintos. Sin embargo, éste no es el caso del grupo que contiene a los linajes II-III porque éstos se separan sólo el 53% de las veces, pero se asume que las cepas venezolanas pertenecen al linaje III dividido en dos subclados: IIIA y IIIB<hr/>A molecular study of five strains of the eastern equine encephalitis virus (EEEV) was conducted by sequencing of products generated using the reverse transcriptase assay, coupled to the polymerase chain reaction (RT-PCR). Sequences of approximately 500 pair of bases from the non-translated 3’ region of the genome were compared with homologous sequences of the South American (SA) and North American (NA) variants of the EEEV complex. The EE VE02 EL PAO strain, used to develop an inactivated oil vaccine had most resemblance with the EE VE76 EL DELIRIO strain, followed by the EE VE75 CATATUMBO, the EE VE96 MLLANO and the EE VE84 TUCACAS strains, respectively. The alignment of sequences among typical strains revealed differences of 2.82 and 8.48%, respectively, while the EE VE02 EL PAO strain differed in 8.86% from the Perú strain (EE PE-0.0155, from lineage III) and in 38.08% from the NA strain (EE-PE-6, from lineage I, used in commercial vaccines). The results of the present investigation indicate that the autochthonous strains have more similarly with the SA strain than with the NA strain. Four major lineages (I-IV) in the EEEV complex have been identified. The phylogenetic tree evidenced two clades, one which groups lineages I-III and another, lineage IV. Most of the sequences are contained in the first clade, which at the same time, separates into two groups containing sequences of lineage I and lineages II-III; and, due to the high bootstrap value (99%) the groups are considered completely different. However, this was not the case of the group which enclosed the lineages II and III; because these only separate 53% of the time, but it is assumed that the Venezuelan strains belong to linage III, which diverges into two subclades: IIIA and IIIB
