El presente estudio se efectuó con el propósito de evaluar la distribución de los mastocitos en las glándulas salivales mayores (parótida, submaxilar y sublingual) de rata. Las muestras de tejido glandular fueron incluidas en parafina y los cortes histológicos obtenidos se colorearon con Azul alcian-safranina y Azul de Toluidina. Posteriormente se efectuó la cuantificación de mastocitos/mm2 considerando dos áreas glandulares: el estroma intralobulillar y el interlobulillar. Los resultados no muestran variaciones significativas en la población de mastocitos al comparar las tres glándulas (p>0,05), pero si se encontró una mayor presencia de mastocitos en relación con la vía excretora principal. <abstract abstract-type="short" xml:lang="en"><p><![CDATA[Mast cells (MT) are connective tissue cells widely distributed throughout the body, especially in immune mucosal response sites like the digestive and air way tract. The aim of the present study was to find the number and the pattern of distribution and possible differences in mast cell population present in the mayor salivary glands (parotid, submandibular and sublingual glands) of rats. Fragments from salivary glands were collected, processed an included in paraffin wax, cut and stained with alcian blue-safranin and toluidine blue. The total number of MT was counted to estimate the mm² population from both intralobulillar and interlobulillar stroma tissues. Statistical analysis showed not significant differences (p> 0.05) between the three analysed glands. <body><![CDATA[<p style="text-autospace: none; word-spacing: 0; line-height: 100%" align="justify"> <font face="Verdana" size="2"><span lang="ES-VE"><b>Recibido  para arbitraje:</b> 16/04/2008 <b> Aceptado para publicación: </b> 06/11/2008</span></font></p>     <p style="text-align: justify; word-spacing: 0; line-height: 100%"><b> <font size="2" face="Verdana">INTRODUCCIÓN</font></b></p>     <p style="text-align: justify; word-spacing: 0; line-height: 100%"> <font size="2" face="Verdana">&nbsp;Los mastocitos &nbsp;son células del tejido conectivo derivadas de  una célula pluripotencial CD34+, localizada en &nbsp;la medula ósea <sup>(1).</sup>&nbsp;  Se distribuyen con preferencia en la dermis cutánea y en varias &nbsp;mucosas como la  digestiva, urinaria y respiratoria, aunque su cantidad y densidad son mayores en  la interfase entre los ambientes externos e internos donde pueden responder a  antígenos extraños <sup>(2).</sup> Presentan en su citoplasma gránulos  basófilos&nbsp; envueltos por membrana y que resultan metacromáticos cuando se tiñen  con anilinas básicas, lo que permite su identificación en cortes histológicos <sup>(3).</sup> </font></p>     <p style="text-align: justify; word-spacing: 0; line-height: 100%"> <font size="2" face="Verdana">Los mastocitos pueden ser activados por mecanismos inmunológicos  o no inmunológicos, liberando una gran cantidad de mediadores&nbsp; incluyendo  citoquinas, aminas vasoactivas y enzimas. La cantidad de mastocitos es mayor en&nbsp;  sitios de inflamación&nbsp; aguda y crónica como se describen en los casos de  psoriasis, enfermedad inflamatoria intestinal, parasitosis y reacciones de  hipersensibilidad alérgica como urticaria y asma <sup>(4,5,6,7).</sup> </font> </p>     <p style="text-align: justify; word-spacing: 0; line-height: 100%"><font face="Verdana" size="2">Se ha demostrado que algunos mediadores liberados por los  mastocitos, especialmente el factor de necrosis tumoral alfa, aumenta la  movilización de&nbsp; neutrófilos ante la aparición de determinadas bacterias y,  además, &nbsp;la migración de linfocitos intraepiteliales <sup> (8).</sup>  Por otro lado, Artuc et al., 1999; describe el papel de los mastocitos&nbsp;&nbsp; en la  reparación de heridas, ampliando el papel funcional de los mastocitos <sup>(9</sup>).</font></p>     <p style="text-align: justify; word-spacing: 0; line-height: 100%"> <font size="2" face="Verdana">En la cavidad bucal varias investigaciones mencionan un  incremento en el número de mastocitos en los casos de enfermedad periodontal,  así como también se destaca la participación de mastocitos en los casos de  liquen plano bucal <sup>(10,11).</sup> </font></p>     <p style="text-align: justify; word-spacing: 0; line-height: 100%"> <font size="2" face="Verdana">Existen trabajos previos que indican la presencia de mastocitos  en glándulas salivales en diferentes mamíferos <sup>(12,13, 14).</sup> La  mayoría de los mastocitos de la cavidad bucal expresan las proteasas, triptasa y  quimasa, mientras que una subpoblación menor son quimasa negativos(<sup>15</sup>).  &nbsp;Evidencias recientes sugieren&nbsp; la participación de la triptasa, contenida en  los gránulos mastocitarios, en la fibrosis que acontece en sialoadenitis crónica  de las glándulas submaxilares <sup>(16</sup>). Los mastocitos han sido  implicados también en varios desordenes inflamatorios inmunes que afectan las  glándulas salivales, como el síndrome de Sögren <sup>(17)</sup>.</font></p>     <p style="text-align: justify; word-spacing: 0; line-height: 100%"> <font size="2" face="Verdana">Aunque hay numerosos reportes sobre la participación de los  mastocitos en diferentes patologías de la cavidad bucal, la información sobre la  participación de los mastocitos glandulares en condiciones de normalidad, es  escasa. </font></p>     <p style="text-align: justify; word-spacing: 0; line-height: 100%"> <font size="2" face="Verdana">Hasta el momento no existen datos que permitan comparar la  distribución de estas células en una glándula de secreción puramente serosa,  como la parótida, en relación con otras glándulas de secreción mixta de  predominio seroso, como la submaxilar, o de predominio mucoso, como las  glándulas sublinguales. </font></p>     <p style="text-align: justify; word-spacing: 0; line-height: 100%"> <font size="2" face="Verdana">El objetivo principal de este trabajo es describir la  distribución de los mastocitos en el estroma de las glándula salivales parótida,  submaxilar y sublingual en forma comparativa. El disponer de esta información  contribuirá a comprender mejor el papel de estas células dentro del sistema  inmune de cavidad bucal.&nbsp;</font></p>     ]]></body>
<body><![CDATA[<p style="text-align: justify; word-spacing: 0; line-height: 100%"><b> <font size="2" face="Verdana">MATERIALES Y MÉTODO:</font></b></p>     <p align="justify" style="word-spacing: 0; line-height: 100%"><font size="2" face="Verdana">Se  utilizaron quince ratas&nbsp; Wistar de&nbsp; dos meses de edad pertenecientes a &nbsp;ambos  sexos. Los animales fueron sacrificados después de ser colocados&nbsp; bajo campana  con vapores de éter, <i>respetando protocolos de cuidado y uso de animales de  laboratorio </i><sup>(18</sup><i>). </i>Se disecaron y extrajeron&nbsp; treinta  glándulas parótidas y &nbsp;quince submaxilares y seis sublinguales. Las glándulas  extraídas fueron seccionadas en pequeños trozos y colocadas en fijador. Cada uno  de los trozos fue inmerso en solución de paraformaldehido al 4 %, preparado una  hora antes de su utilización. Los especimenes se mantuvieron en fijador durante  12 horas. Posteriormente las muestras&nbsp; fueron deshidratadas en concentraciones  crecientes de alcoholes e&nbsp; incluidos en parafina. &nbsp;El tejido incluido en bloque  de&nbsp; parafina se cortó con micrótomo es&nbsp; secciones de 5 micras de espesor</font></p>     <p align="justify" style="word-spacing: 0; line-height: 100%"><font size="2" face="Verdana">Para  observar la organización histológica general de las glándulas salivales se  tiñeron dos cortes por cada glándula con Hematoxilina-eosina. Esta es una  técnica de tinción habitual de muestras histológicas pero no diferencia los  mastocitos en forma específica, por lo cual se utilizó para identificar los  componentes glandulares y como control negativo</font></p>     <p align="justify" style="word-spacing: 0; line-height: 100%"><font size="2" face="Verdana">Otros seis  &nbsp;cortes &nbsp;semiseriados provenientes de cada una de las glándulas &nbsp;fueron  coloreados con &nbsp;las técnicas que se detallan a continuación:</font></p>     <p style="text-indent: 0cm; word-spacing: 0; line-height: 100%; margin-left: 0cm" align="justify"> <font face="Verdana" size="2">a)<span style="font-style: normal; font-variant: normal; font-weight: normal; font-family: Verdana">&nbsp;</span>Azul Alcian-safranina. Coloración utilizada según la técnica descripta por Bancroft y  Stevens (1982) <sup>(19).</sup></font></p>     <p style="text-indent: 0cm; word-spacing: 0; line-height: 100%; margin-left: 0cm" align="justify"> <font face="Verdana" size="2">b)<span style="font-style: normal; font-variant: normal; font-weight: normal; font-family: Verdana">&nbsp;</span>Azul  de toluidina) es una anilina básica ya utilizada para identificar y contabilizar  mastocitos en tejidos glandulares por lo que se utilizó como control positivo <sup>(3).</sup>&nbsp;</font></p>     <p align="justify" style="word-spacing: 0; line-height: 100%"><font size="2" face="Verdana">Las  muestras se observaron con un microscopio Zeizz Axiostar plus, se fotografiaron  con una cámara digital Canon. Para analizar la distribución celular, las  imágenes se digitalizaron utilizando el programa Image Pro plus. &nbsp;Para conocer  la población de mastocitos por mm<sup>2</sup> se tomaron ocho zonas al azar en  &nbsp;cada uno de&nbsp; los cortes.&nbsp; Se contaron y promediaron los mastocitos por mm<sup>2</sup>,&nbsp;  considerando&nbsp; dos sectores topográficos: a) estroma interlobulillar&nbsp;&nbsp; y,&nbsp; b)  &nbsp;estroma intralobulillar.</font></p>     <p align="justify" style="word-spacing: 0; line-height: 100%"> <font size="2" face="Verdana"><span style="font-weight: 700"> Análisis estadístico</span></font></p>     <p align="justify" style="word-spacing: 0; line-height: 100%"><font size="2" face="Verdana">Los  resultados se expresan como la media aritmética ± error estándar. Las  comparaciones entre las diferentes medias se llevaron a cabo mediante la prueba  de ANOVA y para el la comparación entre los dos sectores analizados se utilizó&nbsp;  el test de Student con un nivel de significación de 0,05%</font></p>     <p align="justify" style="word-spacing: 0; line-height: 100%"><b><font size="2" face="Verdana">&nbsp;  RESULTADOS</font></b></p>     ]]></body>
<body><![CDATA[<p align="justify" style="word-spacing: 0; line-height: 100%"><font size="2" face="Verdana">&nbsp;Las  técnicas histológicas empleadas permitieron identificar los mastocitos por la  presencia de sus gránulos metacromáticos característicos, los cuales  &nbsp;reaccionaron adecuadamente con los colorantes empleados (<a href="#fig1">figura 1</a>).</font></p>     <p align="center" style="word-spacing: 0; line-height: 100%"> <a name="fig1"> <font face="Verdana" size="2"> <img border="0" src="/img/fbpe/aov/v48n1/art02fig1.gif" width="555" height="464"></font></a></p>     
<p align="justify" style="word-spacing: 0; line-height: 100%"><font size="2" face="Verdana">Los  mastocitos aparecen localizados, con especial preferencia en el tejido conectivo  interlobulillar donde su numero fue significativamente alto en relación al  estroma intralobulillar (P&lt;=0.001), &nbsp;tanto en&nbsp; la glándula parótida como en la  submaxilar y sublingual (<a href="#tab1">tabla 1</a>). Solo un escaso porcentaje de mastocitos sufrió  perdida de los gránulos citoplasmáticos degranulación), con los métodos  empleados. Llamó la atención la adaptación de la forma celular&nbsp; que observamos&nbsp;  en algunos sectores glandulares; a pesar de que la mayoría de los mastocitos de&nbsp;  los espacios interlobulillares presentaron forma redondeada o ligeramente  ovalada (<a href="#fig2">figura 2b</a>), las células localizadas en &nbsp;periferia de cada lobulillo  adoptaron un aspecto mas alargado con su eje mayor paralelo al lobulillo y,  aunque las células están muy separados entre sí, asemejan una pseudo-empalizada  (<a href="#fig3">FIGURA 3a</a>&nbsp; y <a href="#fig3">3b</a>).</font></p>     <p align="center" style="word-spacing: 0; line-height: 100%"> <a name="fig2"> <font face="Verdana" size="2"> <img border="0" src="/img/fbpe/aov/v48n1/art02fig2.gif" width="555" height="461"></font></a></p>

<p style="text-align: justify; word-spacing: 0; line-height: 100%"><b> <font size="2" face="Verdana">DISCUSION</font></b></p>     <p align="justify" style="word-spacing: 0; line-height: 100%"><font size="2" face="Verdana">La  presencia de mastocitos en las glándulas salivales es conocida, pero su función  dentro de la Walsh, (2003) menciona que la distribución de  mastocitos en compartimientos titulares se relaciona con el potencial de los  mediadores mastocitarios en influenciar células próximas, resultado esto en  efectos estimulatorios, inhibitorios o tóxicos <sup>(22).</sup> </font> </p>     ]]></body>
<body><![CDATA[<p style="text-align: justify; word-spacing: 0; line-height: 100%"> <font size="2" face="Verdana">Nuestros hallazgos indican que los mastocitos además de reunirse  cerca de los vasos sanguíneos también lo hacen en las proximidades de la vía  excretora principal de las glándulas salivales. Estudios previos mencionan al  factor de crecimiento mastocitario (MGF, del inglés mast cell growth factor)  como un mediador clave que actúa influenciando la migración y asentamiento &nbsp;de  los mastocitos. Este mediador puede ser sintetizado tanto por células  endoteliales como por keratinocitos <sup>(23), </sup>lo que sustenta nuestras  observaciones.</font></p>     <p style="text-align: justify; word-spacing: 0; line-height: 100%"><font face="Verdana" size="2">Los mastocitos expresan las moléculas MHC I y II, por lo que  pueden interactuar con otras células del sistema inmune ya que poseen la  capacidad para procesar antígenos virales y bacterianos <sup>(24).</sup> Son  considerados células presentadoras de antígenos no profesionales, y por su  localización perivascular comparten una ubicación estratégica con células  presentadoras de antígenos profesionales como células dendríticas <sup>(25).</sup>  En concordancia con esto hemos descrito previamente la presencia de célula  dendríticas en relación de proximidad&nbsp; con las vías excretoras principales de  glándulas salivales mayores &nbsp;y, especialmente, la localización dentro del  epitelio ductal de la célula de &nbsp;Langerhans, este último dato también reportado  por Stasulis et al., (2003). <sup>(26, 27)</sup></font></p>     <p style="text-align: justify; word-spacing: 0; line-height: 100%"><font face="Verdana" size="2">Recientemente se describió la interacción&nbsp; célula&nbsp; Langerhans-mastocitos <sup>(28, 29</sup>) y al mismo tiempo Heib et al., (2007) ha demostrado el papel  crucial de los mastocitos activados promoviendo, vía factor de necrosis tumoral  alfa e interleuquina-1, &nbsp;la migración de las células de Langerhans  intraepiteliales <sup>(30).</sup></font></p>     <p style="text-align: justify; word-spacing: 0; line-height: 100%"> <font size="2" face="Verdana">En conjunto los reportes mencionados señalan&nbsp; la posibilidad,&nbsp;&nbsp;  por parte de los mastocitos, &nbsp;de iniciar una activación de la respuesta inmune  específica al poseer la capacidad de capturar y procesar antígenos o  indirectamente por su interacción con otras células del sistema inmune.</font></p>     <p style="text-align: justify; word-spacing: 0; line-height: 100%"> <font size="2" face="Verdana">Algunas patologías infecciosas e inflamatorias afectan con  preferencia las glándulas salivales pudiendo los microorganismos alcanzar la  glándula por vía sanguínea o en forma ascendente a través de la vía excretora <sup>(31</sup>). En otras patologías inflamatorias como la parotiditis  recurrente en niños y jóvenes la causa permanece aun incierta&nbsp; postulándose  entre su posibles causas los mecanismos inmunes <sup>(32</sup>)&nbsp; Es posible  suponer, en base a nuestros hallazgos, que la distribución de los mastocitos en  relación con la vía excretora principal&nbsp; les brinda mayor chance de interactuar  con los antígenos que ingresan a la cavidad bucal en forma directa o  indirectamente por su estrecha relación con las células dendríticas pudiendo  estar involucrados en la repetición de procesos inflamatorios crónicos como los  mencionados.</font></p>     <p style="text-align: justify; text-indent: 0cm; line-height: 100%; word-spacing: 0"> <font size="2" face="Verdana"> <span style="font-weight: 700"> AGRADECIMIENTOS</span></font></p>     <p align="justify" style="word-spacing: 0; line-height: 100%"> <font size="2" face="Verdana"><span lang="ES-AR"> Agradecemos especialmente la excelente asistencia técnica de&nbsp;  Juana Dora Dávila.
