No Brasil, a adição destes aditivos é permitida no limite máximo (LMP) de 50 mg/kg. O método analítico validado fundamenta-se na redução do nitrato a nitrito em coluna de cádmio e quantificação espectrofotométrica por reação de diazoconjugação com ácido sulfanílico/a-naftol. Das 43 amostras analisadas, 38 (88,37%) não apresentaram nitrato e nitrito em níveis detectáveis; em apenas 5 (11,63%) foi encontrado nitrato, sendo que em 4, acima do LMP. Do total, somente l (2,33%) apresentou nitrito, porém abaixo do LMP. Do ponto de vista toxicológico, a maioria das amostras podem ser consideradas seguras, apesar dos resultados também poderem signficar maior risco de contaminação bacteriana.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[Nitrate and nitrite in homemade and industrialized cheeses commercialized in the Southern region of Minas Gerais, Brazil. The evaluation of nitrate and nitrite in homemade and industrialized cheeses commercialized in the southern region of Minas Gerais State, was the aim of this work. In Brazil, the use of these additives is permitted at maximum levels of 50 mg/kg. The basis of the previously validated method is the quantitative reduction of nitrate to nitrite through cadmium column and spectrophotometric determination after nitrite diazotation with sulphanilic acid/a-naphtol reagent. From all samples analyzed. 38 (88.37%) showed neither a nitrate nor nitrite detectable content; 5 samples (11,63%) presented nitrate, 4 of them being above the tolerated level and only one (2,33%) showed detectable nitrite, however below the permissible limit. The majority of samples did not show nitrate or nitrite at detectable levels. However, though probably safe from a toxicological point of view, the results can implicate an incrcased risk of bacteriological contamination.]]></p></abstract>

<body><![CDATA[<P ALIGN="JUSTIFY">Recibido: 14-12-1998  Aceptado: 02-06-1999</P> <B> <FONT FACE="Arial">    <P>INTRODU&Ccedil;&Atilde;O</P> </FONT> </B>    <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; OS queijos podem ser afetados por diversos tipos de defeitos, o mais temido &eacute; o estufamento tardio, sendo na maioria das vezes causado pelo <I>Clostridium tyrobotyricum</I> <sup><b>(1,2)</b></sup> cuja presen&ccedil;a causa fermenta&ccedil;&atilde;o do lactato <b><sup> (1)</sup></b> produzindo na massa &aacute;cido but&iacute;rico, g&aacute;s carb&ocirc;nico e hidrog&ecirc;nio. <b><sup>(2</sup></b><sup><b>-4)</b></sup> O ac&uacute;mulo destes gases causa estufamento interno, com n&uacute;mero vari&aacute;vel de olhaduras grandes e irregulares. Pode apresentar trincas na casca, odor de ran&ccedil;o desagrad&aacute;vel e sabor estranho, devido ao &aacute;cido but&iacute;rico. Os queijos duros, com casca, ou os embalados em pel&iacute;cula pl&aacute;stica possuem facilidade a reten&ccedil;&atilde;o de gases e aos estufamentos. Portanto, quando o defeito ocorre, os queijos s&atilde;o profundamente alterados do ponto de vista est&eacute;tico, f&iacute;sico-qu&iacute;mico e organol&eacute;ptico, de maneira que dificilmente podem ser vendidos no mercado regular. <b><sup> (5)</sup></b> </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Corno conseq&uuml;&ecirc;ncia do estufamento tardio, muitas pesquisas t&ecirc;m sido realizadas para testar m&eacute;etodos que evitem o crescimento destes microrganismos. <b><sup> (6) </sup></b> O mais eficaz e conveniente para aplica&ccedil;&atilde;o em ind&uacute;strias <b><sup> (4-9)</sup></b> &eacute; adi&ccedil;&atilde;o de nitrato de s&oacute;dio ou de pot&aacute;ssio <b><sup> (6,10,11)</sup></b> que s&atilde;o utilizados na fabrica&ccedil;&atilde;o de queijos na concentra&ccedil;&atilde;o de 10 a 30 g/100L de leite. </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; No Brasil, esta adi&ccedil;&atilde;o &eacute; permitida em queijos, com exce&ccedil;&atilde;o de queijos frescos, no limite de 50 mg/kg (ppm), quantificado como &iacute;on nitrito, no produto a ser consumido. <b><sup> (12)</sup></b> </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Os nitratos t&ecirc;m efici&ecirc;ncia comprovada contra microrganismos but&iacute;ricos por tempo ilimitado e n&atilde;o possuem efeito prejudicial sobre a flora l&aacute;tica desej&aacute;vel no queijo. <b><sup> (5)</sup></b> Uma vez adicionado ao leite, ele sofre redu&ccedil;&atilde;o enzim&aacute;tica pela xantina oxidase a nitrito que inibe o crescimento e desenvolvimento do <I>Clostridium. </I><b><sup>(4,13)</sup></b> </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Por outro lado, nitratos e nitritos podem causar ef'eito t&oacute;xico em indiv&iacute;duos expostos atrav&eacute;s de alimentos, dependendo da quantidade ingerida e da susceptibilidade do organismo. Os nitritos convertem a hemoglobina a metemoglobinemia, o que afeta o transporte de oxig&ecirc;nio. Existe tamb&eacute;m a possibilidade de rea&ccedil;&atilde;o desses &iacute;ons com aminas secund&aacute;rias e terci&aacute;rias tanto "in vivo" como no pr&oacute;prio alimento, originando compostos N-nitrosos de elevado potencial carcinog&ecirc;nico, teratog&ecirc;nico e mutag&ecirc;nico. <b><sup> (14-18)</sup></b> Fabricantes de queijo que utilizam tal m&eacute;todo e reaprovcitam o SOTO pa¡'a fabricar outros produtos, como porexemplo aricota e alimentos infantis. devem monitorizar a concentra&ccedil;&atilde;o de nitrato e nitrito nestes, visto que a maioria dos &iacute;ons s&atilde;o dissolvidos e arrastados pelo sara. <b><sup> (19)</sup></b> </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; A avalia&ccedil;&atilde;o peri&oacute;dica dos teores de nitratos e nitritos em alimentos deve ser realizada a fim de que a IDA (ingest&atilde;o di&aacute;aria aceit&aacute;vel) n&atilde;o seja ultrapassada, o que colocaria em ( risco a popula&ccedil;&atilde;o. Os valores de IDA para nitratos e nitritos preconizados no Brasil e em todo Mercosul, s&atilde;o os mesmos da FAO/OMS, ou seja, de 0,06 mg/kg/ dia de nitrito (como &iacute;on) e de 3,7 mg/kg/dia para nitrato, ressaltando que a IDA n&atilde;o deve ser aplicada &agrave;s crian&ccedil;as menores de 3 meses de idade. Alimentos destinados &agrave;s crian&ccedil;as com menos de 6 meses de idade n&atilde;o podem conter nitrito como aditivo. <sup><b> (20)</b></sup> </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; N&atilde;o se tem conhecimento de trabalho desenvolvido na regi&atilde;o sul de Minas relativo &agrave; determina&ccedil;&atilde;o de nitratos e nitritos em queijo; esta regi&atilde;o do &eacute; uma das maiores produtoras de queijos do Pa&iacute;s. N&atilde;o se sabe se fabricantes est&atilde;o usando tal aditivo, e, caso sim, se os valores ultrapassam ou n&atilde;o o permitido, o que motivou a realiza&ccedil;&atilde;o deste trabalho. Para tal, a metodologia escolhida foi submetida a valida&ccedil;&atilde;o a fim de garantir a qualidade dos dados gerados. </P> <B><FONT FACE="Arial">    <P>MATERIAL E METODO</P> </FONT> </B>    ]]></body>
<body><![CDATA[<P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Foram analisadas 43 (quarenta e tr&ecirc;s) amostras de queijo, sendo 16 (dezesseis) industrializados e 27 (vinte e sete) caseiros, adquiridos em supermercados, mercearias, a&ccedil;ougues, mercados municipais, latic&iacute;nios e feiras livres de Alfenas e regi&atilde;o. Dentre as amostras de queijos caseiros, 14 eram do tipo fresco. Estas an&aacute;lises foram realizadas no per&iacute;odo de maio a junho de 1998. </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; O m&eacute;todo empregado para determina&ccedil;&atilde;o de nitrato e nitrito em queijos foi baseado nas t&eacute;cnicas descritas pelas normas da AOAC (m&eacute;todo 976.14) <b><sup> (21)</sup></b> e tamb&eacute;em segundo as Normas Anal&iacute;ticas do Instituto Adolfo Lutz, <b><sup> (22)</sup></b> s&oacute; diferindo na vidraria utilizada: ao inv&eacute;s de um funil de separa&ccedil;&atilde;o acoplado a um tubo de vidro, utilizou-se bureta de 50 mL, o que evitou a forma&ccedil;&atilde;o de bolhas de ar. </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Para otimiza&ccedil;&atilde;o do m&eacute;todo, estudou-se: a linearidade, construindo a curva padr&atilde;o de nitrito de s&oacute;dio em &aacute;gua e em queijo; a capacidade redutora da coluna de c&aacute;dmio, realizada logo ap&oacute;s a montagem da coluna pela passagem de solu&ccedil;&atilde;o padr&atilde;o de nitrato de s&oacute;dio, num total de 7 experimentos; a precis&atilde;o intra-ensaio, em adicionados com concentra&ccedil;&atilde;es de 100, 500 e 1000 mg/kg, sendo feito quintuplicatas de cada concentra&ccedil;&atilde;o; a precis&atilde;o interensaio, avaliada por an&aacute;lises de uma amostra dia-ap&oacute;s-dia durante 7 dias, conservada sob refrigera&ccedil;&atilde;o; a estabilidade da capacidade redutora da coluna de c&aacute;dmio, calculada pela determinac;:ao da capacidade redutora da coluna, 7 vezes antes do in&iacute;cio da an&aacute;lise das amostras e durante esta, por mais 4 vezes. </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Os aparelhos utilizados foram: banho-maria Modelo 100 Fanem Ltda., espectrofot&ecirc;metro de duplo feixe Micronal Modelo B493 e liq&uuml;idificador Roma Walita RI 6745. </P> <B>    <P ALIGN="JUSTIFY">RESULTADOS</P> </B>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; A trav&eacute;s do estudo da linearidade, pela constru&ccedil;&atilde;o das curvas padr&atilde;o, observou-se pequena influencia da matriz nas leituras de absorv&acirc;ncias (<a href="#fig1">Figura 1</a>).</P> <B><a name="fig1"></a><FONT FACE="Arial">    <P ALIGN="CENTER" style="line-height: 100%; margin-top: 0; margin-bottom: 0">FIGURA 1</P> </FONT> </B><FONT FACE="Arial">    <P ALIGN="CENTER" style="line-height: 100%; margin-top: 0; margin-bottom: 0">Curvas de calibra&ccedil;&atilde;o de nitrito adicionado &aacute; agua e ao queijo</P> </FONT> <FONT FACE="Arial" SIZE=2>    <P ALIGN="CENTER"><IMG SRC="/img/fbpe/alan/v50n1/art12img1.gif" WIDTH=492 HEIGHT=260 align="center"></P> </FONT>     
<P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Portanto, a quantifica&ccedil;&atilde;o de nitrito foi feita utilizando a equa&ccedil;&atilde;o de regress&atilde;o linear obtida da curva preparada pela adi&ccedil;&atilde;o de quantidades crescentes de nitrito em queijo. </P>     ]]></body>
<body><![CDATA[<P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; A capacidade m&eacute;dia redutora da coluna foi de 95,43% e a estabilidade ficou entre 93,66% e 103, 82% (m&eacute;dia 96,17%±3,71). </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; A precis&atilde;o intra-ensaio do m&eacute;todo, avaliada pela % de CV, foi de 4,90% para adicionados de 100 mg/kg de nitrito, 3,45% para 500 mg/kg e 3,41 % para 1000 mg/kg. A precis&atilde;o interensaio, para amostra com valor m&eacute;dio de 892,14 mgl<I> </I>kg±t24,18, foi de 2,71 % de CV. O limite de detec&ccedil;&atilde;o do m&eacute;todo foi estabelecido em 10 mg/kg<I>. </P> </I>    <P ALIGN="JUSTIFY">Os teores de nitratos e nitritos determinados nas amostras podem ser visualizados na <a href="#tab1">Tabela 1</a>. </P> <B><FONT FACE="Arial">    <P>DISCUSS&Atilde;O</P> </FONT> </B>    <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; O m&eacute;todo validado apresentou-se adequado para aplica&ccedil;&atilde;o na determina&ccedil;&atilde;o de nitratos e nitritos em queijos, com varia&ccedil;&otilde;es intra e interensaio aceit&aacute;veis e limite de detec&ccedil;&atilde;o 5 vezes menor de que o valor considerado limite m&aacute;ximo permitido de nitratos em queijos no Pa&iacute;s, de 50 mg/kg.<I> </I>Os procedimentos adotados foram de f&aacute;cil execu&ccedil;&atilde;o, relativamente r&aacute;pidos (aproximadamente 3 horas para cada an&aacute;lise, utilizando coluna de c&aacute;dmio), viabilizando-os para an&aacute;lise de rotina de nitrato e nitrito em queijo. </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Pode-se verificar atrav&eacute;s deste trabalho que todos os queijos que n&atilde;o apresentaram nitratos foram tamb&eacute;m negativos para nitritos. Zerfiridis &amp; Manolkidis, <sup><b> (8)</b></sup> na Gr&eacute;cia, tamb&eacute;m encontraram dados similares ou. nitrito em concentra&ccedil;&atilde;o menor que 1 mg/kg, independente do conte&uacute;do de nitrato. Outro trabalho desenvolvido no Brasil, na cidade de S&atilde;o Paulo, por Silveira et al., <sup><b> (27)</b></sup> em 1991, que analisaram 112 amostras de queijos, foi verificado que em 104 (92,85 %) n&atilde;o havia nitrato e nitrito detect&aacute;vel (LD=10 mgikg), e em 8 (7,14%) apenas o nitrato estava presente. Os baixos n&iacute;veis de nitrato e nitrito encontrados em queijos poderiam ser explicados pela alta solubilidade destes &iacute;ons em &aacute;gua, ficando pequena parte adsorvida na superficie das micelas da case&iacute;na coagulada. <b><sup> (28)</sup></b> </P>     ]]></body>
<body><![CDATA[<P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Lemieszec Chodorowska et al., citados por Devoyod, <b><sup> (4)</sup></b> analisaram 1281 amostras de queijo semiduros, e em apenas 1 % das amostras encontraram concentra&ccedil;&otilde;es acima de 100 mg/kg; em 32,5% entre 20 e 100 mg/kg; em 44,9% entre 5 e 20 mg/kg e em 21,6% abaixo de 5 mg/kg. Em rela&ccedil;&atilde;o ao nitrito, 50,3% das amostras apresentaram concentra&ccedil;&otilde;es menores de 1 mg/kg e apenas 0,3% superiores a 10 mg/kg. </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Em 1980, Zerfiridis &amp; Manolkidis <b><sup>(8)</sup></b> analisaram 54 amos tras de queijos da Gr&eacute;cia e verificaram que 46,3% n&atilde;o apresentavam nitrato e nitrito, 24% nitrato inferior a 1 mg/kg; 9,3% entre 1 a l0 mg/kg; 20,4%, entre 35 e 104 mg/kg. Pesquisaram tamb&eacute;m 69 marcas de queijos importados, sendo que o resultado encontrado mostrou grandes varia&ccedil;&otilde;es de teor: entre 1 e 508 mg/kg, sendo que 8,7% n&atilde;o apresentaram nitrato. </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Lipparini &amp; Rampa, <b><sup> (2)</sup></b> em 1983, analisaram 72 amostras de queijo para teores de nitratos e nitritos procedentes de diferentes pa&iacute;ses: It&aacute;lia, Fran&ccedil;a, Alemanha, Holanda, Dinamarca, Su&iacute;&ccedil;a e Inglaterra. As quantidades de nitrato encontradas estiveram entre 0,4 e 18,3 mg/kg. Em todas obteve-se tra&ccedil;os de nitrito at&eacute; o limite de 0,9 mg/kg. </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Baixas tamb&eacute;m foram as concentra&ccedil;&otilde;es encontradas por Karlowski &amp; Bojeski <b><sup> (29)</sup></b> para queijos poloneses, onde a m&eacute;dia de nitrato encontrada foi de 12,2 mg/kg (de nd a 131,8 mg/kg) e de nitrito de 4,2 mg/kg (de 0 a 10,0 mg/kg). </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Plutta et al., <b><sup> (30) </sup></b> analisando n&iacute;veis de nitrato e nitrito em queijo produzido na Pol&ocirc;nia, pnde 0,01 a 0,02% de nitrato de s&oacute;dio foi adicionado durante a fabrica&ccedil;&atilde;o, encontraram quantidades relativamente baixas, m&eacute;dias de 10 mg/Kg de nitrato e 1 mg/kg de nitrito. </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Su et al., <b><sup> (31)</sup></b> na China, conduziram seus estudos sobre a composi&ccedil;&atilde;o de 41 amostras de queijos importados, sendo a concentra&ccedil;&atilde;o de nitrito detectada entre 1,1 e 6,3 mg/kg.</P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Diraman, <b><sup> (32)</sup></b> em 1993, analisando 82 amostras de queijo na Turquia, onde nitrato e nitrito foram usados como agentes antimicrobianos, encontraram nitrato em concentra&ccedil;&otilde;es variando de 0,19 a 191,64 mg/kg, senda que 27 amostra continham menos que 1 mg/kg. </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; A adi&ccedil;&atilde;o de nitrato ao leite utilizado na prepara&ccedil;&atilde;o de queijo na Pol&ocirc;nia &eacute; permitido num limite m&aacute;ximo de 50 mg/kg. <b><sup> (33)</sup></b> Na Bulg&aacute;ria, nitrato de pot&aacute;ssio &eacute; adicionado de l0 a 20 g/l00 L para queijo duro e semi-duro. Esse valor de 20g/ 100L deve ser aderido &agrave; produ&ccedil;&atilde;o de queijo e tal regulamenta&ccedil;&atilde;o n&atilde;o resulta num risco de acumular nitrato, nitrito e N- nitrosanlinas no consumidor. <b><sup> (34)</sup></b> Na Holanda, &eacute; permitida a adi&ccedil;&atilde;o de 15 g de nitrato /100 L de leite e no Canad&aacute;, 20g/100 L de leite. J&aacute; na Fran&ccedil;a, Turquia, Gr&eacute;cia e It&aacute;lia, o uso de tais conservantes n&atilde;o &eacute;e permitido. <b><sup> (32, 35)</sup></b> </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; A adi&ccedil;&atilde;o de nitrato e nitrito durante a fabrica&ccedil;ao de queijo &eacute; uma necessidade tecnol&oacute;gica no Brasil, pois outros m&eacute;todos n&atilde;o contam com apoio legal, ou dependem de melhora da qualidade higienica, <sup><b> (27, 28, 32, 35, 36)</b></sup> de apoio financeiro (salvo a usinas de grande porte), de tecnologia ou mesmo da comprova&ccedil;&atilde;o de sua efici&ecirc;ncia. </P> <B><FONT FACE="Arial">    <P>CONCLUS&Atilde;O</P> </FONT> </B>     ]]></body>
<body><![CDATA[<P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; De acordo com os resultados experimentais obtidos neste trabalho, concluiu-se que: </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Grande parte dos produtores de queijo da regi&atilde;o sul de Minas Gerais, Brasil, n&atilde;o fazem uso de nitratos e nitritos como agentes conservantes ou utilizam quantidades muito baixas e os que o fazem ultrapassam os limites m&aacute;ximos permitidos (amostras 2, 3, 12 e 14). </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Apesar da maioria das amostras n&atilde;o apresentar n&iacute;veis detect&aacute;veis de nitrato e nitrito, estando portanto seguras do ponto de vista toxicol&oacute;gico, isla pode implicar num maior  risco de contamina&ccedil;&atilde;o bacteriana. </P>     <P ALIGN="JUSTIFY">&nbsp;&nbsp;&nbsp; Quando a adi&ccedil;&atilde;o de nitrato e nitrito se fizer necess&aacute;ria, e senda o limite estabelecido pela legisla&ccedil;&atilde;o brasileira coerente, deve-se promover orienta&ccedil;&atilde;o para o uso carreto. Mas, s&oacute; a jun&ccedil;&atilde;o dos esfor&ccedil;os de autoridades sanit&aacute;rias, extensionistas, produtores de leite, t&eacute;cnicos de latic&iacute;nios e pesquisadores poder&aacute; garantir bons produtos aliment&iacute;cios. </P> <B><FONT FACE="Arial">    <P>REFERENCIAS</P> </FONT> </B>     <!-- ref --><p>1.
