El propósito de este trabajo consistió en realizar una comparación entre el método propuesto por la AACC (22-90), basado en medir diferencias de pH y una técnica en la que se mide la actividad ureásica por su acción hidrolítica de la urea y posterior dosaje del amonio producido mediante la reacción de Berthelot. Se utilizaron los dos métodos para procesar 20 muestras de harina de porotos de soja sin tratar y con diferentes tratamientos térmicos de inactivación. <body><![CDATA[<P align=justify>Recibido: 01-12-2000 <FONT face="Times,Times New Roman">Aceptado: 19-06-2001</P></FONT><B>     <P align=center>INTRODUCCION</P></B>     <P align=justify>Los porotos de soja y sus derivados tienen gran riqueza en  proteínas de elevado valor biológico y razonable digestibilidad, que los  convierten en una fuente nitrogenada de reemplazo con costo sustancialmente  inferior a las proteínas de origen animal. Sin embargo, la presencia de  compuestos químicos tales como inhibidores de tripsina y de quimotripsina,  lectinas, taninos, fitatos y saponinas, denominados genéricamente antinutrientes  o factores antinutricionales (1- 4) disminuyen la eficiencia nutricional de  dietas de soja respecto a lo que se podría esperar si sólo se tuviera en cuenta  su composición química. Estos antinutrientes, de naturaleza proteica y diferente  termoestabilidad, son inactivados mediante tratamientos térmicos habituales  durante el acondicionamiento de los productos de soja antes de su consumo. La  temperatura, el tiempo de aplicación y las condiciones de humedad a las que se  lleva a cabo el tratamiento deben ser rigurosamente controladas debido a que se  corre el riesgo de ocasionar, en eventuales casos de temperaturas y tiempos  anormalmente elevados disminución de la biodisponibilidad de aminoácidos  esenciales como la lisina, con defectos nutricionales de gravedad variable. La  determinación del crecimiento en animales de laboratorio, especialmente en ratas  al destete, constituye el indicador más realista de la calidad nutricional de  productos de soja (5,6). Sin embargo esta metodología es poco factible de ser  llevada a cabo a nivel industrial, por lo que debe ser reemplazada por pruebas  más sencillas y rápidas. Entre ellas se destacan la solubilidad proteica (7) y  la actividad ureásica. </P>     <P align=justify>La actividad ureásica presente en los porotos de soja crudos  disminuye notoriamente después de los tratamientos térmicos, y su evaluación es  habitualmente empleada como indicador de la eficacia del tratamiento de  inhibición practicado.</P>     <P align=justify>El propósito de este trabajo consistió en realizar una  comparación entre el método propuesto por la AACC (22-90) -modificación del de  Caskey-Knapp (8)-, con una técnica en la que se mide la actividad ureásica por  su acción hidrolítica de la urea y posterior dosaje del amonio producido,  mediante la reacción de Berthelot (9). </P><B>     <P align=center>MATERIAL Y METODOS</P>     <P align=justify>Productos de soja</P></B>     <P align=justify>Las muestras de porotos de soja provenían de cultivos de la  zona sur de la provincia de Santa Fe. Las harinas de soja las proveyeron Galpro  SA (San Lorenzo, provincia de Santa Fe), Alpro SA (Pergamino, provincia de  Buenos Aires) y Extender S.A. (Provincia de Buenos Aires). Las provistas por  Alpro presentaban información sobre los tratamientos de inactivación a que  fueron sometidas.</P> <B>     <P align=justify>Tratamiento de las muestras</P></B>     <P align=justify>Porotos crudos: se emplearon molidos, tanto con el contenido  original de aceite como desgrasados (dos muestras) por medio de extracción con  hexano normal en frío.</P>     ]]></body>
<body><![CDATA[<P align=justify>Porotos tratados: muestras de 250 g de porotos crudos fueron  sometidas a tratamientos en el laboratorio. Estos consistieron en: (a) Remojo en  agua destilada durante dos horas y hervor en agua destilada en recipiente de  vidrio abierto durante 5, 10, 20 y 45 minutos, molienda y secado a 37 ºC con  corriente de aire. (b) Autoclavado: 5 minutos a vapor fluente; 5, 10 y 15  minutos a 1 atmósfera, molienda y secado a 37 º C (c) Aire seco a 100 ºC,  molienda.</P>     <P align=justify>Harinas comerciales: Galpro (Harinas G): inactivadas  industrialmente, enteras y desgrasadas. Alpro (Harinas A): desgrasada con vapor  de agua; tratada 2 minutos a 152 ºC calor seco; aire seco a 140 ºC; vapor 15  minutos a 1 atmósfera; vapor 5 minutos a 5 atmósferas. Extender: desgrasada,  inactivada industrialmente.</P> <B>     <P align=justify>Preparación de los extractos</P></B>     <P align=justify>Las muestras de porotos molidos y de harinas de soja tratadas  con distintas metodologías se utilizaron para preparar extractos según la  metodología descripta por Liener para la determinación de la actividad  hemaglutinante de productos de soja (10). Se incubaron 5g de productos de soja  con 25 ml de NaCl 0.9% estéril durante 1 hora entre 4-8ºC y centrifugaron  durante 15 minutos a 3500 rpm. Los extractos obtenidos se emplearon para evaluar  la actividad ureásica mediante el nuevo método.</P><B>     <P align=justify>Descripción de la metodología</P></B>     <P align=justify>Método de referencia: AACC Método 22-90 (Método de Caskey-  Knapp modificado): medida de la diferencia de pH registrada entre 200 mg de una  muestra de productos de soja en 10 ml de buffer de fosfatos 0.05 M de pH 7.0  conteniendo 3g% de urea y 200 mg de la misma muestra en 10 ml de buffer sin urea  luego de la incubación durante 30 minutos a 30º C. </P>     <P align=justify>Nuevo método:</P>     <P align=justify>Reactivos<B>:</B> (a) Sustrato:Urea 0.01M </P>     <P align=justify>(b) Reactivos de Color: (I) Fenol 532mmol/l; Nitroprusiato de  Na 0.85 mmol/l; etilén bis ditiocarbamato manganoso 0.3 mmol/l</P>     <P align=justify>(II) NaClO 36,6 mmol/l; p-toluén sulfoncloramida 0,12 mmol/l;  NaOH 0.625 mmol/l</P>     ]]></body>
<body><![CDATA[<P align=justify>Técnica: 20 ul de extracto se emplean como reactivo enzimático  incubándose con 20 ul de sustrato durante 5 minutos a 37 ºC. Se agregan 0.5 ml  de Reactivo I y 0,5 ml de Reactivo II. Se incuba 5 minutos y se diluye con 8 ml  de H20 destilada. Se efectúa la lectura espectrofotométrica a 540 nm. Los  valores de la lectura se corrigen teniendo en cuenta la cantidad de harina que  representa el volumen de extracto y refiriéndola a 200 mg, cantidad de muestra  empleada en el método de referencia.</P>     <P align=justify>La actividad ureásica, que se cuantifica por medio de la  concentración de amonio producido, se compara con la actividad desarrollada por  un extracto de soja comercial provista por laboratorios Wiener a la que se le  asigna 100% de actividad.</P> <B>     <P align=justify>Evaluación del límite de detección y la sensibilidad del método  propuesto</P></B>     <P align=justify>Para evaluar el límite de detección y la sensibilidad del  método propuesto así como la posibilidad de diferenciar entre muestras con y sin  actividad ureásica residual se efectuaron diluciones de la ureasa comercial  provista por Wiener laboratorios con solución fisiológica y se usaron 20 ul de  las mismas reemplazando a las muestras en el caso de la reacción de la AACC y en  reemplazo de los extractos en el caso de la técnica propuesta.</P> <B>     <P align=justify>Evaluación de la actividad ureásica en muestras de productos de  soja</P></B>     <P align=justify>La actividad ureásica residual de 20 muestras de porotos y  harinas de soja sometidas a diferentes tratamientos fue evaluada mediante las  dos técnicas. En el caso de la técnica propuesta se expresó la actividad  ureásica de las diferentes muestras como porcentaje respecto de la actividad de  la ureasa comercial sin diluir a la que se le atribuyó 100% de actividad.</P>     <P align=justify>Los valores de actividad residual expresan la eficacia de los  distintos tratamientos, que según varios autores (11,12) se relacionan con la  humedad, tiempo y temperatura a que se llevan a cabo.</P> <B>     <P align=justify>Análisis estadístico</P></B>     <P align=justify>Los datos obtenidos fueron analizados mediante la aplicación de  correlación lineal con análisis de la linealidad. Se utilizó el programa  estadístico Instat.</P> <B>     <P align=center>RESULTADOS Y DISCUSION</P></B>     ]]></body>
<body><![CDATA[<P align=justify>En la <A HREF="#Tab1">Tabla 1</A> se detallan los resultados obtenidos en el  experimento de evaluación del límite de detección y de la sensibilidad para  detectar actividad de ureasa por medio del nuevo método. Se verifica que hasta  una dilución 1/10 de la enzima comercial se mantiene un nivel de respuesta  similar al de la original. El límite de detección del método propuesto alcanza  hasta niveles de 0.004 partes de ureasa, nivel en el que el método basado en la  determinación de diferencia de pH no acusa respuesta y que por lo tanto  resultaría en informar esa muestra como sin actividad ureásica residual. Los datos son un promedio de tres o cuatro determinaciones y se expresan  como promedio y error estándar del promedio. Los resultados obtenidos permiten  visualizar la relación inversa entre la temperatura, la humedad y el tiempo de  tratamiento con los niveles de actividad ureásica residual. Así se puede  observar que los tratamientos con calor húmedo ocasionan mayor inactivación y  que el remojado previo al hervor en recipiente abierto resulta más efectivo que  el tratamiento con vapor fluente sin remojo previo. Estos hallazgos coinciden  con lo observado por Kabara y otros (13). También resulta interesante el hecho  de que el tratamiento con aire a 100º C no produce disminución de la actividad  ureásica comparada con la que presentaron los porotos sin tratar.</P>     <P align=justify>Los resultados de actividad ureásica residual obtenidos por los  dos métodos fueron analizados estadísticamente y se obtuvo un coeficiente de  correlación de 0.9416 (p<FONT face=Symbol>&lt;</FONT> 0.0001), Test de Pearson.  Empleando el test de Spearman se obtuvo un coeficiente de correlación de 0.7957  (p<FONT face=Symbol>&lt;</FONT> 0.0001). Los valores de error estándar del  promedio de los replicados para las distintas muestras se muestran en la <a A HREF="#Tab2">Tabla  2</A>. <body><![CDATA[<P align=justify>Los resultados obtenidos nos permiten concluir que el límite de  detección del método propuesto alcanza hasta niveles de 0.004 partes de ureasa,  nivel en el que el método basado en la determinación de diferencia de pH no  acusa respuesta. La sensibilidad del método propuesto permite diferenciar  satisfactoriamente muestras con actividades residuales correspondientes a  diferencias en 0.001 partes de ureasa. La correlación obtenida entre el método  de la AACC 22- 90 y el propuesto es altamente significativa. La precisión  alcanzada en la metodología propuesta resulta satisfactoria como se aprecia en  los valores obtenidos para error estándar. Asimismo, el nuevo método presenta  mayor especificidad, ya que se mide la concentración del producto de la  reacción.
