En los últimos años, el uso del horno de microondas dentro de la industria alimentaria ha ganado popularidad. Entre sus usos se incluye el descongelar, secar y cocinar alimentos, mas la inactivación de microorganismos que pueda o no ejercer este tipo de tratamiento es tema de discusión mundial. Por otro lado, la Escherichia coli O157:H7 representa un patógeno emergente, de distribución mundial y asociado a alimentos. Su resistencia a ambientes adversos ha sido ampliamente discutida. El propósito de este estudio fue determinar el efecto de diferentes tiempos e intensidades de cocimiento del horno de microondas sobre la sobrevivencia de esta bacteria en tortas de carne de res. Las tortas de carne fueron inoculadas con una población alta (10(7)-10(9) UFC/mL) o baja (10(5)-10(7) UFC/mL) de E. coli O157:H7, mantenidas en congelación por 3 días a -4ºC y posteriormente descongeladas en un horno Whirlpool según su peso. Fueron sometidas a niveles de potencia de 70%, 80%, 90% y 100% por períodos de 30, 60, 90 y 120 segundos. Se determinó en cada muestra la tasa de sobrevivencia de la bacteria inoculada de acuerdo a la metodología descrita en Vanderzant & Splittstoesser. Según los resultados obtenidos, la tasa de destrucción de las bacterias analizadas fue significativa (p<0,005). El número de bacterias presentes en las tortas de carne disminuyó dramáticamente a medida que aumentaba el tiempo y la temperatura de exposición, no obstante, para eliminar totalmente el microorganismo inoculado se necesitó de una exposición prolongada produciendo características organolépticas indeseables en el alimento.]]></p></abstract>

It is used for thawing, drying and cooking food, but the microorganism’s inactivation that this treatment may exert or not is still a subject of worldwide discussion. At the same time, Escherichia coli O157:H7 now presents itself as an emerging pathogen, distributed worldwide and associated with food. Its resistance to adverse environments has been widely discussed. The purpose of this work was to determine the effect of different times of exposure and cooking intensities of microwave oven on the survival of this bacterium inoculated into minced meat samples. These were inoculated with a high (10(7)-10(9) CFU/mL) or low (10(7)-10(7) CFU/mL) population of E. coli O157:H7, frozen for 3 days at -4ºC and thawed in a Whirlpool microwave oven according to their weight. They were radiated at levels of 70%, 80%, 90% and 100% for periods of 30, 60, 90 and 120 seconds. In each sample the rate of survival of the bacteria was determined according to the methodology described by Vanderzant & Splittstoesser. The results obtained showed that the rate of destruction of the bacteria analyzed was significant (p<0,005). Entre sus usos se incluye el descongelar, secar y cocinar alimentos, mas la inactivaci&oacute;n de microorganismos que pueda o no ejercer este tipo de tratamiento es tema de discusi&oacute;n mundial (1,2).</font></P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Diversos trabajos citan la reducci&oacute;n en el n&uacute;mero de microorganismos al ser tratados con microondas, incluyendo pavo, carne, leche de soya, pollo, papa y alimentos congelados (3-5), pero no su destrucci&oacute;n total; otros estudios citan la sobrevivencia de microorganismos a este tipo de tratamiento, incluyendo bacterias tan importantes en Microbiolog&iacute;a de Alimentos como <I>Salmonella</I> (6) <I>Clostridium perfringens</I> y <I>Bacillus cereus</I> (2,7).</font></P> <I>    <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Escherichia coli</font></I> <font size="3" face="Times New Roman"> O157:H7 representa un pat&oacute;geno emergente, y de distribuci&oacute;n mundial.  Fue por primera vez reconocido como pat&oacute;geno de origen alimentario en 1982, cuando fue asociado a dos brotes de colitis hemorr&aacute;gica en los Estados Unidos (8).  Adem&aacute;s de este cuadro cl&iacute;nico, puede presentar otros dos tipos de manifestaci&oacute;n, incluyendo p&uacute;rpura trombocitop&eacute;nica y s&iacute;ndrome ur&eacute;mico hemol&iacute;tico (9-11).</font></P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Esta bacteria puede ser transmitida por alimentos o agua, directamente de persona a persona y ocasionalmente por exposici&oacute;n ocupacional (12).  La mayor&iacute;a de brotes alimentarios han sido asociados a alimentos derivados del ganado, especialmente carne molida y leche cruda (13), no obstante, frutas, vegetales y alimentos derivados de otras especies animales tambi&eacute;n pueden transmitir este pat&oacute;geno.</font></P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">La frecuencia de recuperaci&oacute;n de esta bacteria a partir de alimentos es muy baja, as&iacute; como bajo es el nivel de contaminaci&oacute;n de alimentos asociados a brotes (10-6200 UFC/g) (14) y la dosis infecciosa descrita (tan baja como 2 c&eacute;lulas/25g).</font></P>     ]]></body>
<body><![CDATA[<P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Tambi&eacute;n, est&aacute;n bien descritas en la literatura las caracter&iacute;sticas de resistencia y sobrevivencia de esta bacteria ante ambientes adversos.  Diversos estudios indican la reducci&oacute;n en 1-2 logaritmos en el n&uacute;mero de <I>E. coli</I> O157:H7 presente en alimentos fermentados o secados (15), refieren tambi&eacute;n la capacidad de multiplicarse a muy bajas temperaturas, a&uacute;n congelaci&oacute;n (16) y su resistencia a ambientes de baja actividad de agua (aw) (17) y pH (18-19).</font></P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Dado que la carne molida mal cocida o cruda ha sido descrita como el principal veh&iacute;culo de transmisi&oacute;n de esta bacteria (20), y que en Costa Rica ha habido al menos siete casos cl&iacute;nicos  relacionados con esta bacteria (21), as&iacute; como que el cocinar en horno de microonda ofrece cortos per&iacute;odos de calentamiento y menos tiempo de exposici&oacute;n al calor que el tratamiento convencional,  se propone el presente estudio, cuyo objetivo es estudiar el efecto de diferentes tiempos y poderes de cocci&oacute;n del horno de microondas sobre la sobrevivencia de <I>E. coli</I> O157:H7 inoculada en tortas de carne de res.</font></P> <B>    <P ALIGN="left"><font size="3" face="Times New Roman">MATERIALES Y METODOS</font></P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Ubicaci&oacute;n</font></P> </B>    <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">El proyecto se llev&oacute; a cabo en el Laboratorio de Microbiolog&iacute;a de Alimentos y Aguas de la Facultad de Microbiolog&iacute;a entre julio y diciembre del 2001.</font></P> <B>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Alimento</font></P> </B>    <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Los ensayos se realizaron sobre muestras de carne molida adquiridas en supermercados ubicados en el &aacute;rea metropolitana de San Jos&eacute;, Costa Rica.   Todos los ejemplares fueron mantenidos entre 2 y 5ºC entre el tiempo de compra  y su an&aacute;lisis, el cual no excedi&oacute; las 4 horas y tratados en forma as&eacute;ptica con el fin de no aumentar su carga.  En cada caso se corri&oacute; a la vez un blanco de muestra con el fin de evaluar la flora natural del producto y descartar la presencia de <I>E. coli</I> O157:H7</font></P> <B>    <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">In&oacute;culo</font></P> </B>    <P ALIGN="JUSTIFY"> <font size="3" face="Times New Roman"> Se utiliz&oacute; una cepa de <I>Escherichia coli</I> O157:H7de origen cl&iacute;nico para los an&aacute;lisis.  Esta fue mantenida a -70ºC en agar tripticase soya (Oxoid) y activada en caldo tripticase soya (pH 7,0) a 35ºC por 24 horas.  Esta &uacute;ltima suspensi&oacute;n  fue considerada como la poblaci&oacute;n alta (10<SUP>7</SUP>-10<SUP>9 </SUP>y  UFC/mL) de <I>E. coli</I> O157:H7 utilizada para inocular los alimentos.  Una suspensi&oacute;n de poblaci&oacute;n baja (10<SUP>7</SUP>-10<SUP>7</SUP> UFC/mL) fue preparada agregando 1 mL de la suspensi&oacute;n de poblaci&oacute;n alta a 99 mL de caldo tripticase soya.</font></P> <B>    <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Inoculaci&oacute;n de la carne</font></P> </B>    ]]></body>
<body><![CDATA[<P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Aproximadamente 1 kg de carne molida colocado dentro de una bolsa de polietileno est&eacute;ril fue inoculado con 50 mL  de la suspensi&oacute;n de poblaci&oacute;n alta de <I>E. coli</I> O157:H7.  Este fue homogenizado por 2 min en un masticador (Stomacher) e inmediatamente dividido en tortas de 50 g cada una, todas con igual di&aacute;metro y espesor.  Las tortas fueron congeladas a -4ºC  por 72 horas y despu&eacute;s fueron procesadas en horno de microondas, simulando el uso industrial, hotelero o dom&eacute;stico del producto.</font></P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Una porci&oacute;n de 25 g de carne fue tomada antes de la inoculaci&oacute;n con el fin de realizar un recuento inicial de <I>E. coli</I> O157:H7.</font></P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">El mismo procedimiento fue realizado con otros 1000 g de carne molida, pero inoculados con 50 mL de la suspensi&oacute;n de poblaci&oacute;n baja de <I>E. coli</I> O157:H7. Cada an&aacute;lisis fue realizado en tres ocasiones diferentes.</font></P> <B>    <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Protocolo de cocimiento</font></P> </B>    <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Se utiliz&oacute; un horno de microondas Whirlpool   modelo MT 11515 GQ-O con diez niveles de energ&iacute;a para descongelar y cocinar las muestras.  Se procedi&oacute; a descongelarlas individualmente seg&uacute;n su peso, despu&eacute;s de lo cual fueron sometidas a 70% (76,7ºC), 80% (82,8ºC), 90% (87,8ºC) y 100% (93,3ºC) de poder durante 30, 60, 90 y 120 segundos cada una.  Se utiliz&oacute; una muestra control sin inocular  (blanco de carne) y otra inoculada pero no tratada con microondas, como controles.</font></P> <B>    <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Cuantificaci&oacute;n de los microorganismos</font></P> </B>    <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Para la determinaci&oacute;n microbiol&oacute;gica se sigui&oacute; la metodolog&iacute;a de N&uacute;mero M&aacute;s Probable (NMP)  seg&uacute;n se describe en Vanderzant &amp; Splittstoesser (22).  Brevemente, luego del proceso de cocci&oacute;n, 25 g de cada muestra fueron resuspendidos en 225 mL de agua peptonada est&eacute;ril (APE) 0,1% y de nuevo homogenizados en Stomacher por 2 minutos.   A partir de esta suspensi&oacute;n se prepar&oacute; diluciones decimales hasta 10<SUP>-7</SUP> en tubos con 9 ml APE 0,1%.  Se preenriqueci&oacute;  cada diluci&oacute;n, por triplicado en caldo Escherichia coli (EC) + novobiocina incubado por 24 h a 35ºC.  Se confirm&oacute; el crecimiento utilizando agar McConkey sorbitol + 4-metil umbeliferil &#946; D glucor&oacute;nido (MUG) 0,2 g/L (Oxoid) incubado  a 35ºC por 24 h.</font></P> <B>    <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Evaluaci&oacute;n de la sobrevivencia de las bacterias</font></P> </B>    <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Se evalu&oacute; la sobrevivencia de las bacterias relacion&aacute;ndola con el poder (temperatura de cocci&oacute;n) y tiempo de cocci&oacute;n.</font></P> <B>    <P ALIGN="left"><font size="3" face="Times New Roman">RESULTADOS</font></P> </B>    ]]></body>
<body><![CDATA[<P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">En la <a href="#TABLA 1"> Tabla 1</a> se detalla el comportamiento de la poblaci&oacute;n alta de <I>E. coli</I> O157:H7 al ser tratada con diferentes intensidades de poder en el horno de microondas.  La carga inicial fue de 10<SUP>7</SUP> NMP/g, la cual se mantiene a los 30 s para los poderes 70%, 80% y 90%.  En la intensidad de 100% hay un descenso de un logaritmo en el n&uacute;mero de microorganismos en este lapso de tiempo. A los 60 s de tratamiento, en las intensidades de 90% y 100% ya no se detecta microorganismos viables.  Para el 70% de poder se necesita  m&aacute;s de 120 s para inactivar las bacterias presentes.</font></P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">La poblaci&oacute;n baja (10<SUP>5</SUP> NMP/g) necesit&oacute; de  60 s a  90% de poder y 90 s a 80% de poder para ser no detectable. <body><![CDATA[<P ALIGN="JUSTIFY">&nbsp;</P> <B>    <P ALIGN="left"><font size="3" face="Times New Roman">DISCUSION</font></P> </B>    <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Son muchas las investigaciones realizadas a nivel mundial sobre el efecto de hornos de microondas en la sobrevivencia de bacterias pat&oacute;genas inoculadas en diferentes alimentos. Los resultados son contradictorios, mostrando variaciones importantes seg&uacute;n sea la composici&oacute;n del alimento, el microorganismo evaluado y hasta la fase de crecimiento en que &eacute;ste se encuentre (3-7). Algunos autores  como Fujikawa <I>et al</I>. citan que este tratamiento es equivalente al cocimiento convencional en lo que respecta a inactivaci&oacute;n de microorganismos  (23).  Otros, como Rosenberg y Bogl citan en su estudio diferentes grados de eficiencia entre el cocimiento tradicional y el de microondas de hasta dos logaritmos en la tasa de muerte de bacterias Gram positivas y Gram negativas (24).</font></P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Seg&uacute;n los resultados obtenidos, la tasa de destrucci&oacute;n de las bacterias analizadas fue significativa (p&lt;0,005).  El n&uacute;mero de bacterias presentes en  las tortas de carne disminuy&oacute; dram&aacute;ticamente a medida que aumentaba el tiempo y la temperatura de exposici&oacute;n, tanto para poblaci&oacute;n alta como para la baja. No obstante, se necesit&oacute; de largos per&iacute;odos de exposici&oacute;n al tratamiento t&eacute;rmico para llegar a eliminar completamente la bacteria inoculada.</font>  </P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Estudios previos realizados con este mismo modelo de torta de carne tratada en horno de microondas determinaron, a trav&eacute;s del indicador enzim&aacute;tico de  inhibici&oacute;n de la fosfatasa &aacute;cida, que se necesita al menos 60 s de tratamiento (independiente del poder utilizado) para considerar al alimento enzim&aacute;ticamente cocinado.  Es decir, luego de 60 s de tratamiento t&eacute;rmico en microondas desaparece esta enzima, lo cual coincide con las caracter&iacute;sticas organol&eacute;pticas de carne cocinada en todo su espesor (25).  Para las muestras analizadas en el presente trabajo, en promedio se necesit&oacute; de un tiempo mayor de cocimiento al descrito para eliminar totalmente el microorganismo inoculado (poblaci&oacute;n alta y baja), lo cual provoca que la torta de carne presente caracter&iacute;sticas organol&eacute;pticas indeseables como son la resequedad y dureza.</font></P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Los resultados obtenidos permiten evidenciar que la efectividad de este tipo de tratamiento es inversamente proporcional al n&uacute;mero inicial de microorganismos, no obstante,  las temperaturas internas alcanzadas en los alimentos  no garantizan la esterilizaci&oacute;n de los  mismos (26).  La distribuci&oacute;n de calor que presentan los hornos de microondas no es homog&eacute;nea (27), por lo que un aumento en la temperatura no es del todo uniforme.  Adem&aacute;s, dada la presencia de prote&iacute;nas y grasa en el alimento, existe mucha  fluctuaci&oacute;n en  las temperaturas internas del alimento (28).</font></P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Estudios similares llevados a cabo con <I>E. coli</I> O157:H7 inoculada en carne cruda demuestran  que se da una reducci&oacute;n importante  en el n&uacute;mero de microorganismos presentes al cocinar con microondas, lo cual permite extender el tiempo de preservaci&oacute;n del alimento (29), mas el efecto de las microondas sobre esta bacteria var&iacute;a seg&uacute;n sea la dosis de irradiaci&oacute;n utilizada, as&iacute; como la temperatura interna alcanzada (30).  Es importante destacar que dadas las caracter&iacute;sticas de patogenicidad de esta bacteria y su baja dosis infectante (&lt;50 UFC/g), su simple  presencia en alimentos procesados es totalmente inaceptable (31).</font> </P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">A&uacute;n cuando los niveles de inoculaci&oacute;n utilizados en este estudio son poco probables de ocurrir, se puede concluir que la carne contaminada con <I>E. coli</I> O157:H7  representa un riesgo para la salud p&uacute;blica a&uacute;n cuando se mantenga a temperatura de congelaci&oacute;n  y luego se cocine en microondas. Al igual que con otros microorganismos pat&oacute;genos, este tipo de tratamiento t&eacute;rmico no es efectivo para su eliminaci&oacute;n a partir de alimentos contaminados. La implementaci&oacute;n de Buenas Pr&aacute;cticas de Manufactura (GMP) as&iacute; como de An&aacute;lisis de Riesgos y Puntos Cr&iacute;ticos de Control (HACCP) en la producci&oacute;n de alimentos y su preparaci&oacute;n puede ayudar a controlar esta bacteria.</font></P>     <P ALIGN="JUSTIFY"><font size="3" face="Times New Roman">Por otra parte es imprescindible que el consumidor est&eacute; informado del riesgo a la salud asociado al consumo de carne que no est&eacute; bien cocinada o que solamente sea calentada antes de su consumo.</font></P> <B>    <P ALIGN="left"><font size="3" face="Times New Roman">AGRADECIMIENTO</font></P> </B>     ]]></body>
