The method is based on the dissolution, or extraction, of the active principle with chloroform and direct analysis of standard and sample solutions by FTIR spectrometry. Quantification of the analyte was carried out at the 1512 (principal band) and 1300 cm-1 bands, using as analytical signal the absorbance at 1512 and 1300 cm-1, respectively, corrected by mean appropriate baselines. The main figures of merit at the principal band (working range 0.1-1.0% (w/v), limit of detection (3 &#963;): 0.004% (w/v), limit of quantification (10 &#963;): 0.0125% (w/v), precision: 0.6% and sample throughput of 30 samples h-1) are attractive and really adequate for the proposed analysis. The accuracy of the method was certified by means comparative analysis of real samples using the official method described by the United Stated of America Pharmacopeia (USP). The method was satisfactorily applied to the determination of atenolol in a series of commercially available solid pharmaceutical samples. All the values obtained for the commercial samples were within the limit prescribed by the official pharmacopoeia.]]></p></abstract>
<abstract abstract-type="short" xml:lang="es"><p><![CDATA[En este trabajo se desarrolla una propuesta metodológica alternativa, simple y rápida, para la determinación de atenolol en productos farmacéuticos. El método se basa en la disolución, o extracción, del principio activo con cloroformo y análisis directo de los estándares y muestras por espectrometría infrarroja con transformada de Fourier. La cuantificación del analito se llevó a cabo, en paralelo, en las bandas del analito centradas en 1512 (banda principal) y 1300 cm-1, utilizando como señal analítica la absorbancia a 1512 y 1300 cm-1, respectivamente, corregida mediante líneas de base apropiadas. Las figuras de mérito del método en la banda principal (intervalo de trabajo: 0,1-1,0% (p/v), límite de detección (3&#963;): 0,004% (p/v); límite de cuantificación (10 &#963;): 0,0125% (p/v), precisión: 0,6% y una frecuencia de análisis de 30 muestras h-1) resultan adecuadas para realizar el análisis propuesto. La exactitud del método se certificó mediante análisis comparativos de muestras reales con el método oficial descrito en la Farmacopea de los Estados Unidos de América (USP). Se analizaron, de forma satisfactoria, diversos productos farmacéuticos que se comercializan en el país. Introducci&#243;n</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> El atenolol, 2-[p-[hidroxi-3-(isopropilamino)propoxi]-fenil]acetamida (<a href="#fig1">Figura  1</a>), es un antagonista </FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Symbol"> b</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana">-adrenaceptor, perteneciente al grupo de los </FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Symbol"> b</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana">-bloqueadores.  Este tipo de drogas se utiliza, generalmente, en el tratamiento de des&#243;rdenes  cardiovasculares como: arritmia cardiaca, infarto del miocardio, paro card&#237;aco  congestivo, migra&#241;a e hipertensi&#243;n [1, 2].</FONT></P>     <P ALIGN="center"><a name="fig1"> <img border="0" src="/img/fbpe/rtfiuz/v32n3/art07fig1.gif" width="304" height="169"></a></P>     
<P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Este principio activo farmac&#233;utico (PAF) fue introducido en el mercado  en el a&#241;o 1976, en reemplazo del propanolol, como anti-hipertensivo. Se  administra por inyecci&#243;n intravenosa o por v&#237;a oral, mediante jarabes o  tabletas con un contenido declarado que oscila entre 50 y 100 mg. En el  tratamiento de la hipertensi&#243;n se puede suministrar de forma aislada, o  en preparaciones compuestas con otros antihipertensivos de tipo diur&#233;tico  [2]. Al igual que otros </FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Symbol"> b</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana">-bloqueadores, el atenolol ha sido usado como  agente de dopaje en actividades deportivas; raz&#243;n por la cual el Comit&#233;  Ol&#237;mpico Internacional (COI) lo ha incluido en la lista de sustancias prohibidas  [3].</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Como cualquier producto farmac&#233;utico, los f&#225;rmacos que contienen atenolol  deben pasar por un estricto control de calidad que verifique el contenido  nominal del PAF, as&#237; como su uniformidad. Esto con el objeto de garantizar  su eficacia.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> La Farmacopea de los Estados unidos de Am&#233;rica (USP) propone para este  fin un m&#233;todo HPLC con detecci&#243;n UV [4]. Los m&#233;todos cromatogr&#225;ficos, por  lo general, son precisos y exactos. Sin embargo, requieren instrumentaci&#243;n  y accesorios costosos; y suelen tener un consumo elevado de reactivos,  solventes y tiempo de an&#225;lisis. Adem&#225;s, algunos de ellos han sido descritos  como tediosos. Por estas razones, el desarrollo de nuevos m&#233;todos anal&#237;ticos  &#150;simples, accesibles, sensibles y selectivos&#150; para el an&#225;lisis de PAF en  f&#225;rmacos representa una l&#237;nea de investigaci&#243;n atractiva y de mucha vigencia.  En esa v&#237;a, los nuevos dise&#241;os metodol&#243;gicos hacen &#233;nfasis en algunos aspectos  fundamentales, entre los que destacan: i) an&#225;lisis directo (sin pre-tratamientos  complejos de la muestra), ii) instrumentaci&#243;n accesible y de bajo costo  (de adquisici&#243;n y mantenimiento), iii) tiempos de an&#225;lisis cortos, iv)  m&#237;nimo consumo de reactivos, solventes y muestras (qu&#237;mica verde) y v)  que sea, o pueda ser, f&#225;cilmente automatizado. En la b&#250;squeda de estos  objetivos, se han propuesto m&#233;todos alternativos basados en t&#233;cnicas electroanal&#237;ticas  (potenciometr&#237;a [5], voltametr&#237;a [6, 7], etc.) y espectrosc&#243;picas (Absorci&#243;n  Molecular UV-visible [8-11], Fluorescencia Molecular [12-14] y Espectroscopia  Infrarroja en el IR cercano (NIR) [15, 16], etc.), entre otras.</FONT></P>     ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> El atenolol presenta bandas de absorci&#243;n caracter&#237;sticas en el UV y en  el infrarrojo medio (MIR). Sin embargo, en conocimiento de los autores  &#150;hasta el momento&#150; no se ha descrito ning&#250;n m&#233;todo para su determinaci&#243;n  en productos farmac&#233;uticos, basado en la espectrometr&#237;a infrarroja con  transformada de Fourier (FTIR), con medidas de transmisi&#243;n en el infrarrojo  medio (MIR).</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> La espectrometr&#237;a FTIR es una t&#233;cnica espectrosc&#243;pica poderosa y vers&#225;til,  que ofrece selectividad y especificidad. Por ello, se ha convertido en  una herramienta muy &#250;til para el an&#225;lisis de innumerables sustancias en  muestras complejas, sin la necesidad de tratamientos complejos previos  a la muestra [17]. En las &#250;ltimas dos d&#233;cadas se ha utilizado &#150;de forma  extensa y exitosa&#150; en la industria farmac&#233;utica, tanto en el &#225;rea de investigaci&#243;n  como de control [18, 19]. La sinergia entre &#233;sta t&#233;cnica (FTIR) y la industria  farmac&#233;utica se debe a varios factores, entre los que destacan: i) la mayor&#237;a  de los PAF absorben radiaci&#243;n IR, ii) la evoluci&#243;n de la t&#233;cnica &#150;en instrumentaci&#243;n  y accesorios&#150; ha brindado soluciones efectivas a muchos de sus problemas  y iii) la concentraci&#243;n de los PAF en los productos farmac&#233;uticos suele  ser elevada.</FONT></P> </MULTICOL> <MULTICOL GUTTER="39" COLS="2">     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> El principal objetivo de este trabajo fue desarrollar un m&#233;todo alternativo  &#150;simple, r&#225;pido y accesible&#150; basado en la Espectrometr&#237;a FTIR, para la  determinaci&#243;n de atenolol en formulaciones farmac&#233;uticas s&#243;lidas; cuyas  caracter&#237;sticas b&#225;sicas lo hacen competitivo frente al m&#233;todo oficial de  an&#225;lisis y accesible a todos los laboratorios de control de la industria  farmac&#233;utica.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>2. Parte Experimental</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>1.1. Instrumentaci&#243;n</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Todos los espectros de absorci&#243;n fueron adquiridos con un espectr&#243;metro  FTIR Perkin Elmer (PE), modelo Spectrum 2000 (Nolwack, CT. USA) equipado  con una fuente de radiaci&#243;n PE para trabajar en el MIR, un divisor &#243;ptico  intercambiable (<I>beamspletter</I>) de KBr, un detector de respuesta r&#225;pida de  mercurio-cadmio-telurio (MCT-MIR) y una celda IR de transmisi&#243;n est&#225;ndar  (CTIR) de geometr&#237;a circular Wilmad (New Jersey, USA) con ventanas circulares  (32 &#215; 2 mm) de ZnSe. Esta celda es de tipo desmontable, con paso &#243;ptico  variable. Este par&#225;metro se fij&#243; en 0,5 mm con un espaciador de PTFE (Tefl&#243;n).  El control del equipo, as&#237; como la adquisici&#243;n y procesamiento de los espectros,  se llev&#243; a cabo con el software Spectrum 2000, de PE.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> La unidad de trabajo se mantiene con un ambiente controlado &#150;mediante acondicionadores  de aire y des-humificadores&#150; a una temperatura de 25 &#177; 2&#176;C y una humedad  relativa de 45 &#177; 5%.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Para la preparaci&#243;n de las muestras se utiliz&#243; un ba&#241;o ultras&#243;nico Branson,  modelo 2100 (USA) y para el filtrado se us&#243; un sistema de guarda columna  con membranas de <I>nylon</I> de 13 mm de di&#225;metro y 2 &#181;m de tama&#241;o de poro.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>2.2. Reactivos, est&#225;ndares y muestras</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Los reactivos utilizados en este trabajo fueron de grado anal&#237;tico. Se  utiliz&#243; cloroformo estabilizado con etanol, calidad HPLC (T.J. Baker Xalostoc,  M&#233;xico), como disolvente. Previo al an&#225;lisis, el disolvente fue destilado  y secado por varias horas sobre CaCl</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUB>2</SUB></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2"> anhidro (BDH, Poole, Inglaterra)  [20, 21]. Se prepar&#243; una soluci&#243;n patr&#243;n (1% (p/v)) de atenolol (Sigma  Aldrich, 99,5% de pureza, St. Louis, USA), disolviendo la cantidad apropiada  en cloroformo. Los est&#225;ndares de trabajo, entre 0,1 y 1,0% (p/v), fueron  preparados en cada sesi&#243;n de trabajo por diluci&#243;n adecuada del patr&#243;n en  el mismo medio.</FONT></FONT></P>     ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Las muestras analizadas corresponden a productos farmac&#233;uticos comerciales  provenientes de cinco Laboratorios Farmac&#233;uticos nacionales, que se encuentran  disponibles en el mercado local. La presentaci&#243;n comercial de las mismas  corresponde a tabletas que declaran un contendido de 50 y 100 mg del principio  activo.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Para la preparaci&#243;n de las soluciones-muestra se toman inicialmente 10  tabletas del producto farmac&#233;utico, se trituran y pulverizan &#150;de forma  manual&#150; con la ayuda de un mortero, hasta obtener un fino polvo homog&#233;neo  representativo de la muestra. Se pesa la cantidad de polvo equivalente  a 100 &#243; 50 mg de atenolol, y se coloca en un tubo de ensayo con tapa. A  continuaci&#243;n se lleva a cabo el proceso de extracci&#243;n s&#243;lido-l&#237;quido con  20 mL de cloroformo, colocando el recipiente en un ba&#241;o ultras&#243;nico por  15 minutos. Finalmente, la soluci&#243;n resultante se filtra para separar los  excipientes insolubles, se transfiere el filtrado a un matraz de 25 mL  y se lleva a aforo con el mismo disolvente. En este punto, la soluci&#243;n-muestra  est&#225; lista para el an&#225;lisis por FTIR.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Para el an&#225;lisis cromatogr&#225;fico de referencia, las muestras y la fase m&#243;vil  (1-heptnosulfonato de sodio/fosfato dib&#225;sico de sodio anhidro/ butilamina)  se prepararon siguiendo rigurosamente el protocolo descrito en la USP [4].</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>2.3. Procedimiento general</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> La celda IR de medida se llena, de forma manual y secuencial, con el blanco  (cloroformo), los est&#225;ndares y las soluciones-muestra bajo evaluaci&#243;n;  y se adquiere en cada caso el espectro FTIR correspondiente, acumulando  3 barridos (<I>scans</I>) a una resoluci&#243;n de 4 cm</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">. Se procesan los espectros  y la data espectral de inter&#233;s. La concentraci&#243;n de atenolol se obtiene  por interpolaci&#243;n de la se&#241;al anal&#237;tica correspondiente a las soluciones-muestra  en la curva de calibraci&#243;n simple construida a partir de los est&#225;ndares,  analizados y procesados en forma an&#225;loga a las muestras (ver secci&#243;n 3.4.1).  El estado de las ventanas de ZnSe, as&#237; como el paso &#243;ptico, se verific&#243;  peri&#243;dicamente mediante el m&#233;todo tradicional, estableciendo el <I>background</I>  &#150;y adquiriendo el espectro&#150; con la celda vac&#237;a [22, 23]. La variaci&#243;n en  el paso &#243;ptico en una sesi&#243;n de trabajo t&#237;pica, es inferior al 1% (b</FONT></FONT><SUB><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana">(n</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana">=3)</FONT></SUB><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">  = 0,500 &#177; 0,004 mm).</FONT></P> </MULTICOL> <MULTICOL GUTTER="39" COLS="2">     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>2.3.1. M&#233;todo de referencia</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Los an&#225;lisis de referencia se hicieron por HPLC con detecci&#243;n UV, utilizando  el m&#233;todo descrito en la USP [4]. Se inyectan, de forma autom&#225;tica, 10  &#181;L de la muestra en el cromat&#243;grafo, equipado con una columna <I>Nova Pack  C18</I> (3,9 &#215; 300 mm) de 4,6 &#181;m d.p. (Waters) con un flujo de fase m&#243;vil de  0,6 mL min</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">. La se&#241;al de absorbancia se monitorea de forma continua a  226 nm.</FONT></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>3. Resultados y Discusi&#243;n</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>3.1. Estudios preliminares. Caracterizaci&#243;n espectral del sistema</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> El cloroformo es un disolvente apropiado para llevar a cabo la determinaci&#243;n  propuesta. Por su parte,  el comportamiento espectral del atenolol en cloroformo evidencia algunos  cambios que son producto de las interacciones con el disolvente y, evidentemente,  de las zonas opacas &#150;o de pobre transparencia&#150; del mismo. El cloroformo  presenta bandas de absorci&#243;n caracter&#237;sticas a 3018; 1214; 740 y 667 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">  (<a href="#fig2a">Figura 2.1A</a>), que afectan, obviamente, las ventanas de transparencia del  solvente en esas zonas; y por ende, al blanco en el sistema utilizado en  esas zonas espec&#237;ficas.</FONT></FONT></P>     <P ALIGN="center"><a name="fig2b"> <img border="0" src="/img/fbpe/rtfiuz/v32n3/art07fig2b.gif" width="579" height="449"></a></P>     
<P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> De esta forma, se observan ligeros desplazamientos en pr&#225;cticamente todas  las bandas,del analito, siendo este comportamiento notorio en las bandas  de 3514 y 3401 cm</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">, as&#237; como en la de 1645 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1 </SUP></FONT> <FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">que aparece centrada en  1671 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">. No obstante, la mayor&#237;a de las bandas se ven muy poco afectadas  en este medio, probablemente por las caracter&#237;sticas del disolvente, condici&#243;n  que las hace potencialmente &#250;tiles desde el punto de vista anal&#237;tico.</FONT></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>3.2. Estudios de viabilidad de la propuesta metodol&#243;gica</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Para establecer la viabilidad de la propuesta de trabajo, se estudi&#243; el  comportamiento espectral de dos est&#225;ndares, una soluci&#243;n-muestra y la misma  muestra fortalecida con el principio activo, en el intervalo espectral  comprendido entre 1100 y 1600 cm</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">. Como se puede observar en la  <a href="#fig3">Figura  3</a>, en esta regi&#243;n se localizan varias bandas de inter&#233;s. Sin embargo, algunas  est&#225;n cerca de zonas de absorci&#243;n del disolvente (1177 y 1245 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">), otras  no est&#225;n bien resueltas, en algunas se observan fuertes desplazamientos  (1671 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">) y otras son muy d&#233;biles; condiciones todas conducentes a potenciales  errores anal&#237;ticos.</FONT></FONT></P>     <P ALIGN="center"><a name="fig3"> <img border="0" src="/img/fbpe/rtfiuz/v32n3/art07fig3.gif" width="577" height="1300"></a></P>     
<P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Para el desarrollo de este trabajo se seleccion&#243; la banda centrada en 1512  cm</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2"> como banda principal y la de 1300 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2"> &#150;de menor intensidad&#150; como  banda secundaria; ya que ambas lucen libres de interferencias. En todas  las disoluciones usadas para este estudio, se observa &#150;en ambas bandas&#150;  un crecimiento centrado, sin ning&#250;n tipo de deformaci&#243;n o desplazamiento  (</FONT></FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Symbol">l</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUB>Max</SUB></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">, forma de la banda, etc.) que es consistente con la concentraci&#243;n  del analito (<a href="#fig3">Figuras 3B y 3C</a>). Este comportamiento avala la viabilidad  del m&#233;todo propuesto.</FONT></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> La se&#241;al anal&#237;tica seleccionada, en ambos casos, fue la absorbancia en  el m&#225;ximo de absorci&#243;n (</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Symbol" SIZE="2">l</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUB>Max</SUB></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">: 1512 y 1300 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">, respectivamente) con correcci&#243;n  mediante una l&#237;nea de base apropiada: 1525-1494 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2"> y 1313-1282 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">,  respectivamente. Optimizaci&#243;n de par&#225;metros&nbsp;experimentales</B></FONT></P>     ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> El m&#233;todo de an&#225;lisis propuesto requiere la optimizaci&#243;n del proceso de  extracci&#243;n del analito en la muestra, as&#237; como la selecci&#243;n de las condiciones  instrumentales-espectrosc&#243;picas de operaci&#243;n del espectr&#243;metro FTIR.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>3.3.1. Par&#225;metros espectrosc&#243;picos-instrumentales</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Se estudi&#243; la influencia que tienen sobre la se&#241;al anal&#237;tica algunos par&#225;metros  inherentes a la espectrometr&#237;a FTIR como son: la resoluci&#243;n y la acumulaci&#243;n  de barridos (<I>scans</I>). De igual forma, se evaluaron las condiciones para  establecer la se&#241;al de fondo, el <I>background</I>. Los estudios se realizaron  usando dos referencias: un est&#225;ndar de atenolol en cloroformo 0,4% (p/v)  y una soluci&#243;n-muestra de concentraci&#243;n equivalente.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> La experiencia obtenida en investigaciones previas [21, 24] indica que  el n&#250;mero de interferogramas acumulados (n) es un par&#225;metro que tiene poca  influencia sobre la relaci&#243;n se&#241;al/ruido, cuando se utiliza cloroformo  como disolvente. En este trabajo se evidenci&#243; el mismo comportamiento.  Sobre esta base, se fij&#243; el valor de n en 3, tanto para establecer el <I>background  </I>como para la adquisici&#243;n de los espectros correspondientes al blanco, a  est&#225;ndares y muestras. En estas condiciones se obtiene un blanco de calidad,  sin mayores costos en el tiempo de an&#225;lisis.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Con relaci&#243;n a la resoluci&#243;n nominal (R</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUB>N</SUB></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">), se estudi&#243; el efecto de este  par&#225;metro sobre la se&#241;al de las soluciones de referencia, en el intervalo  de 2 a 32 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">. Como se puede observar en la  <a href="#fig4">Figura 4</a>, la se&#241;al anal&#237;tica  (Abs en </FONT> </FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Symbol"> l</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUB>max</SUB></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">, corregida por l&#237;nea de base) se mantiene pr&#225;cticamente constante  en el intervalo de 2 a 4 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">; y a partir de ese valor disminuye de forma  considerable.</FONT></FONT></P>     <P ALIGN="center"><a name="fig4"> <img border="0" src="/img/fbpe/rtfiuz/v32n3/art07fig4.gif" width="518" height="535"></a></P>     
<P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Por otra parte, el tiempo de adquisici&#243;n de los espectros decrece al aumentar  el valor R</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUB>N</SUB></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">. Sobre la base de estos resultados, se seleccion&#243; un valor  de 4 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2"> para el resto del trabajo.</FONT></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>3.3.2. El proceso de extracci&#243;n. Preparaci&#243;n de la soluci&#243;n-muestra</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Las caracter&#237;sticas qu&#237;micas y f&#237;sicas de las tabletas pueden variar de  un producto a otro; ya que los excipientes y su concentraci&#243;n; as&#237; como  sus propiedades f&#237;sicas pueden diferir considerablemente en los distintos  productos comerciales.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> La primera etapa del m&#233;todo propuesto involucra la extracci&#243;n (s&#243;lido-liquido)  cuantitativa del analito presente en los comprimidos, usando CHCl</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUB>3</SUB></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2"> como  medio extractante. Para facilitar el lixiviado del principio activo, esta  etapa se realiz&#243; en un ba&#241;o ultras&#243;nico; y se vari&#243; el tiempo de tratamiento  con ultrasonido (t</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUB>Son</SUB></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">) entre 0 y 60 min. El estudio se realiz&#243; para todas  las muestras evaluadas. Sin embargo, por razones de simplicidad, en la  <a href="#fig5">Figura 5</a> solo se muestra el comportamiento observado para dos muestras.</FONT></FONT></P>     ]]></body>

<P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> En ausencia de ultrasonido (t</FONT><SUB><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana">Son</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> </FONT> </SUB><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"> <FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">= 0), se observa una se&#241;al anal&#237;tica significativa.  Esto indica que una fracci&#243;n importante del principio activo &#150;presente  en el s&#243;lido&#150; pasa con facilidad al bulto de la soluci&#243;n extractante. A  partir de ese punto, se observa un incremento en la se&#241;al con t</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUB>Son</SUB></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">, hasta  los 10 minutos. Para valores superiores, la se&#241;al anal&#237;tica se hace independiente  de este par&#225;metro; lo que indica, en principio, una extracci&#243;n cuantitativa.  El uso de mayores tiempos no aporta ning&#250;n beneficio; y se traduce en un  incremento en el tiempo de an&#225;lisis.</FONT></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Para corroborar la eficiencia del proceso de extracci&#243;n, se compar&#243; el  espectro FTIR obtenido a partir de un est&#225;ndar de atenolol 0,4% (p/v),  preparado directamente en cloroformo, con el correspondiente a una soluci&#243;n-muestra  de concentraci&#243;n equivalente, preparada y analizada mediante el procedimiento  descrito. No se observan diferencias significativas entre ambos espectros  (<a href="#fig5">Figura 5B</a>), lo cual corrobora que el proceso de extracci&#243;n fue cuantitativo.</FONT></P> </MULTICOL> <MULTICOL GUTTER="39" COLS="2">     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>3.4. Validaci&#243;n del m&#233;todo propuesto</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Para establecer la confiabilidad de la propuesta metodol&#243;gica, se procedi&#243;  a validar el m&#233;todo de an&#225;lisis. Para ello se realizaron experiencias dirigidas  a caracterizar anal&#237;ticamente el sistema, a estudiar posibles efectos interferentes  por parte de la matriz y a evaluar la exactitud del mismo.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>3.4.1. Caracter&#237;sticas anal&#237;ticas  del m&#233;todo propuesto</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Bajo las condiciones de trabajo seleccionadas, se construyeron curvas de  calibraci&#243;n simples (CCS) en las dos bandas seleccionadas. En ambos casos  se encontr&#243; excelente proporcionalidad entre la se&#241;al anal&#237;tica y la concentraci&#243;n  de atenolol en el intervalo de 0 a 1,0% (p/v), concentraci&#243;n l&#237;mite estudiada  (concentraci&#243;n del patr&#243;n). Las ecuaciones de las rectas obtenidas por  regresi&#243;n lineal (blanco y 7 est&#225;ndares), fueron: A</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUB>(1512 cm-1)</SUB></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2"> = 9.10 </FONT> <FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;5</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">  + 0,9032 [Atenolol] y A</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUB>(1300 cm-1)</SUB></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2"> = 1. 10 </FONT> <FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;4</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2"> + 0,1426 [Atenolol], con  coeficientes de correlaci&#243;n (r) de 0,9997 y 0,998, respectivamente. Los  l&#237;mites de detecci&#243;n (3</FONT></FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Symbol">s</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">) y cuantificaci&#243;n (10</FONT></FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Symbol">s</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">), fueron de 0,004 y 0,0125%  (p/v), respectivamente, para la banda de 1512 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">; y de 0,025 y 0,083%%  (p/v) para la banda de 1300 cm</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">. La frecuencia de an&#225;lisis, sin tomar  en cuenta la etapa de preparaci&#243;n de las muestras, es de aproximadamente  30 muestras h</FONT><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">.</FONT></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> La repetibilidad del m&#233;todo se evalu&#243; mediante 5 r&#233;plicas de soluciones-muestra  preparadas de forma independiente a partir de productos con 50 y 100 mg  del analito. La desviaci&#243;n est&#225;ndar relativa (DER) en la banda principal  fue de 0,6% y 0,4%, respectivamente. Por su parte, la precisi&#243;n intermedia  (entre d&#237;as) dio valores de 0,8% y 0,6%. El estudio equivalente, realizado  en la banda de 1300 cm</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2">, mostr&#243; valores similares.</FONT></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>3.4.2. Estabilidad de las soluciones  de trabajo</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> La estabilidad de las soluciones de referencia se evalu&#243; monitoreando su  se&#241;al anal&#237;tica por un periodo de tres horas. No se observaron variaciones  significativas en ninguna de las bandas, lo cual es indicativo de la estabilidad  de est&#225;ndares y muestras; as&#237; como de la robustez de m&#233;todo.</FONT></P>     ]]></body>

Efecto de matriz</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> La extracci&#243;n del analito de la muestra con un disolvente org&#225;nico permite  la separaci&#243;n parcial del mismo de la matriz de la muestra, en una etapa  previa a la detecci&#243;n espectrom&#233;trica. Por esta v&#237;a, se a&#237;sla al analito  de potenciales interferentes presentes en la muestra (excipientes), sobre  la base de la solubilidad diferencial que presentan el analito y los excipientes  en las muestras bajo estudio, en cloroformo. (Estas  muestras provienen del mismo Laboratorio Farmac&#233;utico; muestras 1.1 y 1.2  de la <a href="#Tabla_2">Tabla 2</a>).</FONT></P> <MULTICOL GUTTER="39" COLS="2"> </MULTICOL> <MULTICOL GUTTER="39" COLS="2">     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Las pendientes de las curvas CAE no mostraron diferencias significativas  con las correspondientes a las curvas CCS, en ninguna de las bandas. Estos  resultados indican que el m&#233;todo propuesto esta libre de interferencias  de tipo f&#237;sico y qu&#237;mico por parte de la matriz.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>3.4.4. Estudios de exactitud</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Se realiz&#243; una estimaci&#243;n de la exactitud del m&#233;todo, analizando las soluciones  usadas en las experiencias CAE bajo la &#243;ptica de estudios de recuperaci&#243;n.  Los resultados obtenidos evidencian, en todos los casos, recuperaci&#243;n cuantitativa  (97,5-102,5). Por otra parte, en las muestras analizadas por CAE (<a href="#Tabla_1">Tabla  1</a>), la concentraci&#243;n de atenolol obtenida por ambos m&#233;todos de calibraci&#243;n  (CCS y CAE) no mostr&#243; diferencias estad&#237;sticamente significativas.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Por &#250;ltimo, se analizaron algunas muestras por el m&#233;todo descrito en la  USP. La comparaci&#243;n estad&#237;stica (pruebas F-Fisher y t-student) de los resultados  obtenidos por ambos m&#233;todos, no mostr&#243; diferencias significativas [25].  Estos resultados corroboran la ausencia de interferencias de matriz y la  exactitud del m&#233;todo; as&#237; como su confiabilidad, viabilidad y aplicabilidad.</FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> <B>3.5. An&#225;lisis de muestras reales</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Tras los excelentes resultados obtenidos en los estudios de validaci&#243;n,  el m&#233;todo propuesto fue aplicado a la determinaci&#243;n de atenolol en las  formulaciones farmac&#233;uticas s&#243;lidas (tabletas) descritas en la secci&#243;n  2.2. El an&#225;lisis se realiz&#243; en las dos bandas seleccionadas, utilizando  la misma serie de soluciones-muestra y est&#225;ndares. Por ello, las CCS para  la bandas de 1512 y 1300 cm</FONT><FONT COLOR="#1f1a17" SIZE="2" FACE="Bookman"><FONT COLOR="#1f1a17" FACE="Verdana"><SUP>&#150;1</SUP></FONT><FONT COLOR="#1f1a17" FACE="Verdana" SIZE="2"> se construyeron en el mismo intervalo de  concentraci&#243;n: 0-1,0% (p/v). Como se puede observar en la  <a href="#Tabla_2">Tabla 2</a>, la concentraci&#243;n  de atenolol encontrada en las dos bandas es similar; y muestra en la mayor&#237;a  de los casos una buena concordancia con los valores declarados en la etiqueta  del producto. Conclusiones</B></FONT></P>     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> En este trabajo se desarroll&#243; una nueva propuesta metodol&#243;gica para la  determinaci&#243;n de atenolol en formulaciones farmac&#233;uticas s&#243;lidas. La estrategia  de trabajo utilizada, preparaci&#243;n de est&#225;ndares y soluciones-muestra en  cloroformo y an&#225;lisis directo por espectrometr&#237;a FTIR, propici&#243; el desarrollo  de un m&#233;todo de an&#225;lisis simple, r&#225;pido y robusto. Adem&#225;s, de muy bajo  costo y accesible a muchos laboratorios. En relaci&#243;n al consumo de cloroformo,  este se limita a la preparaci&#243;n de est&#225;ndares y muestras; y comparado con  otros m&#233;todos que utilizan disolventes org&#225;nicos [4, 8-11], es razonablemente  bajo. El uso de la espectrometr&#237;a FTIR como t&#233;cnica de detecci&#243;n, proporciona  selectividad, rapidez y simplicidad al m&#233;todo. Las Figuras de m&#233;rito relacionadas  con sensibilidad, precisi&#243;n y exactitud; as&#237; como los resultados obtenidos  en los estudios de estabilidad de las soluciones y en el paso &#243;ptico de  la celda, confieren al m&#233;todo un perfil muy adecuado para ser utilizado  en el control de calidad de este tipo de productos.</FONT></P> </MULTICOL> <MULTICOL GUTTER="39" COLS="2">     <P ALIGN="justify"><FONT COLOR="#1f1a17" SIZE="2" FACE="Verdana"> Nuevos estudios se est&#225;n llevando a cabo con el fin de automatizar el m&#233;todo  de an&#225;lisis propuesto, incorporando en el dise&#241;o metodol&#243;gico el uso de  sistemas de flujo.
