Al finalizar el período experimental los conejos fueron anestesiados después de 12 horas de ayuno. Se extrajo la sangre y en el plasma se analizó el porcentaje de ácidos grasos de los TG y ECT, así como el contenido de TG y CT en las fracciones de lipoproteínas. En el grupo experimental se encontró un aumento no significativo de los TG y CT plasmático, así como en los TG y CT de la VLDL. La dieta con aceite crudo de palma contiene mayor relación de ácidos grasos S/P, produciendo diferencias en el contenido de C16:0 y C18:2,n-6 de los TG, sin cambio significativo en los ácidos grasos de los ECT, reflejándose en la concentración de los TG de la VLDL que aumentó de 19,7 a 42,4 mg/dL no significativamente, lo cual se debería considerar en el control dietético de pacientes con hiperlipidemia tipo IV, caracterizada por una elevación en los TG de la VLDL.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[The Scientific literature does not show a clear effect of a diet rich in palm oil on the plasma content of lipoproteins, triglycerides and cholesterol, some of the reports are in disagreement. The aim of this work was to study the effect of palm oil on triglycerides and cholesterol content on plasma in lipoproteins and fatty acid composition in plasma lipids. The experiment was carreid out in New Zealand male rabbits, fed with a diet 10% (wt/wt) fat content during 9 weeks. The fats used were corn oil (control group) or palm iol (experimental group). The animals were anaesthetized after 12 hour of starvation, fatty acid composition on triglycerides, cholesterol plasmatic and lipoproteins levels were determined. The animals fed with palm oil showed a not significant increase in very low density lipoprotein (VLDL) and plasma triglycerides and cholesterol. Palm oil diet showed high relation of satured/poliinsatured fatty acids and increased the contend of palmitic acid in plasma triglycerides with reduction of linoleic acids in this lipids fraction; However, the cholesteryl ester fatty acids did not change between corn oil and palm oil diet. The non significant increase in plasma triglycerides and VLDL content of the palm oil group must be taken into account in the dietetic control of type IV hiperlipidemic patient.]]></p></abstract>

<body><![CDATA[<P ALIGN="JUSTIFY">La aterog&eacute;nesis es una enfermedad de origen multifactorial y las lesiones a&oacute;rticas y coronarias comiezan en las primeras decadas de la vida<SUP>(1,2)</SUP>. Existen poblaciones con cierta predisposici&oacute;n gen&eacute;tica a poseer elevada la lipoprote&iacute;na de alta densidad (HDL)<SUP>(3)</SUP>. Goldstein y col.<SUP>(4)</SUP> observaron que el tipo de &aacute;cido graso de la dieta juega un papel importante en el catabolismo de la lipoprote&iacute;na de baja densidad (LDL), ya que, el n&uacute;mero de receptores de la LDL en el hepatocito es inverso a la concentraci&oacute;n de &aacute;cidos grasos saturados. Jeremiach<SUP>(5)</SUP> report&oacute; correlaci&oacute;n positiva entre ingesta de &aacute;cidos grasos saturados, niveles altos de colesterol en el suero y mayor incidencia de infartos al miocardio; pero no todos los &aacute;cidos grasos saturados tienen el mismo efecto hipercolesterol&eacute;mico, el &aacute;cido mir&iacute;stico, C14:0, es cuatro veces m&aacute;s aterog&eacute;nico que el l&aacute;urico, C12:0, y el palm&iacute;tico, C16:0,<SUP>(6,7)</SUP>. Kritchevsky y col.<SUP>(8)</SUP> encontraron que conejos alimentados con una dieta libre de colesterol, rica en grasas saturadas y celulosa, como fibra diet&eacute;tica, presentaron severos ateromas que disminuyeron al cambiar la celulosa por alfafa. Rollandelli y col.<SUP>(9)</SUP> han demostrado que las pectinas por fermentaci&oacute;n producen &aacute;cidos grasos saturados de cadena corta que promueven la lipog&eacute;nesis hep&aacute;tica.</P>     <P ALIGN="JUSTIFY">El aceite crudo de palma se obtiene del mesocarpio del fruto de la palma <I>Elaeis guineensis</I> originario de Guinea Occidental y actualmente el mayor productor y exportador de aceite de palma es Malasia. Este aceite se caracteriza por ser rico en &aacute;cido palm&iacute;tico y oleico, con menor proporci&oacute;n de &aacute;cido la&uacute;rico, mir&iacute;stico y linoleico. Para el consumo humano se utiliza el aceite de palma y la ole&iacute;na de la palma, una subfracci&oacute;n, incluido en la actualidad en las mezclas de aceite comestibles en Venezuela. En cuanto a sus efectos aterog&eacute;nicos existen controversias, Vles<SUP>(10)</SUP> demostr&oacute; que conejos alimentados con aceite de palma presentaban un alto grado de aterosclerosis. Denke y col.<SUP>(11)</SUP> encontraron que el &aacute;cido palm&iacute;tico es m&aacute;s hipercolesterol&eacute;mico que el &aacute;cido la&uacute;rico. Alfonso P&eacute;rez C.<SUP>(12)</SUP> observ&oacute; que conejos alimentados con aceite crudo de palma presentaron mayor agregaci&oacute;n plaquetaria respecto a los que recibieron aceite de ma&iacute;z, por el contrario, Honstra y col.<SUP>(13,14)</SUP> se&ntilde;alan que la ingesta de aceite de palma disminuy&oacute; la relaci&oacute;n de tromboxano A2 y prostaciclina I2 en conejos y en personas normolipid&eacute;micas el consumo del aceite de palma disminuy&oacute; la Lp(a). Ng TK y col.<SUP>(15)</SUP> encontraron que el consumo de la fracci&oacute;n de ole&iacute;na de palma disminuy&oacute; en 8% la relaci&oacute;n de LDL/ HDL en humanos. Baudet y col.<SUP>(16)</SUP> demostraron que el aceite de girasol respecto al aceite de palma es m&aacute;s efectivo en disminuir el colesterol total del plasma. El objetivo de este trabajo es comparar el efecto de la ingesta del aceite crudo de palma con respecto al aceite de ma&iacute;z sobre la concentraci&oacute;n de triacilglic&eacute;ridos (TG) y colesterol (CT) del plasma y las lipoprote&iacute;nas; as&iacute; como, determinar la composici&oacute;n de los &aacute;cidos grasos de los TG y &eacute;steres del colesterol (ECT) del plasma en conejos, animales susceptibles a sufrir hiperlipemias.</P>     <P ALIGN="JUSTIFY"></P>     <P ALIGN="JUSTIFY">&nbsp;</P> <B>    <P ALIGN="JUSTIFY">MATERIALES Y M&Eacute;TODOS</P> </B>    <P ALIGN="JUSTIFY"></P>     <P ALIGN="JUSTIFY">Se utilizaron conejos de la raza Nueva Zelandia con 2 Kg de peso y colocados en jaulas individuales se les suministr&oacute; 200 g de conejarina Purina<SUP>&reg;</SUP>/d&iacute;a, enriquecida con 10% de aceite de ma&iacute;z purificado (dieta control) o aceite crudo de palma (dieta experimental) por un per&iacute;odo de 9 semanas y agua <I>ad libitum</I>. Los animales con ayuno de 12 h. fueron anestesiados con 25 mg de tiopental s&oacute;dico por Kg de peso y la sangre se extrajo de la aorta abdominal. El plasma se obtuvo por centrifugaci&oacute;n a 1500 G durante 8 min con EDTA como anticuagulante y se determinaron los niveles de CT, TG y el porcentaje de &aacute;cidos grasos del plasma. Para el an&aacute;lisis de los &aacute;cidos grasos se extrajeron los l&iacute;pidos por el m&eacute;todo de Folch<SUP>(17)</SUP>. El extracto clorof&oacute;rmico se evapor&oacute; bajo corriente de nitr&oacute;geno, las fracciones lip&iacute;dicas se separaron en una l&aacute;mina de silica gel G 60 de 0,1 mm de espesor y como fase m&oacute;vil: &eacute;ter de petr&oacute;leo, &aacute;cido ac&eacute;tico y &eacute;ter et&iacute;lico (90:1:10:v/v/v), se separaron los TG y ECT para su transesterificaci&oacute;n seg&uacute;n el m&eacute;todo de Kates<SUP>(18)</SUP> y el porcentaje de &aacute;cidos grasos se determin&oacute; por cromatograf&iacute;a gas-l&iacute;quido con una columna de vidrio de 4 mm de di&aacute;metro y 1,83 metros de largo ampacadas con dietilen glicol-succinato al 4% sobre Chromosorb<SUP>&reg;</SUP>, la temperatura de la columna fue 185°C, en cada ensayo se us&oacute; patrones de &aacute;cidos grasos Sigma<SUP>&reg;</SUP>. En estas condiciones no se separan los &aacute;cidos grasos C18:3 del C20:0, ni el C20:4 del C22:0. Todos los solventes conten&iacute;an como antioxidante el butilhidroxitolueno, 1 mg/10 mL. El extracto clorof&oacute;rmico tambi&eacute;n fue utilizado para cuantificar el CT y TG del plasma por los m&eacute;todos de Abell, L.L y col.<SUP>(19)</SUP> y Zilversmit, I.B. y col.<SUP>(20)</SUP> respectivamente; las lipoprote&iacute;nas del plasma se aislaron por ultracentrifugaci&oacute;n seg&uacute;n el m&eacute;todo de Sunderman, F. W. y col.<SUP>(21)</SUP>. Para el an&aacute;lisis estad&iacute;stico se utiliz&oacute; la t de Student.</P>     <P ALIGN="JUSTIFY"></P>     <P ALIGN="JUSTIFY">&nbsp;</P> <B>    <P ALIGN="JUSTIFY">RESULTADOS</P> </B>    ]]></body>
<body><![CDATA[<P ALIGN="JUSTIFY"></P>     <P ALIGN="JUSTIFY">La dieta enriquecida con aceite de palma contiene mayor porcentaje de C16:0 (47,5%) y menor cantidad (8,9%) de C18:2,n-6 (&aacute;cido linoleico) respecto a la dieta con aceite de ma&iacute;z, <B><I>Tabla 1</B></I>. En dieta con aceite de palma la relaci&oacute;n de &aacute;cidos grasos saturados/poliinsaturados (S/P) es mayor y la concentraci&oacute;n de TG en la lipoprote&iacute;na VLDL del plasma aument&oacute; no significativamente de 19,7 mg/dL a 42,4 mg/dL, <B><I><a href="#tab2">Tabla</a> 2</I></B>. <body><![CDATA[<P ALIGN="JUSTIFY">&nbsp;</P>     <P ALIGN="JUSTIFY">&nbsp;</P> <B>    <P ALIGN="JUSTIFY">DISCUSI&Oacute;N</P> </B>    <P ALIGN="JUSTIFY"></P>     <P ALIGN="JUSTIFY">El aumento no significativo de los TG en la VLDL con el aceite m&aacute;s saturado, tambi&eacute;n fue reportado por Kohout y col.<SUP>(22)</SUP>, ya que, la s&iacute;ntesis de la lipoprote&iacute;na VLDL est&aacute; inversamente relacionada con el grado de insaturaci&oacute;n de los &aacute;cidos grasos. Las dietas con igual concentraci&oacute;n de l&iacute;pidos y diferentes relaciones S/P, no modificaron la concentraci&oacute;n de CT del plasma, aunque el &aacute;cido graso C16:0, abundante en el aceite de palma, no es un buen sustrato para esterificar el CT hep&aacute;tico e incrementar&iacute;a la concentraci&oacute;n del colesterol libre, que es una se&ntilde;al para disminuir la actividad de receptores hep&aacute;tico para la LDL<SUP>(23)</SUP>, adem&aacute;s, este efecto regulador de los &aacute;cidos grasos sobre los receptores de la LDL se pone en evidencia cuando se suministra colesterol en la dieta<SUP>(24)</SUP>, los resultados del presente trabajo fueron realizados con dietas bajas en colesterol. Baudet y col.<SUP>(16)</SUP> demostraron que el aceite de girasol, que contiene alto contenido de &aacute;cidos grasos poliinsaturados, fue m&aacute;s efectivo en disminuir el colesterol total del plasma y aumentar el colesterol de la HDL en mujeres hipercolesterol&eacute;micas. En humanos, el consumo de aceite de palma disminuy&oacute; el colesterol del plasma y de la LDL respecto al consumo de aceite de coco posiblemente por las diferencias en el contenido y tipo de &aacute;cidos grasos saturados<SUP>(25)</SUP>.</P>     <P ALIGN="JUSTIFY">Cuando se eval&uacute;a el efecto lip&eacute;mico de diferentes aceites comestibles se debe considerar el periodo de ingesta, Torrealba de Ron A y col.<SUP>(26)</SUP> encontraron que a los dos meses de dieta, el aceite de palma respecto al aceite de ma&iacute;z no produce cambios en el contenido de CT plasm&aacute;tico cuando la dieta se enriquece con 0,1% de colesterol, no obstante, a los 6 meses el CT del plasma aument&oacute; en ambos grupos. Otro factor que puede influir en los resultados es la fracci&oacute;n no saponificable, en el aceite crudo de palma corresponde principalmente a los antioxidantes tocoferoles y tocotrienoles, cuya concentraci&oacute;n disminuye cuando se purifican los aceites, sin olvidar los factores gen&eacute;ticos y estilo de vida que pueden influir en el metabolismo lip&iacute;dico, controlados en nuestro dise&ntilde;o experimental y que pueden variar en las poblaciones humanas.</P>     <P ALIGN="JUSTIFY">Las diferencias en el contenido de &aacute;cidos grasos de las dietas por el uso de diferentes aceites vegetales, no alter&oacute; el porcentaje del C18:2,n-6 en los ECT del plasma, al parecer las enzimas que esterifican el colesterol, como la lecitin colesterol acil transferasa (LCAT) poseen diferencia de afinidad con la estructura del acil-CoA, de tal forma que, aunque exista menor concentraci&oacute;n de C18:2,n-6, &eacute;ste es captado selectivamente, la enzima intracelular acilcolesterol aciltransferasa (ACAT) tambi&eacute;n esterifica el colesterol y utiliza con preferencia el &aacute;cido C18:1,n-9 respecto a los &aacute;cidos grasos saturados<SUP>(23)</SUP>. El aumento no significativo de los TG del plasma y de la VLDL en el grupo con dieta suplementada con aceite de palma, <B><I>Tabla 2</B></I>, puede ser por una disminuci&oacute;n de la actividad de la lipasa plasm&aacute;tica, no obstante, en condici&oacute;n post-prandial los &aacute;cidos grasos saturados son absorbidos a menor velocidad que los &aacute;cidos grasos insaturados y producen menor respuesta lip&eacute;mica<SUP>(27)</SUP>. La relaci&oacute;n del tipo de &aacute;cidos grasos, su concentraci&oacute;n en los alimentos y sus efectos sobre la concentraci&oacute;n de los TG del plasma deber&iacute;a ser considerada en el tratamiento diet&eacute;tico de la hiperlipemia tipo IV caracterizada por la elevaci&oacute;n de los TG en la VLDL.</P>     <P ALIGN="JUSTIFY"></P>     <P ALIGN="JUSTIFY">&nbsp;</P>     <P ALIGN="JUSTIFY">&nbsp;</P>     ]]></body>
