La tarjetas FTA® fueron impregnadas con diluciones seriadas de fluido alantoideo con un título inicial de 10(8,8) DL/50 del VEN cepa LaSota y analizadas mediante la prueba de reacción en cadena por la polimerasa transcriptasa reversa (por sus siglas en ingles RT-PCR) a las 24 horas, 7 y 14 días. La concentración más baja del virus detectada en el fluido alantoideo fue 10(5,8) DL/50. La detección del virus a partir de la tarjeta fue posible hasta 14 días después de su inactivación. El re-aislamiento viral en huevos embrionados a partir de las tarjetas resultó negativo. La inactivación del virus en las tarjetas no afectó la calidad de la secuencia de nucleótidos, permitiendo la determinación de su virulencia mediante la secuenciación de los nucleótidos que codifican la zona de corte de la proteína de fusión resultando ser lentogénico en concordancia con la cepa inicial de virus aplicada a las tarjetas. Se concluye que las tarjetas FTA® representan una alternativa válida para el muestreo, inactivación y diagnóstico molecular del VEN, con un alto grado de bioseguridad.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[The feasibility of using FTA® cards (Flinders Technology Associates) for inactivation and transportation of allantoic fluid infected with Newcastle disease virus (NDV) was evaluated. Serial dilutions of allantoic fluid with a titer of 10(8.8) ELD/50 of LaSota strain NDV were loaded on the FTA® cards and analyzed after 24 hour, 7 and 14 days using reverse transcriptase-polimerase chain reaction (RT-PCR). The lowest virus concentration that could be detected from the FTA® cards was 10(5.8) ELD/50. The detection of the inactivated virus was possible after 14 days of virus inactivation. No virus re-isolation in embryonating eggs was possible from the cards. No negative effects of the FTA® card inactivation on the nucleotide sequences were observed, allowing the determination of its virulence by direct nucleotide sequencing of the F protein cleavage site, resulting in a lentogénic strain in concordance with the initial virus applied on the cards. It can be conclude that FTA® cards are a valid alternative for NDV sampling, inactivation and molecular diagnostic with a high degree of biosecurity.]]></p></abstract>

La presencia  en Latinoamérica de cepas velogénicas del virus de la enfermedad de Newcastle  (VEN) con un patrón epidemiológico de endemia, es una de las preocupaciones  máximas de la industria avícola [17]. La implementación de técnicas de  diagnóstico molecular como herramientas en la identificación y vigilancia  epidemiológica del agente causal de la enfermedad, se constituye en un  área activa de investigación. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" size="2" face="Verdana"> El VEN es un virus ARN encapsulado, de cadena simple y de sentido negativo  que pertenece a la familia <I>Paramyxoviridae</I>, subfamilia <I>Paramyxovirinae,  </I>que recientemente fue ubicado<I> </I>en el nuevo género <I>Avulavirus</I>, estableciéndose  así una separación definitiva entre los paramixovirus aviares y los <I>Rubulavirus</I>  de mamíferos [2, 14]. El virus posee un genoma no segmentado que codifica  para 6 proteínas estructurales: hemoaglutinina-neuraminidasa (HN), proteína  de fusión (F), nucleocápside (NP), matriz (M), polimerasa (L) y fosfoproteína  (P) [1, 14]. La proteína de fusión es sintetizada como un precursor inactivo  (Fo), el cual debe ser separado enzimáticamente para que se active y se  generen las porciones F<SUB>1</SUB> y F<SUB>2</SUB>, lo que permite la dispersión del virus de  célula a célula. Los virus velogénicos presentan fenilalanina en la posición  117 y una mayor cantidad de aminoácidos básicos en el sitio de corte de  la proteína de fusión, lo cual es reconocido específicamente por la proteasa  furina, que se encuentra distribuida en un amplio rango de tejidos en el  ave, haciendo más eficiente la colonización de los virus velogénicos [1,  10, 13]. De allí la importancia de identificar y caracterizar esta porción  del genoma viral. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" size="2" face="Verdana"> La manipulación y transporte de VEN de alta patogenicidad, representa un  riesgo potencial para la diseminación de la enfermedad, por lo que se deben  tomar todas las medidas de bioseguridad que permitan el traslado seguro  de muestras entre laboratorios. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" size="2" face="Verdana"> Durante los últimos años se ha ensayado la utilización de tarjetas de papel  de filtro Flinders Technology Associates (tarjetas FTA<SUP>®</SUP>) producidas y comercializadas  por Whatman<SUP>®</SUP> Internacional Ltd. Reino Unido., para la inactivación y traslado  de microorganismos [9, 11, 15], incluso algunos patógenos aviares tales  como Mycoplasma y el virus de la bronquitis infecciosa [7, 8]. Las tarjetas  FTA<SUP>®</SUP>, están constituidas de una membrana de celulosa que contiene químicos  liofilizados capaces de inactivar un amplio rango de microorganismos preservando  sus ácidos nucleicos [3, 12, 15]. Las muestras obtenidas de las tarjetas  pueden ser procesadas en el laboratorio a partir del virus inactivado en  el papel de la tarjeta. El objetivo de este trabajo fue demostrar las ventajas  de utilizar tarjetas FTA<SUP>®</SUP> para la inactivación, transporte, diagnóstico  molecular del VEN en muestras de fluido alantoideo. </FONT></P>     ]]></body>
<body><![CDATA[<p ALIGN="justify"><B><FONT COLOR="000000" size="2" face="Verdana"> MATERIALES Y MÉTODOS </FONT></B>      <P ALIGN="justify"><B><FONT COLOR="000000" size="2" face="Verdana"> Virus </FONT></B> </P>      <P ALIGN="justify"><FONT COLOR="000000" size="2" face="Verdana"> Una vacuna liofilizada contra el VEN (tipo I, cepa LaSota) fue diluida  según las recomendaciones del fabricante (Merial Select, Gainesville, EUA)  y 0,1 mL inoculados en la cavidad alantoidea de huevos embrionados libres  de patógenos específicos (white leghorns) de nueve días de edad (Merial  Select, Gainesville, EUA). Pasadas 72 horas, el fluido alantoideo proveniente  de los embriones inoculados se tituló según la técnica descrita por Villegas  [18]. La concentración inicial del virus en el fluido alantoideo utilizado  en el ensayo fue de 1&nbsp;× 10<SUP>8,8</SUP> DL<SUB>50</SUB>/mL. De este fluido alantoideo, se realizaron  nueve diluciones seriadas (1 en 10) utilizando agua estéril y libre de  ARNasas (Quiagen Sciences. Maryland, EUA). Alícuotas de 50 µL de cada una  de las diluciones se aplicaron a las tarjetas y otras fueron almacenadas  a -20°C hasta su procesamiento. </FONT></P>      <P ALIGN="justify"><font face="Verdana" size="2"><B><FONT COLOR="000000"> Detección del VEN a partir de las tarjetas FTA</FONT></B><SUP><FONT COLOR="000000"><B>®</B></FONT></SUP></font></P>      <P ALIGN="justify"><FONT COLOR="000000" size="2" face="Verdana"> Con la finalidad de determinar el título del VEN más bajo que puede ser  detectado en fluido alantoideo a partir de las tarjetas FTA<SUP>®</SUP>, se realizó  la prueba de transcriptasa reversa-reacción en cadena por la polimerasa  (por sus siglas en inglés RT-PCR) para cada una de las diluciones de fluido  alantoideo muestreadas. El procedimiento se repitió los días 7 y 14, para  evaluar la capacidad de las tarjetas de preservar el ARN viral en la matríz  de la mismas luego de inactivado y permitir la detección del virus luego  de un tiempo de almacenaje oportuno para el traslado de las muestras hacia  los laboratorios. </FONT></P>      <P ALIGN="justify"><font face="Verdana" size="2"><B><FONT COLOR="000000"> Extracción y amplificación del ácido ribonucle</FONT></B><FONT COLOR="000000"><B>ico del VEN </B></FONT></font></P>      <P ALIGN="justify"><FONT COLOR="000000" size="2" face="Verdana"> Se extrajo la matriz de la tarjeta impregnada con el virus, para ser resuspendida  en 200 µL del buffer de extracción (Whatman International, Ltd. Reino Unido)  y colocada en hielo por 15 minutos, mezclando vigorosamente cada 5 minutos.  La fase líquida obtenida, así como 50 µL del fluido alantoideo homólogo  al colocado en las tarjetas (fluido control positivo), fueron sometidos  a un proceso de extracción de ARN empleando Trizol (Life Technologies Inc.,  Grand Island, NY, EUA), siguiendo las recomendaciones del fabricante. El  precipitado final fue diluido en 50 µL de agua estéril y libre de ARNasas  (Qiagen Sciences. Maryland, EUA). </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" size="2" face="Verdana"> La detección del VEN tanto en las tarjetas como en el fluido alantoideo  control, se realizó mediante la amplificación de un segmento de 250 pares  de bases localizado en el gen de la proteína de fusión (zona de corte),  empleando para ello los iniciadores degenerados, 5’-CTGCCACTGCTACTTGIGATAATCC-3’y  5’-CCTTGGTGAITCTATCCGIAAGG-3’ siguiendo el protocolo de RT-PCR descrito  por Seal y col. [13]. Las muestras amplificadas fueron sometidas a electroforesis  en un gel de 1,5% de agarosa y fueron visualizadas en una cámara de luz  ultravioleta empleando bromuro de etidio a una concentración de 0,5 µg/mL  [4, 7, 8, 13]. </FONT></P>      <P ALIGN="justify"><B><FONT COLOR="000000" size="2" face="Verdana"> Secuenciación y análisis de los nucleótidos del segmento amplificado </FONT></B> </P>      <P ALIGN="justify"><FONT COLOR="000000" size="2" face="Verdana"> Los segmentos de ADN del tamaño esperado (250 pares de bases) fueron escindidos  del gel y purificados empleando el paquete comercial QIAquick PCR Purification  Kit (Qiagen Inc., Valencia, CA, EUA) siguiendo las instrucciones del fabricante.  Los productos purificados fueron secuenciados empleando el paquete comercial  de secuenciación BigDye Terminador v3,1 y el secuenciador ABI PRISM 310  Genetic Analizer (Applied Biosystems, Foster City, CA, EUA). Las secuencias  de nucleótidos y patrón de aminoácidos inferido del gen de la proteína  de fusión del VEN, fueron analizadas con la ayuda del programa DNAStar  (DNAStar, Inc., Madison, WI, EUA). </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><font face="Verdana" size="2"><B><FONT COLOR="000000"> Inactivación del VEN en las tarjetas </FONT></B><FONT COLOR="000000"> <B> FTA</B><SUP><B>®</B></SUP> </FONT></font></P>      <P ALIGN="justify"><FONT COLOR="000000" size="2" face="Verdana"> Para demostrar la inactivación del virus en la tarjeta FTA<SUP>®</SUP>, se intentó  el re-aislamiento viral a partir de la matriz de la tarjeta impregnada  con VEN. Brevemente, muestras de las tarjetas impregnadas con 50 µL de  fluido alantoideo con un título de 1 × 10<SUP>8,8</SUP> DL<SUB>50</SUB>/mL del VEN, fueron colocadas  en tubos estériles con 200 µL del buffer de extracción (Whatman International,  Ltd. Reino Unido) e incubadas a temperatura ambiente por una hora. Posteriormente,  0,2 mL del sobrenadante de las tarjetas y del fluido alantoideo positivo,  fueron inoculados en huevos embrionados. Se realizaron tres pasajes sucesivos  (cada 48 horas), el fluido alantoideo fue recolectado y analizado mediante  RT-PCR determinar la presencia del virus. </FONT></P>     <p ALIGN="justify"><font face="Verdana" size="2"><B><FONT COLOR="000000"> RESULTADOS </FONT></B><FONT COLOR="000000"> <B> Y DISCUSIÓN</B> </FONT></font>      <P ALIGN="justify"><FONT COLOR="000000" size="2" face="Verdana"> La combinación de tecnologías de punta, representadas por la utilización  de las tarjetas FTA<SUP>®</SUP> [7-, 11, 15] y la prueba de RT-PCR acompañada de secuenciación  de nucleótidos [4-6, 13], representan un aporte para la vigilancia epidemiológica  de las enfermedades que afectan la industria avícola ya que facilitan el  diagnóstico y determinación de la patogenicidad de los virus de interés  en la sanidad avícola. Los productos amplificados mediante RT-PCR para  cada una de las diluciones de fluido alantoideo analizadas desde la tarjeta  y del fluido alantoideo antes de su inactivación, se muestran en la <a href="#f1"> FIG. 1</a>. La sensibilidad de las tarjetas fue alta, se detectó el virus en el  fluido alantoideo con un titulo inicial de 1 × 10<SUP>8,8</SUP> DL<SUB>50</SUB>/mL, incluso después  de una dilución de 1/1000, siendo capaz de permitir la identificación del  VEN en fluido alantoideo con una concentración de 1 × 10<SUP>5,8</SUP> LD/<SUB>50</SUB>. Esta  sensibilidad es equivalente a la de pruebas clásicas de detección para  el VEN, como la prueba de hemoaglutinación rápida en placa, la cual permite  identificar la presencia del virus en fluido alantoideo con un titulo de  1 × 10<SUP>5</SUP> LD/<SUB>50</SUB> [18]. La prueba de RT-PCR realizada al fluido alantoideo  control, permitió detectar niveles de hasta 1 × 10<SUP>4,8</SUP> LD/<SUB>50</SUB>, lo que demuestra  una diferencia en la sensibilidad debido a la muestra utilizada. La diferencia  en sensibilidad entre las tarjetas y el fluido alantoideo previo a su inactivación,  probablemente se deba al proceso de inactivación química que ocurre en  la matriz de la tarjeta y ha sido reportado para otros microorganismos  [9, 11, 12, 15]. </FONT></P>      <P ALIGN="center"><font face="Verdana" size="2"><a name="f1"><img border="0" src="/img/fbpe/rc/v16n2/art04.1.jpg" width="458" height="478"></a></font></P>  <b>

La identificación  del virus fue posible en cada uno de los días analizados, proporcionando  un tiempo suficiente entre la toma de muestra y la realización de la prueba,  incluso si esta es enviada entre laboratorios de distintos países. La disminución  de un logaritmo en la capacidad de detección observada el día 14 (<a href="#f2">FIG. 2</a>), ha sido explicada en la literatura como la consecuencia de la desnaturalización  natural del ARN a lo largo del tiempo, debido a efectos medioambientales  [7]. </FONT></p>      <P ALIGN="justify"><FONT COLOR="000000" size="2" face="Verdana"> La secuenciación de los nucleótidos amplificados del gen de la proteína  de fusión del VEN inactivado en las tarjetas, permitió predecir la secuencia  de aminoácidos en la zona de corte de la proteína de fusión del virus (<a href="#f3">FIG. 3</a>), donde se ubican los marcadores moleculares que permiten determinar  su patogenicidad [10, 13]. En concordancia con la cepa del VEN aplicada  a la tarjetas (cepa LaSota), el análisis de la secuencia de aminoácidos  demostró la presencia de un virus lentogénico. El proceso de inactivación  del VEN en las tarjetas FTA<SUP>®</SUP>, no afectó la secuencia de nucleótidos ni  el patrón inferido de aminoácidos, respetando la integridad del ARN viral.  Estos resultados concuerdan con lo reportado sobre la calidad de ácidos  nucleicos que pueden recuperarse de la tarjetas FTA<SUP>®</SUP> luego de la inactivación  de los microorganismos [7, 9, 11, 15]. </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" size="2" face="Verdana"> Utilizando RT-PCR, no fue posible detectar partículas virales en el fluido  alantoideo de embriones inoculados con una solución de virus inactivado  en la tarjeta. Por su parte, las muestras provenientes de embriones inoculados  con el fluido alantoideo no inactivado resultaron positivas a la identificación  molecular (<a href="#f4">FIG. 4</a>).
