El objetivo de esta investigación fue identificar y cuantificar las aminas biógenas: triptamina, putrescina, cadaverina, tiramina, histamina, espermina y espermidina, en 3 tipos de quesos madurados de producción nacional (Manchego, Brie y Parmesano), así como identificar Streptococcus, Brevibacterium y bacterias ácido lácticas presentes en estos quesos. Se analizaron 2 marcas para cada tipo de queso, por medio de la técnica de Cromatografía Líquida de Alta Resolución (HPLC), y se logró detectar 7 aminas biógenas. En el queso Manchego las aminas biógenas encontradas en mayor y menor concentración, respectivamente, fueron espermidina (120,22 ppm) y tiramina (43,10 ppm); en el queso Brie, la putrescina presentó la mayor concentración (145,25 ppm) mientras que la cadaverina fue detectada en menor concentración (38,41 ppm); en el queso Parmesano, espermina (142,88 ppm) y espermidina (49,82 ppm) fueron las aminas detectadas en mayor y menor concentración, respectivamente. Los contenidos promedios (ppm) de aminas biógenas por marca de queso madurado fueron: Manchego (M1): 617,60; (M2): 549,51; Brie (M1): 630,80; (M2): 823,67; Parmesano (M1): 667,72 y (M2): 633,47. Ninguno de los valores obtenidos excedió los límites máximos establecidos por la FAO/WHO como tolerables. El efecto de la marca de queso sobre la concentración de aminas biógenas no fue determinante, mientras que el efecto del tipo de queso influyó significativamente en la concentración de estas aminas. Se detectó la presencia de Streptococcus, Brevibacterium y bacterias ácido lácticas en cantidades inferiores a las reportadas en otras investigaciones. Los  cultivos iniciadores hidrolizan las cadenas polipeptídicas liberando aminoácidos;  sustrato de las enzimas descarboxilasas microbianas para la producción  de aminas biógenas [19]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Durante este proceso existen condiciones ambientales y de almacenamiento  como son: temperatura, humedad, grado de acidez y concentración de sales,  que afectan la textura del queso y pueden favorecer el crecimiento de las  enterobacterias. Estos microorganismos podrían estar presentes por inadecuada  manipulación higiénica, y tienen la capacidad de producir aminas biógenas  [14, 18]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Por otra parte, diversos autores, han reportado que los cultivos iniciadores  utilizados en la elaboración de los quesos madurados, se asocian con la  producción de aminas biógenas tales como: histamina, tiramina, triptamina,  putrescina, cadaverina, espermina y espermidina. Los cultivos iniciadores  <I>Lactobacillus, Streptococcus, Leuconostoc </I>y <I>Pediococcus</I>, que se utilizan  como inóculos en los quesos madurados, han sido identificados como bacterias  que poseen enzimas decarboxilasas de los aminoácidos con lo cual son potencialmente  productoras de aminas biógenas [3, 5, 12, 13, 26, 28]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> En Venezuela, estudios realizados por Izquierdo y col. [8] sobre el contenido  de aminas biógenas en quesos madurados, detectaron que varias especies  del género <I>Lactobacillus</I> tienen la capacidad de producir histamina. El  objetivo de éste trabajo fue determinar la presencia de aminas biógenas  en quesos madurados tipo: Manchego, Brie y Parmesano producidos en el país,  así como identificar <I>Streptococcus,</I> <I>Brevibacterium</I> y bacterias ácido lácticas  en estos tipos de quesos. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>MATERIALES Y MÉTODOS</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Recolección de la muestra</B> </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Se analizaron 8 muestras de queso Manchego, Brie y Parmesano de dos marcas  de producción nacional (denominadas M1 y M2), para un total de 48 muestras.  Las muestras de queso eran pertenecientes a un mismo lote de elaboración  por tipo y marca, y tenían un vencimiento de dos meses a partir de la fecha  del muestreo. Se adquirieron 250 g de cada queso en panaderías del municipio  Maracaibo, estado Zulia, Venezuela. Las muestras fueron trasladadas al  laboratorio en una cava con temperatura promedio de 15°C para realizar  los siguientes análisis por duplicado: </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Determinación y cuantificación de aminas biógenas</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Se determinaron las aminas: tiramina, triptamina, putrescina, cadaverina,  histamina, espermina, y espermidina utilizando el método recomendado por  Eerola y col. [4]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Para la preparación de las muestras, se homogeneizaron 5 g de queso con  10 mL de ácido perclórico 0,4 M en un homogenizador Ultra-turrax<sup>®</sup> y se  centrifugaron por 10 minutos a 3000 rpm. Luego fueron filtrados al vacío,  usando papel Whatman N° 2, y trasvasados a balones aforados de 25 mL. La  operación se repitió tres veces y los sobrenadantes fueron combinados y  ajustados a 25 mL con ácido perclórico 0,4 M. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Para la formación de derivados fluorescentes se tomó 1 mL del filtrado,  que fue mezclado con 200 µL de hidróxido de sodio 2 N y luego tamponada  añadiendo 300 µL de bicarbonato de sodio saturado. Posteriormente se adicionaron  2 mL de solución de cloruro de Dansilo (10 mg de cloruro de Dansilo en  1 mL de acetona) y fue transferido a una incubadora a 40°C por 45 min.  El cloruro de Dansilo residual fue removido añadiendo 100 µL de amoniaco  puro, se dejó en reposo por 30 min y se ajustó a un volumen de 5 mL con  acetonitrilo. Posteriormente se centrifugó por 5 minutos a 2500 rpm y el  sobrenadante fue filtrado a través de un filtro millipore de 0,22 µm de  poro para su inyección en el cromatógrafo. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Se preparó un estándar de 100 ppm a partir de patrones de tiramina, triptamina,  putrescina, cadaverina, histamina, espermina, y espermidina de alta pureza  (Sigma Chem. Co. EUA). La formación de derivados fluorescentes de los estándares  se realizó de la misma manera que las muestras. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El cromatógrafo marca Shimadzu<sup>®</sup>, estuvo integrado por una válvula de inyección  Rheodyne<sup>®</sup>, una bomba Shimadzu<sup>®</sup> modelo LC-6 A (Japón), una columna de fase  reversa Merck® RP-18 Lichosper (12,5 cm, 4,5 mm Ø, 5 µm) (Alemania), un  detector de UV-Visible marca Shimadzu<sup>®</sup> modelo SPD-6 A (Japón), a una longitud  de onda de 254 nm. Para el registro de los resultados se utilizó el software  cromatográfico Class-Vp Shimadzu®. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Se utilizó un gradiente de elusión con una mezcla de acetato de amonio  0,1 M (solvente A) y acetonitrilo (solvente B) comenzando 80%: 20% respectivamente,  incrementando B a 80% en 25 minutos. La corrida se mantuvo isocráticamente  por 5 minutos. El flujo fue de 0,7 mL/min y el volumen de la inyección  fue de 20 µL. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Para determinar la concentración de las aminas, se comparó el tiempo de  retención de cada muestra con los estándares de referencia y la concentración  del analito se calculó por comparación de las áreas de los picos. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Análisis microbiológico</B> </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Recuento de </B><I><B>Streptococcus</B></I><B>:</B> Se le realizó al queso Manchego según la metodología  de la ICMSF [7]. Se pesaron 11 g de muestra y se homogeneizaron con 99  mL de citrato de sodio al 2%. A partir de esta dilución (10<SUP>1</SUP>) se prepararon  diluciones seriadas desde 10<SUP>2</SUP> hasta 10<SUP>4</SUP>, las cuales se sembraron por superficie  en placas de agar M17 HIMEDIA<sup>®</sup>, y se incubaron en una estufa Thelco® a  35°C por 24 horas. Una vez transcurrido el tiempo se procedió al recuento  de colonias típicas (lisas, brillantes y de un color blanco amarillento). </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Recuento de Bacterias Ácido Lácticas y </B><I><B>Brevibacterium</B></I><B>:</B> El recuento de bacterias  ácido lácticas se le realizó al queso Parmesano y el recuento de <I>Brevibacterium</I>  al queso Brie empleando la metodología de la ICMSF [7]. Se pesaron 11 g  de muestra y se homogeneizaron con 99 mL de citrato de sodio al 2%. A partir  de esta dilución (10<SUP>1</SUP>) se prepararon diluciones seriadas desde 10<SUP>2</SUP> hasta  10<SUP>5</SUP>, y se sembró en placas de agar MRS (De Man Rogosa y Sharpe) Merck<sup>®</sup>,  que fueron incubadas por 48 h a 37°C en condiciones de microaerofilia (5-10%  de CO<SUB>2</SUB>), mediante el empleo de una campana de Gas-Pack. El contenido promedio más alto de  las aminas detectadas corresponde a la putrescina en el queso Brie, con  145,25 ppm, concentración que no mostró diferencias significativas (P&lt;0,05)  para esa amina con los otros quesos analizados. La cadaverina fue la amina  detectada en menor concentración con 38,41 ppm en el queso Brie. <body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Con relación a las otras aminas detectadas en los quesos estudiados se  observó que el contenido de triptamina en el queso Manchego es significativamente  menor (P&lt;0,05) con respecto al Parmesano y el Brie. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El queso Manchego presentó la menor concentración de tiramina (43,10 ppm)  diferente significativamente (P&lt;0,05) a los contenidos de la misma amina  en los quesos Parmesano (94,05 ppm) y Brie (109,94 ppm), los cuales no  presentaron entre ellos diferencias significativas. Detectándose el contenido  más alto en el Brie. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> No se observaron diferencias significativas en cuanto a la concentración  de histamina entre los tres tipos de quesos estudiados. Presentando la  mayor concentración el queso Brie (127 ppm) y la menor para el Parmesano  (82,07 ppm). </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La espermina se encontró en mayor concentración en el queso Parmesano (142,88  ppm) y difiere significativamente (P&lt;0,05) del contenido en el queso Brie  (80,77 ppm) el cual presentó el menor contenido de esta amina. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El queso Manchego mostró la mayor concentración de espermidina (120,22  ppm) observándose diferencias significativas (P&lt;0,05) en comparación con  el queso Parmesano (49,82 ppm) que presentó el menor contenido de la amina. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Diversas investigaciones [6, 12, 26, 27] han identificado y cuantificado  las aminas biógenas: tiramina, triptamina, putrescina, cadaverina, histamina,  espermina y espermidina en quesos madurados, reportando resultados similares  a este estudio. El alto contenido y variabilidad de aminas biógenas en  quesos madurados podría ser atribuido a las condiciones de procesamiento,  donde el tiempo de maduración en conjunto con los microorganismos presentes  en la elaboración del queso, promueven la proteólisis, resultando en un  incremento progresivo de los aminoácidos precursores de las aminas biógenas  [13, 19]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> En la <a href="#tab2">TABLA II</a> se presentan las concentraciones de aminas biógenas, expresadas  en ppm, para los distintos tipos y marcas de quesos analizados. Al analizar los datos de cada tipo de queso no  se encontraron diferencias significativas entre las marcas. Los valores  observados son inferiores a los encontrados por otros autores los cuales  reportan valores entre 241,92 y 540 ppm [13]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La putrescina se encontró en concentraciones de 101,05 hasta 151,2 ppm  y no presentó diferencias significativas entre los tipos y marcas estudiadas.  Valsamaki y col. [27] reportaron concentraciones similares a las encontrados  en este trabajo (77,7-193 ppm) y sugieren que los contenidos elevados de  putrescina pueden ser debidos a la presencia de enterobacterias durante  la fabricación de los quesos, ya que estas presentan alta actividad en  la descarboxilación de aminoácidos. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Las concentraciones de cadaverina variaron entre 32,82 y 66,72 ppm. Se  encontraron diferencias significativas (P&lt;0,05) entre los tipos de queso  Parmesano (M2) y Brie (M1), y entre las marcas M1 y M2 del queso Manchego.  Las concentraciones reportadas por otros autores [13, 14, 27] varían entre  82,8 y 215,28 ppm, superiores a las encontradas en este trabajo. El queso  tipo Parmesano presentó la mayor concentración de esta amina, coincidiendo  con lo reportado por Aygun y col. [1], quienes afirman que la cadaverina  puede estar presente en los quesos duros en concentraciones mayores a las  encontradas en quesos semiduros y blandos. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La concentración de tiramina varió de 38,57 hasta 120,63 ppm. No se observaron  diferencias entre las marcas para los distintos tipos de queso. Se encontraron  diferencias significativas (P&lt;0,05) entre las Marcas M1 y M2 de los quesos  tipo Parmesano y Manchego. Stratton y col. [17] y Rice y col. [22], reportaron  la presencia de tiramina en queso Parmesano en concentraciones de hasta  290 ppm. Rice y col. [17] en quesos Brie detectó cantidades máximas 260  ppm, coincidiendo con lo encontrado en los quesos analizados. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La histamina se encontró en un rango de 67,2 a 153,05 ppm. Se observaron  diferencias entre los tipos de queso, siendo significativamente mayor (P&lt;0,05)  la concentración en el queso Brie (M2) a las encontradas en Parmesano (M1)  y Manchego (M2). Los valores encontrados son superiores a los reportados  por Valsamaki y col. [27] (47 a 84 ppm), mientras que son inferiores a  los de Aygun y col. [1] (122 a 1720 ppm). Sumner y col. [23] señalan que  el hallazgo de histamina en los quesos madurados podría deberse a la disponibilidad  de histidina libre presente debido a la proteólisis que ocurre durante  la maduración. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La histamina es la amina principalmente asociada a intoxicaciones por consumo  de alimentos, la FAO/WHO [25] recomienda que su contenido no sea mayor  a 100 ppm. Los valores encontrados en el queso tipo Brie superan esta cifra.  Stratton y Taylor [21, 24] reportaron casos de intoxicación en personas  que estaban tomando medicamentos (isoniazida) y consumieron quesos que  contenían un rango de histamina entre 30 y 100 ppm. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Los valores de espermina variaron entre 69,5 y 149,13 ppm y no se encontraron  diferencias significativas para los tipos y marcas de quesos estudiados.  Vale y col. [26] reportaron la presencia de esta amina en 46 muestras de  queso analizadas con un promedio de 5,85 ppm, la diferencia de estos valores  con lo obtenido podría ser atribuida a contaminación con enterobacterias  [27]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La espermidina alcanzó concentraciones de 27,07 hasta 169,5 ppm, encontrándose  diferencias significativas (P&lt;0,05) entre los tipos Manchego (M1 y M2),  y el tipo Brie (M1). Entre marcas se encontraron diferencias en el tipo  Brie (M1 y M2). Los valores encontrados en este estudio son superiores  a los reportados por Novella y col. [13], quienes detectaron concentraciones  entre 0 a 43,04 ppm. </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Analizando en conjunto los datos discutidos previamente se puede decir  que, el efecto de la marca de queso sobre la concentración de aminas biógenas  no es determinante, no así el efecto del tipo de queso, factor que influye  en la concentración de estas aminas significativamente, tal vez debido  a las diferencias en el uso de cultivos iniciadores y en el proceso de  manufactura de los mismos [16, 19]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La <a href="#tab3">TABLA III </a>muestra el contenido total de aminas biógenas para los tipos  y marcas de los quesos madurados estudiados (expresados en ppm). Los valores  detectados por tipo de queso variaron desde 583,56 ppm para tipo Manchego,  hasta 731,34 ppm para tipo Brie. Aygun y col. [1] reportaron que, los contenidos  de aminas biógenas para los quesos blandos pueden ser de 1700 ppm, semi  duros 705 ppm y los duros 2250 ppm, valores que son superiores a los obtenidos  en este estudio. Según FAO/WHO, el contenido total de aminas biógenas no  debe exceder 1000 ppm [25]. Estos autores evidenciaron que el contenido de histamina y tiramina encontrado  en el cultivo, disminuyó en un 70% y 55% respectivamente, agotando los  nutrientes disponibles en un tiempo de maduración de 4 semanas. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> En el queso parmesano se estudió la presencia de bacterias ácido lácticas,  cuyos valores se ubican entre 5,74 y 6,93 Log UFC/g. Joosten y Northolt  [9] reportaron recuentos similares de <I>Lactobacillus</I> (5,47 hasta 6,77 Log  UFC/g). Se ha demostrado que los <I>Lactobacillus </I>tienen la capacidad de descarboxilar  la histidina para producir histamina. Izquierdo y col. [8] analizaron la  relación entre los <I>Lactobacillus</I> y la histamina en quesos madurados, encontrando  histamina en queso parmesano en concentraciones de 95,23 ppm, que resultaron  positivos a la presencia de <I>Lactobacillus. </I>Una disminución en el recuento  de los cultivos iniciadores podría estar asociada a la elaboración de los  quesos con leche pasteurizada, debido a que se produce una disminución  en el conteo general de microorganismos por efecto del tratamiento térmico  de la leche [15]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> En la <a href="#tab5">TABLA V </a>se muestra el recuento, expresado en Log UFC/g de <I>Streptococcus</I>  en el queso manchego, <I>Brevibacterium</I> en el Brie y bacterias ácido lácticas  al queso parmesano, por tipo y marca de queso madurado estudiado. Para  el queso Manchego en las dos marcas estudiadas no se encontraron diferencias  significativas, con promedios en M1 de 2,53 y para M2 de 3,13; para el  queso Brie tampoco se detectaron diferencias significativas y sus promedios  fueron para M1 de 6,03 y en M2 de 5,81. En el queso parmesano la cantidad  de bacterias lácticas presentó diferencia significativa (P&lt;0,05) entre  las marcas, obteniéndose promedios de 5 para M1 y 3,52 UFC/g para M2. En  el queso parmesano, los microorganismos utilizados como cultivos iniciadores  con mayor frecuencia son las bacterias ácido lácticas pertenecientes al  género <I>Lactobacillus, </I>las cuales han sido indicados como microorganismos  capaces de producir aminas biógenas.
