El estudio se realizó en 30 granjas con 28 a 2.000 vientres y con diferentes niveles de tecnificación. Los verracos al llegar a la granja fueron sometidos a un periodo de adaptación al manejo y al estatus sanitario. En las granjas se empleaba la monta natural, la inseminación artificial o ambas técnicas. La alimentación de los verracos consistió en alimento comercial. Se realizó un estudio transversal por conglomerados desde septiembre 2005 hasta febrero 2006, muestreándose 170 verracos. La presencia de anticuerpos y partículas virales se determinó mediante las pruebas de ELISA y RT-PCR (Reacción en Cadena de la Polimerasa- Transcriptasa Reversa), respectivamente. Cincuenta y seis verracos fueron positivos a la prueba de ELISA, 21 a la prueba de RT-PCR y 67 a una o ambas pruebas. La prevalencia en los verracos fue 39,4% (67/170) y dentro de granjas varió de 0 a 100%. Veintiún granjas tuvieron al menos un animal positivo a la prueba de ELISA, 13 a la prueba de RT-PCR y 25 a una o ambas pruebas. El riesgo de un animal positivo a PRRS fue 3,6 veces mayor en granjas positivas al virus de PRRS y 2,6 veces mayor en granjas donde no se realizaban pruebas diagnósticas antes de introducir los sementales. Las granjas que utilizaban sementales en la detección de celo tuvieron 7,0 veces mayor riesgo de un verraco positivo al virus de PRRS.]]></p></abstract>

A cluster cross-sectional study was carried out from September 2005 to February 2006, and 170 boars were sampled. The presence of antibodies and virus particles was determined using ELISA and RT-PCR (Reverse transcriptase- polymerase chain reaction) tests respectively. Fifty six out of 170 boars were positive to ELISA test, 21 to RT-PCR and 67 to one or both tests. Boar prevalence was 39.4% (67/170) and within farm varied from 0 to 100%. Twenty one farms (70%) had at least one ELISA positive animal, 13 positives to RT-PCR test (43.3%) and 25 (83.3%) to one or both tests. Aceptado: 20 / 02 / 2009</FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>INTRODUCCIÓN</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El Síndrome Reproductivo y Respiratorio Porcino (PRRS) es una enfermedad  viral que se caracteriza por signos respiratorios en animales de todas  las edades, aumento en las repeticiones de celo en hembras, aborto tardío  y disminución de la fertilidad; incremento de cerdos (<I>Sus scrofa</I>) momificados,  nacidos muertos y débiles, y alta mortalidad de lechones durante la lactancia  [4].</FONT></P>    <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La enfermedad es causada por un virus ARN de cadena sencilla, perteneciente  al orden <I>Nidovirales</I>, familia <I>Arteriviridae, </I>género <I>Arterivirus</I>, grupo  Nidovirus [15]. En los EUA se ha descrito al PRRS como la enfermedad más  devastadora en las explotaciones porcinas [20]. En las piaras, ocurre un  desbalance en las infecciones por microorganismos primarios y secundarios,  por lo que se manifiesta un incremento de enfermedades de diversa índole. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El uso extensivo de la inseminación artificial en las granjas porcinas  ha facilitado el intercambio de características genéticas deseables; sin  embargo, la diseminación de patógenos vía semen es un aspecto importante  que se debe considerar al implementar un programa de salud animal [6]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El virus del PRRS afecta a cerdos de todas las edades, produciendo diversos  signos clínicos de acuerdo a la edad y estado reproductivo. En el caso  de los sementales puede presentarse disminución del apetito, fiebre, disminución  de la libido y alteraciones en la calidad del semen, principalmente disminución  de la motilidad, aumento en anormalidades, daño en acrosomas y un aumento  en el porcentaje de eyaculados rechazados [6]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La transmisión del virus del PRRS mediante el semen de animales infectados  implica un riesgo para las explotaciones porcinas, ya que los animales  infectados eliminan el virus vía semen durante algunos periodos, principalmente  durante la fase aguda de la enfermedad. Se ha notificado que la fase de  eliminación del virus vía semen es muy variable entre los individuos, desde  un mínimo de cuatro hasta 92 días postinfección [6]. También se ha notificado  que la eliminación del virus en semen puede ser de forma intermitente y  no necesariamente estar correlacionada con la viremia o el estatus serológico  [6,15]. El grado de eliminación depende de la cepa viral, de las condiciones  del medio ambiente, y de la cantidad de virus infectante [3]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> En granjas porcinas de Yucatán, México, se encontró una seroprevalencia  individual del 55% al virus del PRRS en la población de cerdos y 58% en  los sementales [8]. En este mismo estudio se menciona que, el uso de reemplazos  externos, el vaciado parcial de la granja y mantener animales de diferentes  edades dentro de una nave, favorecen la presencia del virus en una granja.  El PRRS es una de las enfermedades que causa mayores pérdidas a la industria  porcina [5]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Se ha demostrado que el empleo de sementales portadores del virus de PRRS  es un factor importante en la transmisión y diseminación de la enfermedad  entre granjas y dentro de granja, debido a que el virus puede encontrarse  en el semen [15]. Para llevar a cabo un programa de control del virus de  PRRS en granjas porcinas es necesario conocer la prevalencia de sementales  positivos al virus de PRRS y cuáles son los factores de riesgo asociados  a su presencia. </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Los objetivos de este estudio fueron: Estimar la prevalencia del virus  de PRRS en suero de sementales y determinar algunos factores de riesgo  asociados a la presencia del virus en granjas porcinas de Yucatán, México. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>MATERIALES Y MÉTODOS</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Localización y clima</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El estudio se llevó a cabo en 30 granjas porcinas de Yucatán, localizado  en el sureste de México. El clima de la región es tropical subhúmedo con  lluvias en verano y rango de temperatura de 7 a 42°C, siendo la media de  26,6°C. La humedad relativa varía de 65 a 100% con un promedio de 78% y  la precipitación anual varía de 415 a 1290 mm dependiendo del área. Los  vientos predominantes son de norte a sureste [7]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Selección y descripción de las granjas</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Las 30 granjas encuestadas fueron seleccionadas por conveniencia e interés  de los productores por participar en la encuesta de las 105 granjas pertenecientes  a la asociación de productores de cerdos del estado de Yucatán. Las granjas  se localizaban en un área de alta densidad porcina y se consideraron representativas  de la región centro del Estado. El número de vientres en las granjas encuestadas  varió de 28 a 2.000 y el número de sementales por granja de 1 hasta 32.  El nivel de tecnificación también varió entre granjas, el cual se encuentra  en relación con el número de vientres. Se muestrearon todos los sementales  que tuvieran al menos un mes de estancia en las granjas. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Manejo general de los sementales en las granjas</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Los verracos al llegar a las granjas se sometieron a un periodo de adaptación  al manejo y al estatus sanitario. En las granjas porcinas se manejan dos  tipos de servicio reproductivo: la monta natural y la inseminación artificial.  Algunas explotaciones manejan ambas opciones. La alimentación de los verracos  consistía en el uso de dietas comerciales basadas en soya (<I>Glycine max  L</I>) y sorgo (<I>sorghum vulgare</I>). </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Diseño del estudio</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Se realizó un muestreo por conglomerados en un diseño transversal desde  septiembre 2005 hasta febrero 2006. El total de animales a muestrear (n=170  verracos) se calculó de acuerdo a la fórmula proporcionada por Segura y  Honhold [13].</FONT></P>     ]]></body>

<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> donde: </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana">  <img border="0" src="/img/fbpe/rc/v20n1/art03for2.gif" width="82" height="35">  tamaño de muestra para el muestreo simple para una población infinita;  N = 735 (número estimado de verracos en las 105 granjas porcinas pertenecientes  a la asociación de porcicultores de Yucatán); Z = 1,96<B> </B>valor de tablas  de la distribución de Z con un 95% de confianza; d = 0,10 precisión o error  deseado; D = 2, efecto de diseño; P = 0,50, prevalencia individual esperada;  q = 1-<I>p</I>; n<SUB>c</SUB> = 170, número de verracos a muestrear. Dado que el efecto de  diseño se desconocía a principios del estudio, por conveniencia se tomó  el valor de 2. El número de granjas a visitar (n=30) se obtuvo dividiendo  n<SUB>c</SUB> entre el número promedio de cerdos por granja (k=5,5). </FONT></P>      
<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Obtención y procesamiento de muestras</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Las muestras sanguíneas se obtuvieron de la vena cava anterior utilizando  tubos estériles al vacío de 10 mL y agujas 21 x 1½ mm. Las muestras de  sangre se conservaron a temperatura ambiente aproximadamente por una hora,  y posteriormente se transportaron con temperatura de refrigeración al laboratorio  de Inmunología de la Facultad de Medicina Veterinaria y Zootecnia de la  Universidad Autónoma de Yucatán, donde se centrifugaron a 600 g x 10 minutos  en centrifugadora Damon (Modelo DPR 6000, Damon/IEC División, EUA). Posteriormente,  los sueros se transfirieron a tubos de microcentrifugadora de 1,5 mL por  duplicado, previamente identificados, y se conservaron en un congelador  (marca VWR, modelo 5700; VWR Sales group, EUA) a -70°C hasta su análisis. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Prueba serológica</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La prueba de ELISA indirecta se realizó empleando el kit HerdChek*PRRS  2XR (IDEXX Laboratorios Inc. Westbrook, Maine, EUA), siguiendo las instrucciones  especificadas por el fabricante para detectar la presencia de anticuerpos  contra el virus del PRRS en el suero porcino. La presencia o ausencia de  anticuerpos contra el virus de PRRS se determinó con el valor diagnóstico  conocido como S/P, el cual es la razón del valor de densidad óptica de  la muestra entre la densidad óptica del testigo positivo. Cuando la razón  S/P fue menor a 0,4 la muestra se consideró como negativa; si la razón  S/P fue igual o mayor a 0,4 se consideró como positiva (según indicaciones  del fabricante). </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Extracción del ARN y Prueba de RT-PCR.</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Para la extracción del ARN viral de las muestras de suero, se utilizó el  Kit QIAamp Viral RNA Mini Kit de los laboratorios QIAGEN Inc., EUA. El  ARN viral fue recuperado según las indicaciones del fabricante con búfer  de elución y almacenado a -70°C. Para las pruebas de PCR se utilizó el  kit comercial OneStep RT-PCR QIAGEN, siguiendo las instrucciones del fabricante  y utilizando 8 µL del ARN extraído en un volumen total de 50 µL de la mezcla  (master mix). Los iniciadores que se utilizaron fueron del marco de lectura  abierta 7 (ORF7) derivados de la secuencia del VR-2332 PRRSV y sintetizados  por la compañía Accesorios para Laboratorios, S.A. de C.V. México DF. Para  la amplificación en RT-PCR se utilizó el iniciador de sentido (forward)  5’-TCGTGTTGGGTGGCAGAAAAGC-3’ (nucleótidos 2763 a 2785) y el iniciador antisentido  (reverse) 5’-GCCATTCACCACACATTCTTCC-3’. Los productos amplificados de 482  pb fueron visualizados en geles de agarosa al 2% y teñidos con bromuro  de etidio. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Prevalencia y factores de riesgo</B> </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Un caso se definió como aquel cerdo positivo a la prueba de ELISA o RT-PCR.  La prevalencia se estimó como el número de verracos positivos entre el  total de verracos muestreados. Se aplicó un cuestionario para determinar  posibles factores de riesgo asociados al virus de PRRS en sementales porcinos.  El cuestionario incluyó los datos generales de cada granja así como preguntas  asociadas a la población animal y a la bioseguridad. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La información sobre la positividad de los verracos se utilizó para el  estudio de los factores de riesgo, el cual se realizó en dos etapas: 1)  Se corrieron pruebas de Ji-cuadrado para la positividad a PRRS con cada  uno de los factores de riesgo. 2) Los factores de riesgo que resultaron  significativos (P&lt;0,20) a la prueba de Ji-cuadrado se incluyeron en un  análisis de regresión logística marginal (ajustado por hato) multivariado.  Este modelo consideró el efecto de la correlación entre verracos dentro  de granja y para ello se utilizó el procedimiento GENMOD [12]. El modelo  de regresión logística marginal se especificó por la distribución binomial  y la función de ligue logit. Las razones de momios (OR) ajustados y errores  estándares se obtuvieron mediante el método de ecuaciones generalizadas  estimadas usando una matriz de correlaciones de simetría compuesta entre  observaciones dentro de una misma granja. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>RESULTADOS Y DISCUSIÓN</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Prevalencia</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Cincuenta y seis animales de los 170 sementales fueron positivos a la prueba  de ELISA, 21 fueron positivos a la prueba de RT-PCR y 67 sementales resultaron  positivos a una o ambas pruebas. La prevalencia encontrada al virus de  PRRS en sementales de las granjas porcinas fue 40,6% (67/170). Veinticinco  de las treinta granjas muestreadas (83,33%) tuvieron al menos un semental  positivo al virus de PRRS. La prevalencia encontrada en este estudio (40,6%)  es menor que la notificada en la misma región por otros autores [8], quienes  mencionan prevalencias en sementales del 58 y 55% de animales positivos  en todas las edades y etapas del ciclo productivo. En Guanajuato, Puebla,  México, Sonora y Veracruz, se obtuvo una seroprevalencia global del 62,2%  en cerdos de todas las edades [14]. La seroprevalencia en el presente trabajo  es menor, debido posiblemente, a un control en las granjas donde se realizan  pruebas diagnósticas y se eliminan los sementales que resulten positivos. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Estudios realizados en México mencionan, del 78 al 100% de hatos positivos  al virus del PRRS [8,14]. En el presente estudio sólo se muestrearon a  los sementales y se encontró que el 83% de las granjas tienen al menos  un semental positivo; sin embargo, no se puede inferir que las otras granjas  se encuentre libres de la enfermedad, ya que no se colectaron muestras  de otros segmentos de la población como las hembras reproductoras, lechones,  engorda, etc., donde podría estar presente el virus. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Los sementales en granjas porcinas se encuentran expuestos a la enfermedad  debido a que están en constante contacto con las hembras, principalmente  cuando se les utiliza para detectar celos y/o en monta natural. Por otra  parte, un semental infectado pone en riesgo a todas las hembras con las  que tiene contacto, además el PRRS puede transmitirse por semen y por lo  tanto, pone en riesgo también a las hembras inseminadas artificialmente. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Factores de riesgo</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Los nueve factores de riesgo estudiados se presentan en la <a href="#tab1">TABLA I</a>. Las  pruebas de Ji cuadrado indicaron que de los nueve factores, sólo cinco  fueron significativos (P&lt;0,20): Conocer si la granja es positiva a PRRS,  no realizar pruebas diagnósticas antes de ingresar los sementales a la  granja, no realizar pruebas diagnósticas periódicamente, no cuarentenar  a los sementales y utilizar los sementales para detección de celo. Pero  en el modelo de regresión logística marginal sólo tres factores resultaron  significativos (P&lt;0,05): granja positiva a PRRS, no realizar pruebas diagnósticas  antes de ingresar los sementales a la granja y utilizar a los sementales  para la detección de celo (<a href="#tab2">TABLA II</a>). Los sementales en granjas positivas  presentaron 3,6 veces más riesgo de infección que aquellos que se encontraban  en granjas identificadas como negativas al PRRS. Esto se debe a que los  sementales se encuentran a poca distancia (dentro de la misma granja) del  resto del hato y entran en contacto con las hembras para la monta o para  detección de celo lo cual aumenta el riesgo de infección. Por otro lado,  los sementales en granjas negativas están en menor riesgo ya que no entran  en contacto directo con animales infectados. Algunos autores [9,16,18]  mencionan que el uso de sementales positivos o de semen proveniente de  sementales positivos es un factor de riesgo para la infección de hembras  reproductoras, esto debido a que cualquier animal positivo dentro de una  granja representa un riesgo para el resto del hato, tanto los sementales  para las hembras como las hembras, hacia los sementales. Aunque se debe  tomar en cuenta que el riesgo que presentan los sementales puede ser mucho  más importante debido a la inseminación artificial, poniendo en riesgo  a gran cantidad de hembras. En las 30 granjas muestreadas se tiene una  población de 7.816 vientres, de las cuales 7.051 se encuentran en las 25  granjas con sementales positivos, además si el semen de animales infectados  se utiliza para inseminar hembras de otras granjas se aumenta el número  de hembras en riesgo.</FONT></P>     <P ALIGN="CENTER"><b><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <a name="tab1">TABLA I</a></FONT></b></P>     ]]></body>

Es importante  realizar muestreos para prevenir la introducción de animales en periodo  de incubación, en fase latente o con enfermedad subclínica a la granja.  Weigel y col. [16] y Mousing y col<I>.</I> [10] mencionan que el introducir animales  de reemplazo es un factor de riesgo para la presencia de la enfermedad,  lo cual demuestra la importancia de realizar pruebas diagnósticas para  asegurase que los animales se encuentren sanos. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Utilizar a los sementales para la detección del celo es el factor de riesgo  detectado más importante, representando 7,0 veces más probabilidades de  infección comparada con aquellas granjas donde no se emplea a los sementales  para identificar a las hembras en celo. Esta práctica pone en riesgo a  los sementales, ya que entran en contacto directo con las hembras que pueden  ser positivas al virus de PRRS. El virus del PRRS se transmite principalmente  por medio del contacto directo entre un animal enfermo y uno susceptible,  probablemente por contacto nariz-nariz o por contacto con orina y/o heces  infectadas [1, 2, 11]. Los animales infectados pueden eliminar el virus  del PRRS en saliva hasta 42 días postinfección [17], en secreciones nasales  hasta 38 días postinfección, en orina 28 días postinfección [11] y en heces  hasta 35 días postinfección [19], por lo que el riesgo de infección al  juntar animales susceptibles con animales enfermos es alto. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Los factores: no realizar pruebas diagnósticas periódicamente y no cuarentenar  a los sementales, fueron estadísticamente significativos (P&lt;0,05) en el  estudio de regresión logística estándar pero no en el de regresión logística  marginal. Sin embargo, las pruebas diagnósticas periódicas son importantes  para conocer el estado sanitario de la granja y de esta manera poder detectar  un brote a tiempo y tomar las medidas de control necesarias. Por otro lado,  el cuarentenar a los animales antes de introducirlos a la granja es una  medida de bioseguridad importante para una explotación porcina, ya que  los animales pueden llegar en fase de incubación y salir negativos en las  pruebas diagnósticas pero presentar la enfermedad más adelante. El no aislar  a los animales después de la compra incrementa 3,3 veces más las posibilidades  de infección en la granja si no se toma esta medida [16]. </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El empleo de inseminación artificial ha sido notificado como un factor  de riesgo para la infección de los vientres en una granja porcina [16],  encontrándose que la compra de dosis de semen para inseminación representa  un riesgo (OR=2,2) para la infección en el hato reproductor. Sin embargo,  esto no representa un riesgo para los sementales, ya que por medio de la  colecta de semen para la inseminación artificial no tienen contacto directo  con animales infectados. Otra variable estudiada fue la frecuencia de reemplazo  de los sementales, la cual no resultó significativa. Aunque la entrada  de animales nuevos a la granja esté relacionada con la presentación de  enfermedades, si se tienen las medidas de bioseguridad adecuadas, esto  puede prevenirse. La frecuencia con que se reemplacen los sementales no  es tan importante como el realizar pruebas diagnósticas y cuarentenar antes  de introducirlos a la granja. Contar con personal exclusivo para el área  de sementales y el origen de los sementales (local, nacional o internacional)  no son variables significativas para la presentación del PRRS en sementales  de granjas porcinas de acuerdo a lo encontrado en este estudio. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>CONCLUSIONES</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El virus del PRRS se encuentra presente en los sementales de las granjas  porcinas del estado de Yucatán con una prevalencia de 40,6%. El 83,3% de  las granjas muestreadas presenta al menos un semental positivo a esta enfermedad.
