La primera dieta fue una dieta control no pigmentada (C); la segunda contenía 100 mg kg-1 de Carofil Rosa (CR); la tercera y cuarta contenía 200 (CR200) y 250 (CR250) mg kg-1 de oleorresina saponificada de chile rojo (Capsicum annuum), respectivamente. Los resultados mostraron que existieron diferencias significativas (p <= 0.05) entre las dietas C y CR250 con respecto al índice gonadosomático (IGS), peso promedio de la gónada (PG) y el diámetro de los ovocitos (DO), presentando la última dieta los valores más altos de estos factores. Los tratamientos de CP y CR200 presentaron valores similares. Al final del experimento solamente se observó el 10% de ovocitos maduros en la dieta CR250.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[Four groups of rainbow trout (Onchorhynchus mykiss) each with 60 females with an average body weight of 467 g were submitted to an aleatory assay in order to compare the efficiency of four diets containing different concentrations of carotenoid pigments, and to determine if pigment concentration and source had an influence on female gonad development and oocyte maturity. The first diet was the non-pigmented control (C). The second diet contained 100 mg kg-1 of Carophyll Pink (CR). The third and fourth diets contained 200 (RC200) and 250 (RC250) mg kg-1 of saponified red chili oleoresin, respectively. The results indicated significant differences (p <= 0.05) between C and RC250, with respect to the gonado-somatic index (IGS), average gonad weight (PG) and average diameter of oocyte (DO), with CR250 showing higher IGS, PG and DO values. <body><![CDATA[<P ALIGN="JUSTIFY"><font size="3"><b>Recibido: </b> 11-07-2003</font>&nbsp; <font size="3"><b>Aceptado: </b> 31-05-2004</font></P>     <P ALIGN="JUSTIFY"><B><font size="3">INTRODUCCION</font></P> </B>     <P ALIGN="JUSTIFY"><font size="3">Durante los &uacute;ltimos a&ntilde;os, se ha incrementado notablemente el uso de pigmentos carotenoides en el cultivo de la trucha arco iris y el salm&oacute;n, debido a que estos producen un atractivo color en el m&uacute;sculo, que incrementa su aceptaci&oacute;n por los consumidores y resulta en un incremento de hasta el 30% de su valor econ&oacute;mico. Los Salm&oacute;nidos no son capaces de sintetizar de novo estos pigmentos, por lo que resulta necesario incorporarlos en el alimento balanceado durante su proceso de cultivo.</font></P>     <P ALIGN="JUSTIFY"><font size="3">La depositaci&oacute;n de los pigmentos carotenoides en los Salm&oacute;nidos no es constante a lo largo de su ciclo de vida, ya que varia de acuerdo a la edad, la &eacute;poca del a&ntilde;o y el estado de madurez (1,2). Los pigmentos agregados en las dietas balanceadas, no necesariamente son fijados en el tejido muscular del pez, sino que tambi&eacute;n se concentran a trav&eacute;s de diferentes rutas metab&oacute;licas en otros tejidos, especialmente en la piel y en las g&oacute;nadas (3, 4). La maduraci&oacute;n sexual de los peces Salm&oacute;nidos, esta asociada a un periodo inicial de r&aacute;pido crecimiento, seguido por un intenso desarrollo de las g&oacute;nadas, un descenso en la tasa de crecimiento y una disminuci&oacute;n marcada en el consumo de alimento previo a la reproducci&oacute;n (5,6). Durante la madurez sexual, las truchas muestran bajas concentraciones de carotenoides en el tejido muscular, debido a que &eacute;stos pigmentos se distribuyen a otros &oacute;rganos y tejidos, as&iacute; por ejemplo, la astaxantina se mueve del tejido muscular y se acumula en las g&oacute;nadas en el caso de las hembras y en la piel en el caso de los machos (7- 10). Se ha visto que durante la maduraci&oacute;n sexual, cantidades considerables de pigmentos carotenoides son transportados a los ovocitos en donde es posible registrar hasta el 18% del total de carotenoides, que es similar a la encontrada en el tejido muscular (2). Se ha sugerido que la derivaci&oacute;n de los carotenoides a las g&oacute;nadas, esta relacionado con los procesos de maduraci&oacute;n y fertilizaci&oacute;n de los &oacute;vulos maduros. De acuerdo con esta informaci&oacute;n, los pigmentos carotenoides juegan un papel destacado en los procesos de maduraci&oacute;n de las g&oacute;nadas y de los ovocitos, as&iacute; como en el desarrollo embrionario y la sobrevivencia de los alevines en los Salm&oacute;nidos en general.</font></P>     <P ALIGN="JUSTIFY"><font size="3">En relaci&oacute;n a los pigmentos naturales como aquellos obtenidos del Chile rojo (Capsicum annuum), actualmente no existen estudios acerca de sus efectos sobre el desarrollo gonadal en las especies de Salm&oacute;nidos y mucho menos de las v&iacute;as metab&oacute;licas utilizadas por la capsantina (el principal pigmento carotenoide del Chile rojo), aunque se puede asumir, que probablemente las v&iacute;as metab&oacute;licas son similares a la astaxantina sint&eacute;tica, aunque su efecto sobre el desarrollo gonadal y en particular sobre los ovocitos no esta bien definido.</font></P>     <P ALIGN="JUSTIFY"><font size="3">El objetivo del presente trabajo, consisti&oacute; en investigar la influencia de los pigmentos carotenoides como la astaxantina sint&eacute;tica y la capsantina incluida en la oleorresina saponificada del Chile rojo (Capsicum annuum), sobre el desarrollo de las g&oacute;nadas y la maduraci&oacute;n de los ovocitos, en hembras de trucha arco iris de primera reproducci&oacute;n, bajo condiciones de cultivo.</font></P> <B>    <P ALIGN="left"><font size="3">MATERIALES Y METODOS</font></P>     <P ALIGN="JUSTIFY"><font size="3">Preparaci&oacute;n de las dietas</font></P> </B>    <P ALIGN="JUSTIFY"><font size="3">Las dietas experimentales, fueron asignadas de manera aleatoria y consistieron en una dieta control (C) sin ning&uacute;n pigmento, la dieta de Carofil rosa (CR), que incluy&oacute; el pigmento sint&eacute;tico Carofil Rosa y dos dietas con extracto de oleorresina saponificada de Chile rojo (Capsicum annuum) (CR200 y CR250), (<a href="#TABLA 1">Tabla 1</a>). Las dietas experimentales se prepararon utilizando una dieta basal para engorde de trucha arco iris, elaborada por la Empresa Alimentos Balanceados Almaz&aacute;n (bajo Licencia de Silver Cup, USA), la cual conten&iacute;a 40.6 ± 0.12% de prote&iacute;na cruda, 18.8 ± 0.02% de l&iacute;pidos y 1.1 ± 0.11% de fibra cruda. A esta dieta se le a&ntilde;adi&oacute; el aceite de pescado y los pigmento carotenoides. Posteriormente, se coloc&oacute; en un molino para obtener &quot;pellets&quot; de 8 mm de di&aacute;metro, los que fueron secados durante 12 horas a 45 <FONT FACE=Symbol>&#176;</FONT> C y despu&eacute;s de esto, se mantuvieron almacenados en bolsas pl&aacute;sticas negras en refrigeraci&oacute;n a 4 <FONT FACE=Symbol>&#176;</FONT> C hasta su uso.</font></P>     <P ALIGN="CENTER"><a name="TABLA 1"><font size="3">TABLA 1</font></a></P>     ]]></body>

Extracto saponificado de harina de Chile rojo (Capsicum annuum), emulsificado, estabilizado con antioxidantes y dispersado en agua.</font></P>     <P ALIGN="JUSTIFY"><font size="3">Partici&oacute;n relativa por cromatograf&iacute;a: Total de dihidroxipigmentos rojos 40-45%; Dihidroxicapsantina 35-45%; Dihidroxicapsorubina 0.5-2%; Dihidroxiluteina 1-8%; Dihidroxizeaxantina 1-8%; Total de carotenoides 10-20%.</font></P> </DIR> </DIR>  <B>    <P ALIGN="JUSTIFY"><font size="3">Procedimiento experimental</font></P> </B>    <P ALIGN="JUSTIFY"><font size="3">Se utilizaron doscientos cuarenta hembras de trucha arco iris con un peso promedio de 476 ± 63.5 g y nueve meses de edad, las cuales fueron separadas en cuatro grupos experimentales de 60 peces cada uno. Las truchas fueron introducidas en estanques circulares de 4,000 litros cada uno, conectados a un sistema cerrado de recirculaci&oacute;n y reacondicionamiento de agua. Las truchas fueron climatizadas por un periodo de 20 d&iacute;as previo al experimento, con el objetivo de estandarizar el peso total promedio del grupo y el experimento tuvo una duraci&oacute;n de 45 d&iacute;as. Las truchas fueron alimentadas dos veces al d&iacute;a, con el 15% de su peso vivo. La biomasa para cada grupo experimental fue registrada cada 15 d&iacute;as y sirvi&oacute; para ajustar la entrega de las cinco dietas.</font></P>     <P ALIGN="JUSTIFY"><font size="3">Se registraron los siguientes par&aacute;metros de calidad del agua: temperatura del agua y concentraci&oacute;n de ox&iacute;geno disuelto, mediante un ox&iacute;metro YSI modelo 57 (Yellow Springs Instrument Company, Yellow Spring, Ohio, USA) y pH con un potenci&oacute;metro digital marca Orion, modelo 410A. Nitr&oacute;geno amoniacal total (NAT), amoniaco (N-NH<SUB>3</SUB>), nitrito (N-NO<SUB>2</SUB><SUP>-</SUP>) y nitrato (N-NO<SUB>3</SUB><SUP>-</SUP>), con un equipo Hach 2000/DR (Hach Company, Loveland, Co. USA).</font></P> <B>    <P ALIGN="JUSTIFY"><font size="3">Determinaci&oacute;n de los estados de maduraci&oacute;n de las g&oacute;nadas y los ovocitos</font></P> </B>    <P ALIGN="JUSTIFY"><font size="3">Cada 15 d&iacute;as, se sacrificaron aleatoriamente 20 peces de cada grupo y se registr&oacute; la longitud total en cent&iacute;metros (LT), peso total en gramos (PT) y el peso de la g&oacute;nada en gramos (PG). El &iacute;ndice gonadosom&aacute;tico (IGS) fue estimado de acuerdo con el criterio de Rossemblum y colaboradores (11), mediante la f&oacute;rmula siguiente:</font></P>     <P ALIGN="JUSTIFY"><font size="3">IGS = (Pg/Pt) * 100</font></P>     <P ALIGN="JUSTIFY"><font size="3">Donde:</font></P>     <P ALIGN="JUSTIFY"><font size="3">IGS = &Iacute;ndice gonadosom&aacute;tico del pez.</font></P>     ]]></body>
<body><![CDATA[<P ALIGN="JUSTIFY"><font size="3">Pg = Peso de las g&oacute;nadas (g).</font></P>     <P ALIGN="JUSTIFY"><font size="3">Pt = Peso total (g).</font></P>     <P ALIGN="JUSTIFY"><font size="3">Ocho pares de g&oacute;nadas de cada grupo experimental fueron seleccionadas aleatoriamente y fijadas en formol al 10%. De cada g&oacute;nada se tomaron 100 ovocitos, los cuales se observaron al microscopio &oacute;ptico (Kart Zeiss Company) y se midieron con oculares microm&eacute;tricos de 10x, 16x, 40x y 100x, y se obtuvo el di&aacute;metro promedio de los ovocitos (DO). Para determinar el grado de desarrollo de los ovocitos, &eacute;stos fueron sumergidos en una soluci&oacute;n aclaradora (12) y se clasificaron de acuerdo a la escala de madurez de Tyler y colaboradores (5), quienes consideran cinco clases de talla de acuerdo a su di&aacute;metro: I, formada por ovocitos menores de 0.4 mm; II, de 0.4 a 0.59 mm; III, de 0.6 a 0.79 mm; IV, 0.8 a 0.99 mm y V, de 1.0 a 1.2 mm. Los ovocitos mayores de este di&aacute;metro se consideran vitelog&eacute;nicos y los fol&iacute;culos maduros alcanzan un di&aacute;metro promedio de 5 mm.</font></P> <B>    <P ALIGN="JUSTIFY"><font size="3">An&aacute;lisis estad&iacute;stico</font></P> </B>    <P ALIGN="JUSTIFY"><font size="3">Para determinar si exist&iacute;an diferencias significativas entre los grupos experimentales y los tiempos, con respecto al &iacute;ndice gonasom&aacute;tico (IGS), el peso promedio de las g&oacute;nadas (PG) y el di&aacute;metro de los ovocitos (DO), se aplic&oacute; un An&aacute;lisis de Varianza de una sola v&iacute;a y una prueba a posteriori de Tuckey HSD, por medio del Programa Statistica (Statsoft Inc., OK, USA).</font></P>     <P ALIGN="JUSTIFY"><B><font size="3">RESULTADOS Y DISCUSION</font></P> </B>     <P ALIGN="JUSTIFY"><font size="3">Los par&aacute;metros fisicoqu&iacute;micos registrados a lo largo del periodo experimental se mantuvieron constantes (<a href="#TABLA 2">Tabla 2</a>) y sus promedios indican que las condiciones experimentales fueron adecuadas, permitiendo una buena sobrevivencia y un adecuado crecimiento de las truchas, resultados que est&aacute;n acordes con los reportados por Klontz (13) para el cultivo de la trucha arco iris.</font></P>     <P ALIGN="CENTER"><a name="TABLA 2"><font size="3">TABLA 2</font></a></P>     <P ALIGN="CENTER"><font size="3">Valores promedios de los par&aacute;metros fisicoqu&iacute;micos registrados en los estanques de producci&oacute;n a lo largo del experimento</font></P>     <P ALIGN="CENTER">    ]]></body>

Estos datos confirman que la calidad del agua en el sistema cerrado de recirculaci&oacute;n y reacondicionamiento, fue apropiada. Adem&aacute;s, la dieta cubri&oacute; adecuadamente los requerimientos nutricionales. &Uacute;nicamente el amoniaco (N-NH<SUB>3</SUB>) present&oacute; concentraciones altas, que son considerados dentro del rango subletal para la trucha arco iris (&gt; 0.05 mg/L) Klontz (13); no obstante ello, las hembras de trucha arco iris no mostraron s&iacute;ntomas de intoxicaci&oacute;n, ni exhibieron una conducta anormal, ni tampoco presentaron mortalidad.</font></P>     <P ALIGN="JUSTIFY"><font size="3">Los valores promedios del &iacute;ndice gonadosom&aacute;tico (IGS) registrados en la dieta control (C), no mostraron diferencias significativas (p <FONT FACE=Symbol>&#179;</FONT>  0.05) con respecto a los tiempos de muestreo, mientras que las dietas CR y CR200, s&iacute; mostraron diferencias (p <FONT FACE=Symbol>&#163;</FONT>  0.05) entre los 15 y 30 d&iacute;as, pero no entre los 30 y 45 d&iacute;as. Despu&eacute;s de los 30 d&iacute;as, los ovocitos mostraron un considerable incremento en tama&ntilde;o en las dietas CR, CR200 y CR250. En este periodo, los ovocitos presentaron ves&iacute;culas vitelinas en la mayor parte del citoplasma, un n&uacute;cleo esf&eacute;rico con varios nucleolos en la periferia. <body><![CDATA[<P ALIGN="JUSTIFY"><font size="3">A los 45 d&iacute;as, las g&oacute;nadas presentaron ovocitos bien zonificados, con diferentes di&aacute;metros y estados de desarrollo. Los ovocitos en la parte central de la g&oacute;nada mostraron di&aacute;metros m&aacute;s grandes y un mayor desarrollo. Se observaron diferencias entre las dietas C y CR250. La primera mostr&oacute; el 97% de los ovocitos con di&aacute;metros reducidos y sin evidencia de ves&iacute;culas vitelinas. En contraste, en la &uacute;ltima dieta el 10% de los ovocitos mostr&oacute; evidencias de una ves&iacute;cula germinal desplazada hacia la periferia, lo que signific&oacute; la presencia de fol&iacute;culos totalmente maduros. Las dietas CR y CR200, mostraron caracter&iacute;sticas similares, presentando ovocitos de un mayor tama&ntilde;o, con grandes cantidades de ves&iacute;culas vitelinas, aunque la ves&iacute;cula germinal se mantuvo en una posici&oacute;n central.</font></P>     <P ALIGN="JUSTIFY"><font size="3">Los resultados del IGS, PG y DO, se&ntilde;alan que el crecimiento de las g&oacute;nadas a lo largo del experimento fue diferente en cada tratamiento y que los efectos de los pigmentos astaxantina y el extracto de la oleorresina saponificada de Chile rojo, fueron evidentes sobre el desarrollo de las g&oacute;nadas. Debido a que no se observaron diferencias significativas (p <FONT FACE=Symbol>&#179;</FONT>  0.05) entre la ganancia de peso diario en los tratamientos, las diferencias observadas en el desarrollo gonadal no pueden ser atribuidas a la dieta. En este sentido, se estima que las diferencias exhibidas por estos indicadores son debidas principalmente a la concentraci&oacute;n y al origen de los pigmentos carotenoides incorporados en la dieta. El mayor desarrollo de las g&oacute;nadas y de los ovocitos, fue observado en la dieta CR250 y el m&aacute;s bajo en la dieta control.</font></P>     <P ALIGN="JUSTIFY"><font size="3">Los resultados obtenidos en este trabajo, confirman que los pigmentos carotenoides sint&eacute;ticos y naturales, fueron redistribuidos y acumulados durante el desarrollo de las g&oacute;nadas, siendo transportados por la sangre y acumuladas en las g&oacute;nadas (4-10,14). A pesar del grado de desarrollo alcanzado por los ovocitos en la dieta CR250, el 90% de ellos fueron inmaduros, lo cual pudo deberse a que las hembra utilizadas en este experimento, todav&iacute;a no alcanzaban su madurez sexual ya que la temperatura del agua a lo largo del experimento fue en promedio de 19 ºC. Generalmente, las hembras de trucha arco iris alcanzan su primera madurez sexual al a&ntilde;o o a los dos a&ntilde;os de edad y la temperatura &oacute;ptima del agua para la reproducci&oacute;n fluct&uacute;a entre los 5.5 a 8 ºC (15). En nuestro caso particular, estas dos condiciones no se cumplieron. Sin embargo, la influencia directa de los pigmentos utilizados en este experimento sobre la maduraci&oacute;n de las g&oacute;nadas fue evidente. Hasta el momento, la &uacute;nica informaci&oacute;n disponible sobre el efecto de los pigmentos sobre la maduraci&oacute;n de las g&oacute;nadas, es mediante el uso de pigmentos sint&eacute;ticos tales como la astaxantina y cantaxantina (16-19) y queda a&uacute;n mucho que aprender, sobre los efectos en la maduraci&oacute;n gonadal de los pigmentos naturales como la capsantina, que es el componente m&aacute;s importante de extracto de oleorresina saponificada de Chile rojo (Capsicum annuum).</font></P>     <P ALIGN="JUSTIFY"><font size="3">La evidente relaci&oacute;n lineal, encontrada en este estudio entre los pigmentos carotenoides y el desarrollo de las g&oacute;nadas y los ovocitos, indica el papel fundamental que representan &eacute;stos durante el proceso de desarrollo gonadal, sin embargo, es necesario realizar investigaciones m&aacute;s detalladas, que permitan conocer m&aacute;s a fondo los mecanismos de transporte y las v&iacute;as metab&oacute;licas de la capsantina, as&iacute; como encontrar la concentraci&oacute;n &oacute;ptima del pigmento para promover el desarrollo gonadal de las hembras de trucha arco iris. Este &uacute;ltimo aspecto, es de importancia pr&aacute;ctica, ya que permitir&iacute;a mejorar la producci&oacute;n masiva de alevines y cr&iacute;as en las granjas productoras de trucha arco iris, con lo cual se mejorar&iacute;a la rentabilidad econ&oacute;mica.</font></P> <B>    <P ALIGN="left"><font size="3">AGRADECIMIENTOS</font></P> </B>     <P ALIGN="JUSTIFY"><font size="3">Este trabajo forma parte de la tesis Doctoral del primer autor, en el Programa de Doctorado en Ciencias Biol&oacute;gicas, de la Universidad Aut&oacute;noma Metropolitana Iztapalapa.
