<abstract abstract-type="short" xml:lang="en"><p><![CDATA[A number of studies suggest that acetylcholine (ACh) and 5-hydroxytryptamine (5-HT) interrelationship participate on the regulation of physiological and behavioral processes like secretion of corticotropin releasing factor and memory function. The effects of diisopropyl fluorophosphate (DFP), an irreversible inhibitor of acetylcholinesterase, upon the 5-HT and 5-hydroxyindol acetic acid (5-HIAA) levels in six brain regions of the adult rat were studied. The concentration of monoamine and its metabolite were determined by high pressure liquid chromatography (HPLC). The relation metabolite/neurotransmitter was employed as an indicator of neuronal activity. The results indicate that DFP decreased the 5-HIAA levels in hypothalamus, without any modification on the 5-HT levels. In the other brain regions, it was not any change either on 5-HIAA or 5-HT levels.]]></p></abstract>

<body><![CDATA[<p>Key Words:</b> Serotonin, Turnover, Diisopropyl Fluorophosphate.</p>      <p>&nbsp;</p> <b>      <p ALIGN="JUSTIFY">Introducción</p> </b>      <p ALIGN="JUSTIFY">Los hallazgos de varios estudios sugieren que la interacción de la acetilcolina (ACh) y la 5-hidroxitriptamina (5-HT) participa en la regulación de procesos fisiológicos y conductuales tales como la secreción del factor liberador de corticotropina y la memorización (Kile y Turner, 1.985; Lehmann, et al 2000). Más aún, se considera que la pérdida del balance de los sistemas colinérgico y serotonérgico juega un papel importante en el desarrollo de la enfermedad de Alzheimer y la ansiedad (Decker y McGaugh, 1991; File, et al 2000).</p>      <p ALIGN="JUSTIFY">El efecto del aumento de la ACh cerebral, producida por el diisopropilfluorofosfato (DFP), un inhibidor de la acetilcolinesterasa, sobre la actividad serotonérgica depende, entre otros factores, de la vía de administración de la droga y de la región cerebral analizada (Fatehyab, et al 1980; Robinson, et al 1986).</p> <b>      <p ALIGN="JUSTIFY">Objetivo</p> </b>      <p ALIGN="JUSTIFY">En el presente trabajo se estudió el efecto del diisopropilfluorofosfato sobre los niveles de 5-HT y del ácido 5-hidroxiindolacético (5-HIAA) en: hipotálamo, tallo cerebral, hipocampo, corteza cerebral, mesencéfalo y en el estriado de la rata. </p> <b>      <p ALIGN="JUSTIFY">Materiales y Métodos </p>      <p ALIGN="JUSTIFY">Animales experimentales</p> </b>      <p ALIGN="JUSTIFY">Se utilizarn ratas machos adultos de la cepa <i>Sprague Dawley</i>, peso comprendido entre 290-310 g, con ingesta de agua y alimento (Ratarina comercial) <i>ad libitum</i>.</p> <b>      ]]></body>
<body><![CDATA[<p ALIGN="JUSTIFY">Drogas y reactivos químicos</p> </b>      <p ALIGN="JUSTIFY">Diisopropilfluorofosfato, albúmina de bovino, EDTA, NaOH, ácido perclórico y Carbonato de sodio fueron provisto por Sigma Chemical Co. </p> <b>      <p ALIGN="JUSTIFY">Métodos</p> </b>      <p ALIGN="JUSTIFY">Previa anestesia con pentobarbital sódico (50 mg/kg peso), a los animales se les administró DFP (0.1 </font><font FACE="Symbol" SIZE="3">m</font><font SIZE="3">g/1 </font><font FACE="Symbol" SIZE="3">m</font><font SIZE="3">l solución salina) o el vehículo en el ventrículo lateral derecho (Paxinos y Watson, 1982). Después de cuatro horas, los animales fueron sacrificados por decapitación y se realizó la disección de las regiones cerebrales de acuerdo a la técnica de Glowinski e Iversen (1966).</p>      <p ALIGN="JUSTIFY">Los niveles tisulares de las aminas y de sus metabolitos se determinaron por el método de Murai, et al (1988). La concentración de proteínas se determinó por el método de Markwell, et al (1978). </p>      <p ALIGN="JUSTIFY">El análisis estadístico de los resultados se realizó por la prueba de <i>t</i> de <i>Student </i>para datos no pareados. Valores de p&lt;0.05 se consideraron significativos. </p> <b>      <p ALIGN="JUSTIFY">Resultados</p> </b>      <p ALIGN="JUSTIFY">El DFP produjo una disminución de los niveles de 5-HIAA en el hipotálamo, sin modificación de los niveles de 5-HT <i><b><a href="#fig1">(figura 1)</a></b></i>. En el resto de las regiones, no se encontró modificación de los niveles de 5-HT ni de 5-HIAA, tal como puede observarse en la <i><b><a href="#fig1">figura 1</a></b></i>. </p>      <p ALIGN="JUSTIFY">Tanto en el grupo control como en el experimental, la relación 5-HIAA/5-HT fue mayor en el hipocampo en comparación con el tallo (p&lt;0.001), el mesencéfalo (p&lt;0.05) y la corteza (p&lt;0.05) <i><b><a href="#fig2">(figura 2)</a></b></i>. <p ALIGN="JUSTIFY">Los valores representan la X ± EMM de 9 observaciones.</p> </font></i><font SIZE="3"><b>      <p ALIGN="JUSTIFY">Discusión y conclusiones</p> </b>      <p ALIGN="JUSTIFY">En este trabajo se estudió el efecto del DFP sobre los niveles de 5-HT y 5-HIAA en una forma bastante amplia, analizando áreas como el mesencéfalo donde se localizan somas serotonérgicos y áreas que poseen terminales serotonérgicos y colinérgicos tales como la corteza y el hipocampo (Frazer y Hensler, 1999; Jones, 1993). Se utilizó la relación 5-HIAA/5-HT como un índice de la actividad neuronal, basándonos en los hallazgos de Shannon, et al (1986), según los cuales esta relación representa un indicador adecuado de la actividad serotonérgica cerebral. </p>      <p ALIGN="JUSTIFY">A diferencia de trabajos previos, en nuestro estudio se administró el DFP por vía intracerebroventricular, lo cual, tiene la ventaja sobre la administración sistémica, en que se evita la posible participación de mecanismos periféricos en los efectos estudiados; además, permitiría observar mecanismos integradores cerebrales que no serían observados cuando la droga es aplicada <i>in situ</i> (Fatehyab, et al 1980) </p>      ]]></body>
<body><![CDATA[<p ALIGN="JUSTIFY">En nuestro estudio, se observó una disminución de los niveles de 5-HIAA, sin modificación de los niveles de 5-HT, en el hipotálamo después de la administración del DFP, lo cual sugiere que hubo una disminución de la liberación del neurotransmisor e indica que en condiciones normales el sistema colinérgico ejerce una influencia inhibidora sobre el sistema serotonérgico en esta región cerebral. Este hallazgo coincide con los de estudios previos que han encontrado que la disminución de la actividad colinérgica produjo un aumento del recambio de la serotonina en varias regiones del cerebro de rata, entre ellas el hipotálamo (Hortnagl, et al 1987). </p>      <p ALIGN="JUSTIFY">En el hipotálamo se ha demostrado la existencia de neuronas positivas para la acetilcolinesterasa y de terminales serotonérgicos (Saavedra, et al 1974; Sethy, et al 1973), por lo tanto pudiera especularse que la ACh liberada del terminal colinérgico influiría posteriormente en la liberación de 5-HT del terminal serotonérgico.</p>      <p ALIGN="JUSTIFY">&nbsp;</p> <b>      <p ALIGN="JUSTIFY">Referencias bibliográficas</p> </b>  <dir>   <dir>         <!-- ref --><p ALIGN="JUSTIFY">1.
