No forma parte de la flora normal del humano, mientras que los peces dulceacuícolas como las tilapias se tienen como su reservorio primario. Considerando: a) La implicación cada vez más frecuente de este microorganismo como agente causal de enfermedades intra y extra intestinales, b) El incremento de la comercialización de tilapias silvestres y de cultivo a nivel nacional y c) La necesidad de métodos de detección e identificación rápidos y efectivos de patógenos, se aplicó el método biomolecular de la Reacción en Cadena de la Polimerasa (PCR). Se procesaron mediante la PCR, cepas de P. shigelloides y homogeneizados de pools de bazo, riñón y cerebro, y de intestino de tilapias, utilizándose oligonucleótidos iniciadores específicos para el gen 23S ARNr de P. shigelloides. Se obtuvo el reconocimiento del 75% de las 20 cepas identificadas por pruebas fenotípicas convencionales, detectándose además en 27% de los homogeneizados frescos y en 18% de los congelados. Estos resultados confirman la especificidad del método para la identificación de diversas bacterias potencialmente patógenas, su utilidad en el mejoramiento del sistema de control de calidad de granjas dulceacuícolas y en el estudio de casos clínicos, aumentando la efectividad del sistema de salud relacionado con estas áreas]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[Plesiomonas shigelloides is a Gram negative rod, facultative anaerobic, motile, that belongs to the Enterobacteriaceae. It’s not part of the normal human flora, while fresh water fish like tilapias are thought to be its primary reservoir. Considering: a) The more frequent implication of this microorganism as the causal agent of intra and extraintestinal infections, b) The wide spread of national commercialization of feral and cultured tilapias and c) The need for rapid and effective detection and identification methods, the biomolecular technique of Polymerase Chain Reaction (PCR) was applied. Strains of P. shigelloides and pools of homogenates from spleen, kidney and brain and the intestine of tilapias were processed by PCR, using specific primers for the gene 23S rRNA of P. shigelloides. The 20 strains identified by conventional phenotypic traits were identified in 75%, additionally it was detected in 27% of the fresh homogenates and in 18% of the frozen ones. Su cuenca comprende tierras agropecuarias, residenciales  e industriales, donde se asientan centros urbanos importantes y se desarrollan  actividades económicas de gran crecimiento, tales como industrias, servicios  y agricultura [21]. Estas actividades representan una fuente permanente  de contaminación del lago, al verter en él grandes volúmenes de aguas servidas  que aceleran su degradación [6]. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> En este lago, la tilapia de Mozambique (<I>Oreochromis mossambicus</I>) es la  especie íctica más apreciada por los pescadores, existiendo un comercio  clandestino rentable con respecto a su venta en forma de filetes, distribuidos  éstos en diferentes ciudades del país. Un 95% de los pescadores de la cuenca  extraen la tilapia para fines comerciales y consumo directo [6]. En cuanto  a la producción comercial, cabe destacar que el cultivo de la tilapia roja  (tetrahíbrido de <I>O. mossambicus</I> × <I>O. urolepis hornorum</I> × <I>O. niloticus</I> ×  <I>O. aureus</I>) constituye una importante actividad dentro de la acuicultura  nacional debido, entre otros, a su alta tasa de reproducción en granjas  acuícolas [1]. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Las tilapias son susceptibles a infecciones de etiologías diversas, entre  las que destacan las diferentes especies de bacterias consideradas como  patógenos facultativos u oportunistas, ya que son componentes normales  de la flora de los peces y de su ambiente acuático [10, 14, 22]. Se ha  demostrado experimentalmente que <I>Plesiomonas shigelloides </I>es una de las  especies que está presente en forma abundante en las tilapias del Lago  de Valencia y en el agua y sedimento de su entorno, observándose la misma  tendencia en la microflora de tilapias cultivadas y en su medio ambiente  [2]. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Recientes análisis filogenéticos y perfiles antigénicos indican que existe  una relación estrecha entre <I>P. shigelloides</I> y miembros de la familia Enterobactereaceae,  familia en la cual ha sido ubicada taxonómicamente desde el año 2001 [8,  11, 13]. <I>P. shigelloides </I>es la única especie del género, es un bacilo Gram  negativo, anaerobio facultativo, móvil por presencia de flagelos polares,  que no forma parte de la flora normal de los humanos [11, 13]. Este patógeno  ha sido aislado de heces de pacientes con gastroenteritis relacionadas  con el consumo de ostras y pescado crudo o mal cocido, y ha sido asociado  a diarreas espontáneas e infecciones extra intestinales de elevada mortalidad,  tales como septicemias y meningitis del recién nacido [20]. Aún no han  sido totalmente esclarecidos sus mecanismos de patogenia, atribuyéndosele  en parte a la invasividad, enterotoxigenicidad y asociación a plásmidos  [9]. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Debido a la implicación de <I>P. shigelloides</I> como agente causal de enfermedades  tanto en peces como en humanos [4, 12], es importante contar con métodos  de identificación rápidos y efectivos, que puedan superar en especificidad  y sensibilidad a los métodos de diagnóstico convencionales, permitiendo  mejorar el control sanitario requerido para la comercialización de productos  de consumo humano [19]. En respuesta a esta necesidad, la técnica de la  Reacción en Cadena de la Polimerasa (PCR, por sus siglas en inglés: “Polymerase  Chain Reaction”), ha surgido como una opción confiable en la detección  de diferentes bacterias patógenas, sin que ello implique un desplazamiento  absoluto de los métodos de detección convencionales [16]. </FONT></P>      ]]></body>
<body><![CDATA[<P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> La técnica de la PCR implica un proceso extremadamente sensible; ésta requiere  muy poca cantidad de muestra, permitiendo amplificar y analizar el ADN  contenido en muestras tan variadas como células únicas, folículos pilosos  y sedimentos orgánicos, entre otros. La técnica de la PCR se usa también  para detectar agentes infecciosos, en especial bacterias y virus latentes  [18]. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Considerando: a) La implicación cada vez más frecuente de este microorganismo  como agente causal de enfermedades intra y extra intestinales, b) El incremento  de la comercialización de tilapias silvestres y de cultivo a nivel nacional  y c) La necesidad de métodos de detección e identificación rápidos y efectivos  de patógenos, se aplicó el método biomolecular de la Reacción en Cadena  de la Polimerasa (PCR) para la identificación y detección de cepas de <I>P.  shigelloides</I>. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><B><FONT COLOR="000000" size="2" face="Verdana"> MATERIALES Y MÉTODOS </FONT></B></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> La metodología de la PCR se aplicó a 4 tipos de muestras: </FONT></P>  <UL>     <LI>       <p align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Grupo A: Conformado por las cepas de colección de <I>P. shigelloides</I> del laboratorio  de Microbiología de Peces y Crustáceos [2], descritas en la <a href="#TABLA I"> TABLA I</a>. </FONT></LI>      <LI>       <p align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Grupo B: Sobrenadante y precipitado de homogeneizado fresco del bazo, riñón,  cerebro (BRC) y del intestino de tilapias de Mozambique silvestres. </FONT></LI>      <LI>       <p align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Grupo C: Sobrenadante y precipitado de homogeneizado congelado (a –20°C)  del bazo, riñón y cerebro, y del intestino de tilapias rojas cultivadas  en el Edo. Falcón, a partir de los cuales fueron aisladas cepas de <I>P. shigelloides</I>.  Estas muestras fueron rápidamente descongeladas previo a su utilización  para la extracción del ADN a temperatura ambiente. </FONT></LI>      ]]></body>

<body><![CDATA[<P align="justify" style="word-spacing: 0; line-height: 100%"><B><FONT COLOR="000000" size="2" face="Verdana"> Preparación de homogeneizados de tejido </FONT></B></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> A partir de una muestra de 20 tilapias de Mozambique se formaron grupos  de cinco ejemplares para la preparación de los pools. Previa desinfección  del área con alcohol, se realizó la necropsia siguiendo la técnica indicada  por Amlacher [3]. Una vez expuestos los órganos internos, se procedió a  tomar muestras de éstos para la preparación de dos tipos de pools: uno  de BRC, y otro de intestino. Se maceraron en condiciones asépticas los  pools antes mencionados, tanto de origen fresco como congelado, éstos fueron  mezclados con PBS (buffer fosfatado) estéril pH 7,4 en una proporción 1:5.  Estas mezclas se centrifugaron a 3500 rpm a 4°C. Luego de centrifugadas  las muestras se almacenaron por separado el sobrenadante y el precipitado,  para la extracción del ADN. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><font size="2"><B><FONT COLOR="000000" face="Verdana"> Colonias puras de cepas </FONT></B><FONT COLOR="000000" face="Verdana"> <B>  de </B><I><B>P. shigelloides</B></I> </FONT></font></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> A partir de los macerados se inocularon mediante siembra directa muestras  de los pools en el medio Inositol Bilis Verde Brillante (IBB), medio selectivo  y diferencial para <I>P. shigelloides </I>[11, 13]. Del aislamiento primario se  escogieron las colonias sospechosas de acuerdo a sus características morfológicas  (colonias con tonalidad rosada luego de 48 horas de incubación a 30°C),  las cuales fueron purificadas mediante un pase en agar tripticasa soya.  Luego se les repitieron las pruebas fenotípicas efectuadas por Álvarez  y Agurto [2] siguiendo los esquemas de identificación de Koneman y col.  [15] y Austin y Lee [5], que llevaron a la identificación y confirmación  de la identificación de las cepas aisladas, de las muestras provenientes  del Grupo B y Grupo A, respectivamente. De cada cepa una colonia pura fue  transferida a 10&nbsp;mL de caldo tripticasa soya (CTS) e incubada luego por  18 horas a 35°C. Se utilizó 1,5 mL de este cultivo para la extracción del  ADN. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><B><FONT COLOR="000000" size="2" face="Verdana"> Extracción del ADN </FONT></B></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Basados en el protocolo de González-Rey y cols. [12], el ADN genómico se  extrajo de cultivos en fase logarítmica por solubilidad en fases inmiscibles  por el método CTAB/fenol-cloroformo (CTAB: por sus siglas en inglés “Cetyl  Trimethyl Ammonium Bromide” SIGMA<SUP>®</SUP>). El desarrollo bacteriano en CTS y  los homogeneizados de tejido fueron centrifugados a 16.000 × g por 5 minutos  a 4°C, luego se resuspendió el precipitado de cada uno en 570 µL de buffer  TE (10mM Tris-HCl, 1,0 mM EDTA, pH 8,0), 30 µL de SDS (Dodecil Sulfato  de Sodio SIGMA<SUP>®</SUP>) al 10%, y 3 µL de proteinasa K (20 mg/mL SIGMA<SUP>®</SUP>), y se  incubó durante 90 minutos a 40-42°C. Posteriormente se añadieron 100 µL  de NaCl 5 M y 80 µL de CTAB/NaCl [10% CTAB en 0,7 M de NaCl (SIGMA<SUP>®</SUP>)],  y se incubó por 15 minutos a 65°C. Se agregó 0,8 mL de cloroformo-alcohol  isoamílico (24:1; SIGMA<SUP>®</SUP>) y se centrifugó a 16.000 × g por 5 minutos a  4°C. Se extrajo el sobrenadante y se agregó el mismo volumen de fenol-cloroformo-alcohol  isoamílico (25:24:1; SIGMA<SUP>®</SUP>), luego se centrifugó a 16.000 × g por 5 minutos  a 4°C. El sobrenadante fue transferido a un nuevo tubo Eppendorf y se agregó  400 µL de propanol frío (–20°C), para luego centrifugar a 16.000 × g por  5 minutos a 4°C. El ADN precipitado se lavó con 500 µL de etanol frío (4°C)  y se centrifugó por 10 minutos, para finalmente resuspenderlo en 100 µL  de agua destilada estéril, conservándose la muestra a –20°C según el tiempo  requerido. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Para llevarse a cabo el proceso de amplificación, se verificó que el ADN  extraído tuviese una concentración igual o mayor a 10 ng/µL y un índice  de absorbancia (260/280) entre 1,5 y 2,0 [12, 17]. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><B><FONT COLOR="000000" size="2" face="Verdana"> Aplicación de la PCR </FONT></B></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><font size="2"> <B><FONT COLOR="000000" face="Verdana"> Oligonucleótidos iniciadores utilizados: </FONT></B> <FONT COLOR="000000" face="Verdana">  Las secuencias de los oligonucleótidos  iniciadores fueron tomadas de González-Rey y cols. [11]: sentido (PS23FW3)  5’-CTC CGA ATA CCG TAG AGT GCT ATC C-3’, y antisentido (PS23RV3) 5’-CTC  CCC TAG CCC AAT AAC ACC TAA A-3’. Estos oligonucleótidos amplifican la  región específica comprendida entre los nucleótidos C-906 y G-1189 del  gen 23S ARNr de <I>P. shigelloides</I>. </FONT></font></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><font size="2"> <B><FONT COLOR="000000" face="Verdana"> Amplificación del ADN: </FONT> </B><FONT COLOR="000000" face="Verdana"> El volumen final de la reacción se ajustó a 50 µL,  mezclando 5 µL de TE buffer PCR 10X (Promega), 0,8 µL de una solución 25mM  de dNTPs (SIGMA<SUP>®</SUP>), 8,5&nbsp;µL de cada uno de los oligonucleótidos iniciadores,  5 µL de ADN muestra (10 ng/µL) ó 10 µL de amplicones en caso de aplicarse  un segundo proceso de amplificación, 2 U de Taq ADN Polimerasa (5 U/µL,  Promega), 5 µL de MgCl<SUB>2 </SUB>25 mM (SIGMA<SUP>®</SUP>), completándose el volumen final  de reacción a 50&nbsp;µL con agua libre de nucleasas (SIGMA<SUP>®</SUP>). </FONT></font></P>      ]]></body>
<body><![CDATA[<P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Todas las reacciones de amplificación se realizaron en un termociclador  (Amplitron<SUP>®</SUP> II Thermolyne Barnstead Internacional, E.U.A.) con los siguientes  parámetros cíclicos de temperatura: 1 ciclo a 4°C por 5 minutos y otro  de 95°C por 5 minutos, seguido de 35 ciclos: de 94°C por 1 minuto (desnaturalización),  68°C por 1 minuto (alineación) y 72°C por 1 minuto (extensión), y un paso  de extensión final de 72°C por 10 minutos. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><font size="2"> <B><FONT COLOR="000000" face="Verdana"> Detección del ADN amplificado: </FONT> </B><FONT COLOR="000000" face="Verdana"> Todos los ADN amplificados se separaron  a un voltaje constante de 140 vatios en geles de agarosa (SIGMA<SUP>®</SUP>) al 1,5%  en 1X TAE amortiguador. Los ADN fueron visualizados al ser teñidos con  bromuro de etidio (1 µg/µL SIGMA<SUP>®</SUP>), y se observaron en un transiluminador  de LUV (TFP-M/WL Vilber Lourmat, Francia). </FONT></font></P>     <p align="justify" style="word-spacing: 0; line-height: 100%"><B><FONT COLOR="000000" size="2" face="Verdana"> RESULTADOS Y DISCUSIÓN </FONT></B>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> A partir de los homogeneizados mantenidos a –20°C se detectaron 6 cepas  de <I>P. shigelloides</I>, mientras que de los homogeneizados frescos se reconocieron  9 cepas. En ambos tipos de homogeneizados, se evidenció la presencia de  la bacteria tanto en el intestino como en el pool de BRC, mediante pruebas  fenotípicas convencionales [2]. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> La extracción de ADN a partir de las cepas aisladas de tilapias silvestres  y de cultivo, caracterizadas fenotípicamente como<I> P. shigelloides </I>(cepas  de colección), arrojó rangos de concentración entre 50 y 110 ng/µL y de  índice de presunción de pureza entre 1,6 y 1,9. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Se sometieron al proceso de extracción del ADN tanto los sobrenadantes  como los precipitados de los homogeneizados de BRC, y de intestino, mantenidos  a –20°C y en fresco, obteniéndose los siguientes resultados: </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Al procesar 16 muestras de homogeneizados frescos sólo 11 (68,75%) cumplieron  con los requerimientos de la técnica de la PCR: rangos de 100 a 1.000 ng/µL  de concentración de ADN y una pureza entre 1,8 y 2,0. El porcentaje  promedio de extracción del ADN fue mayor en los precipitados que en los  sobrenadantes. Los tejidos que conforman al bazo, riñón y cerebro en individuos  aparentemente sanos, poseen una diversidad y densidad bacteriana mucho  menor que el intestino, además de que son más fáciles de escindir que el  de intestino, lo que puede ser la causa de que las concentraciones de ADN  de<I> P. shigelloides</I> a partir de homogeneizados de BRC, sean considerablemente  más altas que las de intestino. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Al aplicarse la metodología de la PCR a las 20 cepas de colección tipificadas,  fenotípicamente como <I>P. shigelloides</I> se obtuvo un reconocimiento de 15  de ellas (75%), y ningún reconocimiento al aplicarla sobre las tres cepas  consideradas grupo control negativo (<a href="#TABLA III">TABLA III</a>). Estos resultados demuestran  la especificidad de los oligonucleótidos iniciadores utilizados para la  mayoría de las cepas identificadas, concordando así con los resultados  obtenidos por González-Rey y col. X = No se obtuvo extracción óptima de ADN.</font></p>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> También es de hacer notar, que la literatura señala que los oligonucleótidos  iniciadores utilizados aquí son específicos para <I>P. shigelloides </I>[12],  sin embargo, no se conoce su especificidad para los distintos serotipos  que puedan existir en Venezuela. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Cabe resaltar la detección de <I>P. shigelloides</I> en un precipitado de intestino  a partir del cual no hubo aislamiento previo de la bacteria, lo que indica  que el método es en extremo sensible ya que no requiere grandes cantidades  del ADN a pesquisar, y brinda rapidez en la detección, en comparación con  el aislamiento en medios de cultivo que requiere de estrictas condiciones  de crecimiento, así como de la viabilidad de la bacteria para que se multiplique,  y del reconocimiento de la colonia para que no pase desapercibida en un  medio que contenga colonias de morfología muy similar. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> La aplicación de la técnica de la PCR brinda confiabilidad y rapidez en  la detección de microorganismos bacterianos, sin embargo, la aplicación  de ésta directamente en homogeneizados requiere un buen proceso de maceración  para facilitar su utilización y optimizar los resultados, sea a partir  del sobrenadante o precipitado. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Como resultado de la aplicación de la PCR a las muestras seleccionadas  se obtuvo una banda definida de aproximadamente 284 bp, correspondiente  al gen 23S ARNr determinado para <I>P. shigelloides</I>, coincidiendo con lo reportado  por González-Rey [12] (<a href="#fig1">FIG. 1</a>). </FONT></P>    <P align="center" style="word-spacing: 0; line-height: 100%"><a name="fig1"><img border="0" src="/img/fbpe/rc/v16n5/art02fig1.jpg" width="397" height="441"></a></P>     
<p align="justify" style="word-spacing: 0; line-height: 100%"><B><FONT COLOR="000000" size="2" face="Verdana"> CONCLUSIONES Y RECOMENDACIONES </FONT></B>      ]]></body>
<body><![CDATA[<P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Es importante proseguir con la evaluación de los oligonucleótidos iniciadores,  así como, con la cuantificación de la sensibilidad práctica de la técnica  al utilizar dichos oligonucleótidos en cepas de <I>Plesiomonas shigelloides</I>  aisladas en Venezuela. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Es imprescindible obtener una extracción óptima del ácido nucleico de la  muestra para tener resultados confiables en la aplicación de la técnica  de la PCR, siendo la cuantificación del índice de absorbancia un buen método  de presunción de pureza. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> La aplicación de la técnica de la PCR brinda rapidez en la detección de  <I>P. shigelloides</I>, sin embargo, su utilización directa en homogeneizados  requiere un buen proceso de maceración para optimizar los resultados. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> El aplicar una segunda amplificación en los homogeneizados optimiza la  detección del microorganismo, ya que se cuenta con mayor cantidad de ADN  blanco para emplear nuevamente la técnica. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Si se utiliza la técnica de la PCR a partir de las cepas bacterianas aisladas  en medios diferenciales y selectivos, se optimizará la detección bacteriana,  sobre todo en brotes de enfermedades –bien sea en humanos o animales– donde  se requiera un diagnóstico rápido y confiable, en cuyo caso se obtendrían  resultados en un período de alrededor de 48 horas. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Los resultados obtenidos a partir del procesamiento de muestras congeladas  y frescas, indican diferencias que pueden relacionarse con el efecto de  la temperatura y/o del tiempo de almacenamiento, sobre la integridad del  ADN bacteriano. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><B><FONT COLOR="000000" size="2" face="Verdana"> AGRADECIMIENTO </FONT></B></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><FONT COLOR="000000" size="2" face="Verdana"> Al Instituto Nacional de Investigaciones Agrícolas (INIA) y al Fondo Nacional  de Ciencia Innovación y Tecnología (FONACIT) por el financiamiento de este  Trabajo de Investigación a través de los Proyectos N° 02-613-04001 y Agenda  N° 2000001218, respectivamente, que hicieron posible este Trabajo de Investigación. </FONT></P>      <P align="justify" style="word-spacing: 0; line-height: 100%"><B><FONT COLOR="000000" size="2" face="Verdana"> REFERENCIAS BIBLIOGRÁFICAS </FONT></B></P>      <!-- ref --><P align="justify" style="word-spacing: 0; line-height: 100%"><font face="Verdana" size="2">1.
