En el primero, la actividad de la enzima a-amilasa pancreática fue determinada en muestras de mucosa duodenal, para lo cual se diseñaron 5 tratamientos: ausencia de Con A (T0), presencia de Con A (T1), Con A preincubada durante 30 minutos con la enzima (T2) o con el sustrato (T3) y Caseína (T4). En el segundo experimento se evaluó el efecto de la Con A sobre la actividad de la tripsina en homogenados de páncreas, aplicando 2 tratamientos: presencia de Con A (T0) y ausencia de Con A (T1). La Con A se utilizó a una concentración semejante a la del sustrato correspondiente para cada enzima. Los resultados fueron analizados a través del Análisis de Varianza de Kruskal-Wallis. La Con A inhibió significativamente la actividad específica de la enzima a-amilasa pancreática, tanto en la tercera como en la sexta semana de edad. No hubo diferencia en la actividad de la enzima entre semanas. La preincubación de la lectina con la enzima afectó significativamente la actividad de la a-amilasa pancreática. No hubo efecto de la preincubación de la lectina con el sustrato. La tripsina no fue inhibida por la Con A bajo las condiciones del ensayo, posiblemente asociado al efecto inhibitorio de la actividad biológica que ejerce la caseína sobre la Con A]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[Two trials were conducted to assess the effect of Concanavalin A on pancreatic a-amylase and trypsin activity in broiler chickens (3 and 6 week old). Pancreatic a-amylase activity was determined in mucous duodenal, applying 5 treatments: Control (without Con A, T0), Con A (T1), Con A preincubated during 30 minutes with enzyme (T2), Con A preincubated during 30 minutes with substrate (T3) and Casein (T4). La lectina purificada a partir de semillas  de <I>Phaseolus vulgaris</I> (PHA) presenta una alta resistencia a la proteólisis  digestiva <I>in vivo</I>, manteniéndose completamente activa durante su paso a  través del sistema digestivo en ratones [12]. Estudios con segmentos de  intestino aislados de ratas han demostrado que la PHA interfiere con la  absorción de glucosa [7] y puede unirse a las membranas de las células  epiteliales del intestino delgado, dependiendo del patrón de glicosilación,  provocando severos daños en la mucosa intestinal de ratas [15] y cerdos  [2]. Asimismo, la PHA puede interactuar con enzimas hidrolasas del borde  en cepillo afectando su actividad [8]. Recientemente se ha comprobado que  la exposición durante 3 días a la lectina PHA administrada en la dieta  induce cambios en la maduración de las proteínas integrales de la membrana  del enterocito en ratas jóvenes [14] pudiendo ser ésta una de las causas  por las cuales es afectada la actividad de algunas enzimas digestivas.  Al igual que la PHA, la lectina del frijol rojo (RKB) disminuyó significativamente  las actividades de las enzimas a-amilasa pancreática y a-amilasa salival  reduciendo la tasa de digestibilidad <I>in vitro</I> del almidón de trigo 27. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Al ser suministrada en la dieta, cantidades significativas de Concanavalina  A, la lectina presente en la <I>Canavalia ensiformis, </I>pudo ser recuperada  en las heces de ratas que recibieron una dosis única de 40 mg de Con A<I>.</I>  La recuperación de la lectina en las heces durante 4 días fue progresiva  hasta alcanzar el 90% del total de la lectina suministrada, demostrándose  que ésta, al igual que la PHA y otras lectinas, son en alto grado resistentes  a la acción de las enzimas digestivas durante su tránsito a través del  tracto gastrointestinal [18]. Si en el tracto digestivo de los animales  las lectinas permanecen activas, ellas podrían reaccionar con algunos componentes  de la digesta, secreciones, glicoconjugados bacterianos o dietarios y/o  con las membranas de las células epiteliales, dependiendo de su especificidad  de unión a carbohidratos, e interferir así con el normal funcionamiento  del sistema digestivo [21]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Al llegar al lumen intestinal, la Con A puede interactuar con la membrana  de la mucosa afectando los procesos que suceden a este nivel, tales como  mecanismos de transporte e hidrólisis final de moléculas provenientes de  la dieta [10]. En ratas se afectó la absorción de algunos nutrientes como  el calcio y la sacarosa [1] y disminuyó significativamente la actividad  de las enzimas sacarasa, fosfatasa alcalina y leucina aminopeptidasa [18]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El objetivo del presente trabajo fue determinar el efecto de la lectina  Concanavalina A (Con A) sobre la actividad de las enzimas digestivas  pancreáticas  </FONT><FONT COLOR="000000" SIZE="2" FACE="Symbol"> a</FONT><FONT COLOR="000000" SIZE="2" FACE="Verdana">-amilasa y tripsina en  pollos de engorde de 3 y 6 semanas de edad.  </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>MATERIALES Y MÉTODOS</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Las semillas de <I>Canavalia ensiformis</I>, variedad Tovar, fueron suministradas  por el laboratorio de Genética Molecular de la Facultad de Agronomía de  la Universidad Central de Venezuela (UCV). </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El aislamiento de la Con A y las determinaciones enzimáticas se llevaron  a cabo en el laboratorio de Enzimología y Toxicología y en el Centro de  Bioquímica Nutricional, de la Facultad de Ciencias Veterinarias de la UCV. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Los análisis de electroforesis fueron realizados en el laboratorio de Genética  Molecular de la Facultad de Agronomía de la UCV. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Reactivos</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Los reactivos utilizados fueron adquiridos de Sigma. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Aislamiento y Purificación de la Concanavalina A</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El aislamiento y purificación de la Con A se realizó mediante la precipitación  selectiva con sulfato de amonio y cromatografía de afinidad [9], liofilizada  en un equipo Labconco LYPH-Lock 6, EUA, y almacenada a -20°C hasta su utilización. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Determinación de la pureza de la Concanavalina A</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La pureza de la Con A fue establecida por SDS-PAGE al 15% comparando con  un patrón comercial de Con A marca Sigma y confirmada por hemaglutinación,  al medir la capacidad de producir aglutinación de una suspensión de eritrocitos  frescos de conejo al 2% en NaCl 0,9% después de 60 minutos de incubación  a temperatura ambiente y comparada con la producida por la Con A, adquirida  comercialmente de Sigma. A fin de comparar la capacidad hemaglutinante  de la Con A en solución salina fisiológica y en presencia de las soluciones  amortiguadoras utilizadas en las determinaciones de las correspondientes  enzimas, se determinó la capacidad hemaglutinante de la Con A en presencia  de 10 mg/mL de caseína y en presencia de Tris-HCl 160 mM a pH 7,5. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Manejo de los animales y dieta</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Pollos de un día de edad, de la línea comercial Ross, adquiridos en la  granja avícola “La Caridad”, estado Aragua, Venezuela, fueron alojados  en jaulas metálicas colectivas, con suministro de agua y alimento concentrado  comercial <I>ad libitum, </I>para suplir todos sus requerimientos nutricionales  [20]. A las 3 y 6 semanas de edad, un total de 30 pollos/edad, fueron sometidos  a ayuno durante 12 horas y sacrificados por dislocación cervical. Segmentos  duodenales y el páncreas fueron removidos rápidamente, sumergidos en solución  salina fisiológica a 0 -4°C y procesados de inmediato para obtener el homogenado  respectivo. </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Preparación del homogenado del páncreas</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Todo el procedimiento se realizó entre 0 y 4°C. Para formar cada muestra,  el páncreas de 5 pollos fue homogeneizado empleando un homogeneizador de  tejido, marca Elvehjem, EUA, a máxima velocidad por 3 minutos en 4 mL/g  de solución salina fisiológica, filtrado a través de un liencillo y el  extracto almacenado en alícuotas a -20°C hasta su uso para las determinaciones  enzimáticas, en un período no mayor de 15 días en ambos casos a fin de  evitar pérdida de actividad [5]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Preparación del homogenado de la mucosa duodenal</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Todo el procedimiento se realizó entre 0 y 4°C. Los segmentos duodenales  de 5 pollos para cada muestra, separados de acuerdo a Siddons [24], fueron  perfundidos con solución salina fisiológica, cortados longitudinalmente  y colocados sobre una placa de vidrio para extraer la mucosa, empleando  una lámina de vidrio. La mucosa fue sumergida en 20 mL/g de un buffer conteniendo  50 mM de manitol y 2 mM de HEPES a pH 7,1 y homogeneizada a máxima velocidad  por 1 minuto empleando un homogeneizador de tejido, marca Elvehjem, EUA.  Se almacenaron alícuotas de los homogenados respectivos a -20°C con fines  de determinaciones enzimáticas. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Determinaciones Enzimáticas</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" FACE="Symbol" SIZE="2">a</FONT><FONT COLOR="000000" FACE="Verdana" SIZE="2">-<I>Amilasa:</I> La actividad de la enzima a-amilasa pancreática (EC 3.2.1.1)  se determinó en muestras de homogenado de la mucosa duodenal [23]. Brevemente,  el homogenado de la mucosa duodenal (50 µL) fue incubado durante 15 minutos  a 41°C en una mezcla de reacción conteniendo 100 µL de buffer Tris-HCl  160 mM a pH 7,5 y 200 µL de almidón al 0,15% en ácido sórbico saturado,  empleado como sustrato. La reacción se detuvo agregando 100 µL de HCl 0,5  N, añadiéndose seguidamente 2 mL de agua destilada y 100 µL de solución  de yodo 0,01 N. La cantidad de almidón degradado por la amilasa se determinó  por espectrofotometría, midiendo la disminución de la coloración azul del  complejo iónico almidón-yoduro a 620 nm. Los valores obtenidos fueron comparados  con los de una curva estándar elaborada a partir de una solución de almidón  al 0,15%. La actividad de la a-amilasa pancreática se expresó como ìg de  almidón degradado/min/mg de proteína [23]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <I>Tripsina</I>: La actividad de la enzima tripsina (EC 3.4.21.4) se determinó  en muestras de homogenado de páncreas 13, para lo cual 100 µL del homogenado  fue incubado durante 15 minutos a 41°C con 200 µL del sustrato caseína  al 1% en buffer fosfato de sodio 0,1M, pH 7,4. La reacción se detuvo con  la adición de 1 mL de ácido tricloroacético al 5% p/v y la mezcla fue filtrada  en papel Whatman 42. Se hicieron reaccionar 250 µL del filtrado con 1250  µL de Na<SUB>2</SUB>CO<SUB>3</SUB> 0,2 M y 250 µL del reactivo de Folin-Ciocalteu, diluido 1<B>:</B>5  en agua destilada. Después de una incubación durante 20 minutos a 41°C  se midió la absorbancia a 625 nm. La actividad de la tripsina se expresó  como Unidades de Actividad Proteolítica (UAP) calculada a partir de la  ecuación propuesta por Namoto y Narahashi [19]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Todos los análisis enzimáticos se realizaron por triplicado y en todos  los casos se elaboraron blancos con igual procedimiento, en los cuales  las muestras fueron añadidas después de detener la reacción. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Proteína</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La concentración de proteína fue determinada por el método de Bradford  3 utilizando el buffer correspondiente como blanco y albúmina sérica bovina  como estándar. </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Descripción de los experimentos</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Experimento 1:</B> Con el objetivo de determinar el efecto de la Con A sobre  la actividad de la enzima </FONT><FONT COLOR="000000" SIZE="2" FACE="Symbol"> a</FONT><FONT COLOR="000000" SIZE="2" FACE="Verdana">-amilasa pancreática y la posible interacción  de la lectina con la enzima o el sustrato correspondiente, se aplicaron  5 tratamientos: ausencia de Con A (T0), presencia de Con A (T1), Con A  preincubada durante 30 minutos con la enzima antes de iniciar la reacción  (T2), Con A preincubada durante 30 minutos con el sustrato antes de iniciar  la reacción (T3) y presencia de caseína (T4). Todos los tratamientos se  aplicaron <I>in vitro</I> en muestras de homogenado duodenal de pollos de 3 y  6 semanas de edad. La actividad de la enzima a-amilasa y la concentración  de proteínas fueron determinadas para los diferentes tratamientos y edades  de los pollos. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Experimento 2: </B>El efecto de la Con A sobre la actividad de la tripsina  se estudió en muestras de homogenado de páncreas de pollos de 3 y 6 semanas  de edad, aplicando 2 tratamientos: ausencia de Con A (T0) y presencia de  Con A (T1). Al igual que en experimento 1, se realizaron determinaciones  de las actividades de la enzima tripsina y concentración de proteínas en  los homogenados respectivos. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> En todos los casos, las soluciones de Con A fueron preparadas en NaCl 0,9%.  La Con A se utilizó en una concentración semejante a la del sustrato correspondiente  para cada enzima (1,5 mg/mL para a-amilasa y 10 mg/mL para tripsina). Todas  las determinaciones se realizaron por triplicado. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Análisis Estadístico</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Los resultados fueron expresados como el promedio ± la desviación estándar  y analizados estadísticamente a través del Análisis de Varianza de Kruskal-Wallis  y la prueba de rangos promedios a un nivel de significancia de 0,05 [25],  utilizando el programa Statistix, versión 8 [26]. El uso de estadísticas  no paramétricas se debió al hecho de que la distribución probabilística  de los valores no se ajustó a una distribución normal [25]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>RESULTADOS Y DISCUSIÓN</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Pureza de la Con A</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La <a href="#fig1">FIG.1</a> muestra el análisis de SDS-PAGE de la Con A aislada, purificada  y liofilizada en comparación con la Con A utilizada como patrón (Sigma,  St. Louis, EUA). En ambos casos se observa claramente una banda bien definida  que se corresponde al valor de 30 kDa de la Con A monomérica. Así mismo  se observan dos pequeñas franjas de un PM aproximado a 20 y 24 kDa, correspondientes  a polipéptidos de Con A, tal como fue encontrado por otros autores [4,  11]. </FONT></P>      <P ALIGN="justify"><a name="fig1"></a></P>      ]]></body>

<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Estos resultados demuestran que la Con A purificada en el presente trabajo  se encuentra libre de contaminación por otras fracciones proteínicas y  su pureza es similar a la Con A comercial, garantizando que los resultados  obtenidos son debidos a la presencia de la lectina, descartándose la interferencia  de otros posibles factores. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>Título hemaglutinante de la Con A</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El título hemaglutinante de la Con A aislada, expresado como porcentaje  del total de hemaglutinación, fue semejante al de la Con A comercial (<a href="#tab1">Tabla  I</a>). No se manifestó actividad hemaglutinante de la Con A en presencia de  caseína al 1%, posiblemente debido a que esta última es una glicoproteína  cuyos residuos de carbohidratos pueden ser reconocidos por los sitios activos  de la lectina e impedir su acción hemaglutinante. Este efecto puede ser  de gran aplicación práctica al elaborar dietas con materias primas alternativas  que contengan esta clase de lectina, ya que al adicionar caseína en la  ración puede neutralizarse su efecto anti nutricional. Para cada tratamiento se  realizaron 18 repeticiones. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La actividad de la enzima a-amilasa pancreática, determinada en el homogenado  de mucosa duodenal, tanto a la tercera como a la sexta semana de edad,  fue significativamente menor en relación con el control (P &lt; 0,05), tanto  en presencia de Con A (T1) como al preincubar la enzima con la lectina  (T2), con reducciones de 49,2 y 41,5% en la tercera semana y 43,4 y 44,2%  en la sexta semana. La disminución de la actividad de la amilasa pancreática  al preincubarla en presencia de Con A durante 30 minutos, sugiere que la  lectina puede unirse a la enzima, por ser ésta una glicoproteína que contiene  residuos de glucosa. Al preincubar la Con A con el almidón, sustrato de  la enzima amilasa pancreática, a la tercera semana de edad, no se observó  el efecto inhibitorio de la Con A sobre la enzima; no obstante, en las  muestras de la sexta semana, se observó una inhibición de la actividad  enzimática. Ha sido demostrado que la Con A se une a los residuos de carbohidratos  del epitelio del intestino delgado [22]. Estos azúcares afines con la Con  A forman parte de la estructura de las mucinas secretadas por el epitelio  intestinal [26]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Por otra parte, estudios previos han revelados cambios con la edad del  patrón de secreción de estas mucosustancias del tracto intestinal de las  aves [6], observándose una disminución de la secreción de estas mucinas  a partir de la tercera semana de edad [16]. Esta característica de las  aves pudiera explicar el efecto inhibitorio de la Con A sobre la amilasa  a la sexta semana de edad, tomando en cuenta que a esa edad hay una disminución  significativa de las mucinas en el intestino y por lo tanto menos residuos  de oligosacáridos que ocupen los sitios de unión de la Con A pudiendo existir  más moléculas de Con A en forma libre capaces de unirse a la enzima y ejercer  su acción inhibitoria. La inclusión de caseína en el medio de reacción,  no tuvo efecto sobre la actividad de la a-amilasa pancreática. Resultados  semejantes fueron obtenidos al determinar el efecto de la Con A y de la  lectina del frijol rojo (LFR) sobre la actividad <I>in vitro</I> de la amilasa  pancreática porcina y amilasa salival humana, reportando una disminución  significativa en las actividades enzimáticas, como consecuencia de una  inhibición no competitiva de ambas lectinas sobre ambas enzimas, lo que  incide negativamente en la digestión de los carbohidratos de la dieta [28]. </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El efecto de la Con A sobre la actividad específica de la tripsina en homogenados  de páncreas de pollos de 3 y 6 semanas de edad se presenta en la <a href="#tab3">Tabla III</a>. No se encontraron diferencias entre los tratamientos ni entre semanas.
