Dieciocho hembras (7 meses de edad) fueron distribuidas en 3 grupos: TA:1 kg alimento 15% PC+Yea Sacc®, (levadura Saccharomyces cerevisiae cepa 1026). Dosis: 5 gr/animal/día+heno ad libitum. TB:1 kg alimento 15% PC+Bioplex Plus®, (cultivo de levaduras vivas y minerales orgánicos). Dosis: 4 gr/animal/día+heno ad libitum. TT:Testigo. 1 kg alimento 15% de PC+heno ad libitum. El estro se sincronizó con esponjas vaginales (40 mg FGA) durante 12d. A los 10d de la inserción de las esponjas se aplicó 225 mg de prostaglandina/oveja. La superovulación se realizó 36h antes de la retirada de las esponjas mediante 100 mg-NIH FSHp y 500 UI eCG en una sola dosis/oveja. 24h de la retirada de las esponjas se aplicaron 100 µg GnRH/oveja. La tasa de ovulación se midió por endoscopia. Los ovocitos se recuperaron del oviducto 48h de la retirada de las esponjas. La TO resultó superior (P<0,05) en los TA y TB (16,6±6,84 y 15,5±4,18), respecto al TT (9,16±3,81). La tasa de recuperación (TR) muestra diferencias (P<0,05) entre el TA (93,9%) respecto a los TB (56,9%) y TT (61,8%). La proporción (P<0,05) de ovocitos de calidad 1 en el TA respecto a los TB y TT fue mayor. En conclusión, la adición de levaduras en la dieta favorece la respuesta a un tratamiento de estímulo superovulatorio en ovejas púberes incrementando la tasa de ovulación y calidad de los ovocitos]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[The superovulatory response of pre-pubertal or pubertal ewes fed with protein suplement that contained yeast or organic minerals was evaluated in order to obtain “in vivo” maturated oocytes. Eighteen females (7 months of age) were allocated to one of the following treatment and fed as follow: Treatment A (TA): 1 kg food with 15% PC+Yea sacc® (yeast Saccharomyces cerevisiae stump 1026), dose: 5 gr/animal/day and hay ad libitum. Treatment B (TB): 1 kg food with 15% PC+Bioplex Plus® (Cultivation of live yeasts and organic minerals), dose: 4 gr/animal/day and hay ad libitum. Treatment C (TT: Control): 1 kg food with 15% PC and hay ad libitum. The estrous cycle was synchronized through the insertion of vaginal sponges with 40 mg of fluorogestone acetate (FGA) during 12 days. At day 10th of the sponges insertion 225 mg of prostaglandin F2á was administered per ewe. To induce supra-ovulation 100 mg-NIH FSHp and 500 UI eCG in a single dose were applied 36 h before the withdrawal of the sponges, and 24 h from the last application 100 µg of GnRH was also injected. The ovulation rate was measured by endoscopy and ovulated oocytes collected by flushing. The total of ovulation (TO) was greater in the TA and TB (16.6±6.84 and 15.5±4.18) treatment, than on the control TT (9.16±3.81) (P<0.05). The oocyte recovery (TR) after flashing was significantly higher in the TA (93.9%) treatment compared to TB (56.9%) and TT (61.8%). Finally, the proportion (P<0.05) of grade I oocytes in the diet supplementation in TA was in higher than the quality of oocytes in the TB and TT treatment. Paralelo a ello, el balance nutricional desempeña un papel importante a  la hora de obtener resultados adecuados [5]. En este sentido, algunos trabajos  con ovejas adultas suplementadas con alimento de alto valor proteico, han  demostrado que las características de la degradación proteica de los nutrientes  de un alimento, pueden ejercer un efecto favorable o detrimental sobre  el desarrollo embrionario temprano [24]. Así mismo, es conocida la acción  benéfica sobre el ovario, cuando se suplementa con aceites vegetales ricos  en ácidos grasos poli-insaturados [39], donde un aumento de energía y proteína  de la dieta permite un incremento en el número y diámetro de los folículos  preantrales [12] y, una mejor calidad y desarrollo de los ovocitos [27].  Por el contrario, un exceso de energía y proteína puede reducir la respuesta  a un estimulo superovulatorio [5]. Evidentemente, en todos los casos se  refleja que la relación nutrición-reproducción debe guardar cierto equilibrio.  Más aún, tratándose de rumiantes donde la microflora y fauna son inestables,  dependiendo sobre todo del tipo y calidad de los alimentos [18]. En este  sentido, la suplementación de la dieta con levaduras y minerales orgánicos,  además de modular los eventos de la digestión ruminal [20] incrementa la  fertilidad y disminuye la mortalidad embrionaria [4]. Ciertas variedades  y combinaciones microbianas para alimentación directa benefician el rendimiento  y la salud de los animales. Estos elementos incrementan el consumo de alimento,  la ganancia de peso corporal y la eficiencia del alimento, a través de  un balance microbiano saludable en el tracto digestivo [15,29], aunque  otros indican que no existe tal efecto [17]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La aplicación práctica de la fertilización <I>in vitro</I> (FIV), requiere que  los óvulos provengan de animales de alto valor económico, genético y de  historial sanitario conocido [2], La técnica de recuperación de ovocitos  por aspiración laparoscópica directa en los ovarios [3,32,36] es una manera  de recolección eficiente de ovocitos procedentes de animales vivos. Aunque,  también puede utilizarse ovocitos recuperados de ovarios de matadero y  en ambos casos se requiere de un cultivo “<I>in vitro</I>” para su uso inmediato  en programas de FIV. Para ello, la posibilidad de obtener ovocitos en ovejas  púberes mediante la recolección de ovocitos madurados <I>in vivo</I> y recolectados  en el oviducto después de la ovulación [33] abre la posibilidad de mejorar  los resultados hasta ahora obtenidos con ovocitos madurados “<I>in vitro</I>”  [8], ya que la ventaja de obtener ovocitos <I>in vivo</I> es que son cultivados  en las primeras etapas en un medio ambiente oviductual, por el contrario,  los ovocitos <I>in vitro</I> requieren de un cultivo artificial. El objetivo de  este proyecto fue medir la respuesta ovárica de ovejas púberes suplementadas  con levaduras y/o minerales orgánicos con el fin de producir ovocitos maduros  “<I>in vivo</I>” para su posterior conservación mediante vitrificación. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>MATERIALES Y MÉTODOS</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> El trabajo se realizó en enero-febrero de 2004, en el Centro de Selección  y Reproducción Ovina (CeSyRO) del Instituto Tecnológico Agropecuario #  2 de Conkal, Yucatán (21°02´ longitud Norte y 89°29´ longitud Oeste) con  clima tropical subhúmedo (Awo), con temperatura media anual de 26,5°C,  precipitación total de 900 mm, y 9 msnm [11]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Se utilizaron 18 ovejas púberes de raza Santa Cruz, de 7 meses de edad,  peso vivo de 32,1±4,3 Kg y una condición corporal de 3 [31], con buen estado  sanitario. Fueron distribuidas en 3 tratamientos (n= 6) de acuerdo a su  peso. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Todas las ovejas fueron mantenidas en estabulación completa y se desparasitaron  con Albendazol micronizado (OVERSOL; Lab. Ovejero de México, S.A. de C.V,  España). </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Tuvieron a su disposición heno de pasto estrella (<I>Cynodon nlemfuensis</I>)  a libre acceso y se les proporcionó un kilogramo de alimento comercial  (Nutrimyn; MyN Distribuidora, S.A. de C.V., México) para ovinos con 15%  de PC, a base de: Sorgo, pastas de soya y canola, cascarilla de soya, sal,  carbonato de calcio, fosfato de calcio, melaza, urea, bicarbonato de sodio,  cloruro de potasio, oxido de magnesio, minerales traza, vitaminas A, D3  y E, azufre, minerales quelatados (Zn). </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Los tratamientos consistieron en: </FONT></P>  <UL>     <LI>     <p align="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> TA: Levaduras (Yea Sacc<SUP>®</SUP> - Alltech). Dosis: 5 gr/animal/día. </FONT></LI>      <LI>     <p align="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> TB: Levaduras + Minerales Orgánicos (Bioplex Plus<SUP>®</SUP> - Alltech). Dosis: 4  gr/animal/día. </FONT></LI>      <LI>     <p align="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> TT: Grupo Control. </FONT></LI>      </UL>     <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La duración del trabajo de campo fue de 25 días. </FONT></P>      ]]></body>
<body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La sincronización e inducción del estro se realizó mediante la inserción  de esponjas vaginales impregnadas con 40 mg de Acetato de Fluorogestona  (FGA; CHRONOGEST<SUP>TM</SUP>; Intervet, México) ocho días después de iniciado el  manejo alimenticio, y se mantuvieron 12 días. A los 10 días de la inserción  de las esponjas se les aplicó 225 mg de prostaglandina (PGF<SUB>2</SUB>Ü; ILIREN<SUP>®</SUP>,  Intervet, México). Para inducir la superovulación, 36 horas antes de la  retirada de las esponjas, se aplicó en dosis única vía intramuscular un  compuesto a base de 100 mg-NIH de Hormona Folículo Estimulante Porcina  (FSHp; FOLTROPIN-V<SUP>®</SUP>; Vetrepharm Inc. Canada) y 500 UI de Gonadotropina  Coriónica Equina (eCG; FOLLIGON<SUP>TM</SUP>; Intervet, México). Para sincronizar  e inducir la ovulación se les aplicó una dosis de 100 µg de Hormona liberadora  de las Gonadotropinas (GnRH; FERTAGYL<SUP>®</SUP>; Intervet, México) a las 24 horas  después de la retirada de las esponjas. La respuesta al tratamiento se  midió por endoscopia [25] a través del conteo de cuerpos lúteos presentes  en el ovario (TO: Tasa de Ovulación) y la calidad del cuerpo lúteo (color,  forma y presencia/ausencia de fosa de ovulación). Los ovocitos se recuperaron  por perfusión directa del oviducto [30] con 10 ml de PBS (BIOLIFE<SUP>TM</SUP> Advantage;  Agtech, Inc) a las 48 horas de la retirada de las esponjas. Posteriormente  se valoraron en base al aspecto morfológico y desarrollo ovocitario, y  se clasificaron en cuatro categorías: Excelente (Calidad 1: C1), Bueno  (Calidad 2: C2), Regular (Calidad 3: C3) y no fertilizados (Calidad 4:  C4) [28], siendo vitrificados en soluciones que contenía: 10% de Glicerol  (G) durante 5 minutos, 15% G + 20% de Etilenglicol (EG) durante 5 minutos,  25% G + 25% EG durante 30 segundos y posteriormente las pajuelas se expusieron  a vapores de nitrógeno durante 20 segundos, sumergidas directamente en  el nitrógeno [14, 22] y almacenados en nitrógeno líquido. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> Los resultados obtenidos se analizaron estadísticamente mediante la prueba  Ji-cuadrado del programa estadístico Statistix [34]. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> <B>RESULTADOS Y DISCUSIÓN</B> </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La administración oral a ovejas púberes de levaduras y minerales orgánicos  mejoró la tasa ovulatoria (16,6 ± 6,84, 15,5 ± 4,18) (9,16 ± 3,81); con  respecto al grupo de control (P&lt;0,05. <a href="#tab1">TABLA I</a>). <body><![CDATA[<P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> ** Una hembra de este grupo resulto muerta al los 10d de iniciado el experimento.</FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> (<SUP>a,b</SUP>) Letras diferentes en fila indican diferencia significativa (P&lt;0,05). </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> De igual forma, el uso de levaduras mejoró la tasa de ovulación con respecto  a protocolos de estímulo superovulatorio múltiples, previamente reportados.  En esos estudios, la respuesta se situó en valores que varían entre 6,8  ±3,91 (ovejas Ile de France) [37], 9,8 ± 1,05 a 11,5 ± 5,04 ovejas Pelibuey  [26, 38]. Aunque se debe tener en cuenta las diferencias entre razas y  condiciones ambientales a las cuales fueron sometidas las ovejas, los resultados  sugieren que la administración de levaduras en la dieta favorece el nivel  de respuesta superovulatoria a través de un incremento de la TO. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> La tasa de recuperación (TR) de ovocitos fue mayor en el tratamiento TA  (93,9%) y significativamente diferente a los tratamientos TA y TT (56,9  y 61,8%, respectivamente, P&lt;0,05. <a href="#tab1">TABLA I</a>). Aunque el número de ovocitos  recuperados a nivel de oviducto resulta satisfactorio, y similar al reportado  por otros investigadores utilizando técnicas similares (83,2%) Folch y  col [7]; (57,4%): Alwan y col [1], no es posible explicar las diferencias  obtenidas entre los tratamientos. Por lo tanto, futuros estudios se deben  realizar para entender si existen diferencias fisiológicas y/o técnicas  en la colección de ovocitos. </FONT></P>      <P ALIGN="justify"><FONT COLOR="000000" SIZE="2" FACE="Verdana"> En la <a href="#tab2">TABLA II</a> se presentan los resultados del número de ovocitos clasificados  de acuerdo a su morfología. La mayoría de los ovocitos colectados en los  diferentes tratamientos fueron clasificados como calidad 1 (C1) y en una  menor proporción, los de calidad 2 (C2) y 3 (C3). Si bien es conocido que  los ovocitos están en metafase II cuando ocurre la ovulación [6], éstos  son captados por la fimbria que rodea al ovario siendo conducidos hacia  el oviducto. Muchos investigadores coinciden en que en el oviducto se expresan  componentes macromoleculares específicos que interactúan con los gametos  [35], favoreciendo su maduración a fin de estar capacitados para su fecundación  [9]. La calidad del ovocito maduro <I>in vivo</I> resulta de especial interés,  dado que el ovocito en esta fase, prácticamente es seleccionado naturalmente  por factores de crecimiento tales como las IGFs (Insulin like growth factors).  El modo de acción autocrino y paracrino de ellas sobre el medio ambiente  uterino, es mediado por el tipo de receptor uterino, ya sea para IGF-I  o IGF-BP (IGF-binding protein) [23]. Sin embargo, el valor y la calidad  de la dieta suelen influir en la secreción de estos factores, ya sea a  través de la secreción endógena de la hormona de crecimiento (GH), niveles  de insulina [16] y energía [19] que parecen influir sobre la calidad del  ovocito, su capacidad para ser fecundado y su posterior desarrollo embrionario  [9].
