El parásito posee glicoproteínas y enzimas que facilitan su penetración y migración en los tejidos hospederos. El objetivo fue evaluar estos componentes en extractos [fracción soluble (FS) y fracción particulada (FP)] de vermes adultos de F. hepatica. Las fracciones fueron analizadas para proteínas, carbohidratos, actividades enzimáticas, y por electroforesis (SDS-PAGE-15%) en condiciones no reductoras (NR) y reductoras (R). En FS, la concentración de carbohidratos fue 5,63 μmol/mL; se detectó la presencia de fosfohidrolasas, glicosidasas y peptidasas, sobresaliendo la actividad Fosfatasa Acida (1,19 μmol/h/mg, pH 5,0). En FP, la concentración de carbohidratos fue más elevada (7,89 μmol/mL); se observaron valores elevados de Fosfatasa Alcalina (5,79 μmol/h/mg), Fosfatasa Acida (1,67 μmol/h/mg) y moderados de Fosfodiesterasa (0,280 μmol/h/mg pH 9,6). Se detectó actividad Acetilcolinesterasa mayor en FS que en FP (3,95 A/h vs. 1,68 A/h). En SDS-PAGE ambos extractos mostraron polipéptidos de ~80 a 10-kDa; el pre-tratamiento de las muestras con sodio-metaperiodato redujo el número de bandas en cada extracto, sugiriendo la presencia de glicocomponentes. En conclusión, se hallaron enzimas y glicocomponentes en los extractos de F. hepatica estudiados, lo cual constituye un aporte en la búsqueda de posibles blancos inmunológicos y farmacológicos para el control de esta parasitosis.<hr/>Fasciola hepatica is a trematode that causes damage to the liver and biliary tract in both herbivores and man, and is thus an important public health concern in agricultural areas worldwide. This parasite contains glycoproteins and enzymes that facilitate its penetration of, and migration through, host tissues. The aim of this study was to evaluate the relative proportions of these components in the soluble fraction (SF) and n-butanol-solubilized extracts of the particulate fraction (PF) of whole homogenates of adult F. hepatica worms. Extracts were analyzed for protein and carbohydrate content, and hydrolytic activity. In addition, 15% SDS-PAGE electrophoresis was performed under non-reducing (NR) and reducing (R) conditions to identify other components. The SF contained 5.63 μmol carbohydrate/mL, as well as phosphohydrolases, glycosidases and peptidases, with a high acid phosphatase activity (1.19 μmol/h/mg, pH 5.0) that seemed to be characteristic of this fraction. The PF contained 7.89 μmol carbohydrate/mL, with high alkaline phosphatase (5.79 μmol/h/mg) and acid phosphatase (1.67 μmol/h/mg), and moderate phosphodiesterase (0.280 μmol/h/mg pH 9.6) values. Acetylcholinesterase activity was higher in the SF than the PF (3.95 A/h vs. 1.68 A/h). SDS-PAGE analysis showed the presence of polypeptides in both the SF and the PF of ~80 to 10-kDa . Pretreatment with sodium-metaperiodate reduced the number of bands in each extract suggesting the presence of glycocomponents. The notable presence of enzymes and glycocomponents in the SF and PF extracts of adult F. hepatica worms is a first step towards the eventual identification of new immunological and pharmacological targets for the control of this disease. <![CDATA[<b>Inflammatory cells in the nasal secretion and serum pro-inflammatory Th1, Th2, Th17 and regulatory cytokines from American cutaneous leishmaniasis patients</b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1690-46482014000200003&lng=pt&nrm=iso&tlng=pt En la forma mucocutánea (LCM) y cutánea (LCL) de la leishmaniasis, se genera una respuesta inflamatoria cuyos mediadores (células y citocinas) se han involucrado en la severidad de las úlceras y en el daño tisular observado en estos pacientes, particularmente en los LCM. Por ello, nos propusimos identificar los grupos celulares predominantes en la secreción nasal de pacientes con LCL y LCM, y relacionarlos con citocinas proinflamatorias y reguladoras. Evaluamos en pacientes LCL (n=20), LCM (n=14) y 20 individuos sanos: a) La cuantificación de tipos de leucocitos en "frotis" de secreción nasal, úlceras cutáneas y sangre periférica teñidos con Giemsa empleando microscopía óptica, b) Concentraciones séricas de IL-8, IL-4 e IL-10 por citometría de flujo (CBA array) e IFN-γ, TNF-α e IL-17 por ELISA. El grupo celular predominante en la secreción nasal de pacientes con LCM fueron los neutrófilos (80,7%) y escasos eosinófilos (0,6%), comparados con los LCL y controles, en los que no se observaron estas células. Mientras que los "frotis" de las ulceras de los LCL presentaron 45,3% de neutrófilos y 43% de linfocitos. En contraste, en sangre periférica, de los pacientes se observó un incremento de neutrófilos y linfocitos junto a una frecuencia significativa de monocitos (LCM: 5,3; LCL: 6,3%) y eosinófilos (LCM: 8,2%; LCL: 5,2%). Todo esto sugiere la participación de los neutrófilos en la inmunopatogénesis en la LCM. Adicionalmente, se demostró una mayor (P=0,03) concentración sérica de IL-8 en los pacientes con LCL (18,5ρg/mL) y LCM (18,2ρg/mL) respecto a los individuos sanos, sugiriendo que esta citocina promueve el reclutamiento de neutrófilos al sitio de infección en los LCM, mientras que en los LCL contribuyen junto con los linfocitos T CD4+ de la subpoblación Th1 y productores de IFN-γ, en la activación de mecanismos leishmanicidas.<hr/>In mucocutaneous (MCL) and cutaneous (LCL) leishmaniasis, the inflammatory mediators (cytokines and cells) have been associated with ulcers severity and tissue damage observed in these patients, particularly in MCL. Therefore, we decided to identify the predominant cell groups in the nasal secretion of LCL and MCL patients, and related pro-inflammatory and regulatory cytokines. It was evaluated in LCL (n = 20), MCL patients (n = 14) and 20 healthy volunteers: a) Differential leukocyte count by optical microscopy performed in: smear of a runny nose, skin ulcers and peripheral blood dyed with Giemsa, b) serum levels of IL-8, IL-4 and IL-10 using cytometric bead array (CBA) assay and IFN-γ, TNF-α and IL-17 by ELISA. In MCL patients, neutrophils (80.7%) were the most abundant cellular group in nose secretion, followed by a small amount of eosinophils (0.6%) compared to the LCL and controls, where no such cells were observed. In contrast, in peripheral blood from ACL patients were observed an abundant amount of neutrophils and lymphocytes together with a significant frequency of monocytes (MCL:5.3%; LCL: 6.3%) and eosinophils (MCL:8.2%; LCL:5.2%). While the smear from skin ulcers of LCL patients showed 45.3% of neutrophils and 43% lymphocytes. All of these indicate that neutrophils might play a role in the MCL immunopathogenesis. Moreover, an increased serum levels of IL-8 (P=0.03) were found in LCL (18.5ρg/mL) and MCL (18.2ρg/mL) patients, suggesting that this cytokine promotes the recruitment of neutrophils to the infection site in MCL; while in LCL patients may contribute with CD4 + Th1 (IFN-γ) cells in the activation of leishmanicida mechanisms. <![CDATA[<b>Biochemical and biological characterisation of lancehead (<i>Bothrops venezuelensis </i>Sandner 1952) snake venom from the Venezuelan Central Coastal range</b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1690-46482014000200004&lng=pt&nrm=iso&tlng=pt Se aislaron fracciones del veneno de Bothrops venezuelensis que demuestran ser un espectro abundante de proteínas con actividades variadas (coagulante, hemorrágica, fibrinolítica, proteolítica y de función plaquetaria), para el análisis de sus propiedades físico-químicas y biológicas, el veneno fue fraccionado por cromatografía de exclusión molecular, corrido en una electroforesis en gel y realizada una batería de ensayos biológicos. La DL50 del veneno de B. venezuelensis fue 6,39 mg/kg de peso corporal, fue determinada inyectando intraperitonealmente en ratones, diluciones seriadas de veneno de B. venezuelensis. Se colectaron doce fracciones a partir del veneno de B. venezuelensis mediante cromatografía de exclusión molecular. Las fracciones 1-5 y 7-9 tenían actividad hemorrágica. Todas las fracciones, con la excepción de las fracciones 3 y 6, tenían actividad fibrinolítica. Ninguna de las fracciones tuvo actividad de gelatinasa significativa, y sólo fracciones 4-6 demostraron actividad en polvo azul de ocultamiento. Con la excepción de las fracciones 1 y 4 , todas hidrolizaron la cadena β de la insulina. Cada fracción del veneno, así como el veneno crudo mostraron actividad procoagulante, cuando se probó en un analizador Sonoclot. Las fracciones 1, 3 , 5 y 9 inhibieron la función plaquetaria. En este estudio se señalan actividades biológicas de un veneno poco estudiado (B. venezuelensis) y sus fracciones. Al detectar actividades hemorrágicas, fibrinolíticas, procoagulantes, proteolíticas y de inhibición de la función plaquetaria. Este estudio preliminar abre el camino para la identificación de moléculas específicas que podrían tener potencial terapéutico en hemostasia y cáncer, que vienen siendo estudiados en nuestro grupo.<hr/>Venom fractions isolated from Bothrops venezuelensis were shown to contain a broad spectrum of proteins with varied activities. This study describes venom fractions with coagulant, haemorrhagic, fibrinolytic, proteolytic and antiplatelet activities, and analyses their physico-chemical properties and biological activities via molecular exclusion chromatography, gel electrophoresis and a bioassay battery. The LD50, determined by injecting intraperitoneally serial dilutions of B. venezuelensis venom into mice, was 6.39 mg/kg body weight. Twelve fractions were collected from B. venezuelensis venom using molecular exclusion chromatography. Of these, fractions 1-5 and 7-9 showed haemorrhagic activity, and all fractions except 3 and 6 showed fibrinolytic activity. However, none of the fractions had significant gelatinase activity, and only fractions 4-6 demonstrated activity on hide powder azure. With the exception of fractions 1 and 4, all fractions hydrolysed the insulin B-chain. In addition, all fractions as well as the crude venom showed strong procoagulant activity when tested using a Sonoclot Analyzer. Fractions 1, 3, 5 and 9 inhibited platelet function. In this study we have described the activities of the crude venom and its size-fractions from the scarcely studied B. venezuelensis. Haemorrhagic, fibrinolytic, procoagulant and proteolytic activities, and the inhibition of platelet function were detected. This preliminary study paves the way for the identification of specific molecules in B. venezuelensis venom that could have therapeutic potential for cancer and aberrant haemostasis treatment. <![CDATA[<b>Pathogens parasites in sand beach and it´s relation to environmental conditions in a beach from Puerto Cabello, Venezuela, 2012-2013</b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1690-46482014000200005&lng=pt&nrm=iso&tlng=pt Venezuela, presenta una gran variedad de playas frecuentadas durante todo el año, por lo que legalmente se establece un monitoreo permanente de niveles de contaminación para su clasificación "apta" o no para bañistas. Sin embargo, esto no contempla la evaluación parasitológica en la arena de playa como criterio para la referida clasificación. Por ello, la presencia de sólo una forma parasitaria patógena indica que existe contaminación fecal dado por personas, animales o acúmulo de basura a lo largo del balneario; además de la influencia de las características geográficas y ambientales en el desarrollo de los parásitos. Esta investigación tuvo como objetivo determinar la presencia de formas parasitarias patógenas para el hombre como indicadores de contaminación fecal en un balneario en Puerto Cabello. Se recolectaron 110 muestras de arena durante un año en diferentes puntos geográficos seleccionados intencionalmente abarcando la extensión de la ensenada, registrándose: temporadas pre o post vacacionales, humedad relativa de la arena, punto geográfico de muestreo y estación climática. Se realizaron los métodos de Lavado con solución salina 0,85%, Rugai modificado y Willis. Resultados: 25% de muestras fueron positivas para parásitos patógenos, distribuyéndose: larvas rabditoides (8,33%) y filarioides (2,08%) de Strongyloides spp., huevos (2,08%) y larvas rabditoides (12,49%) de Anquilostomideos, huevo de Toxocara spp. (4,17%) y Ooquiste de Isospora belli (2,08%), poniendo en evidencia la contaminación fecal de origen animal y humano. Se espera que investigaciones como éstas fomenten la elaboración de normativas de control sanitario y programas de evaluación de niveles de contaminación parasitaria en arena de playa.<hr/>A large number of beaches in Venezuela are visited throughout the year, and there is a legal system in place whereby these beaches are monitored and classified as suitable or not for bathing. However, the presence of parasites in the sand on the beaches is not evaluated as part of this classification The presence of only one pathogenic parasite on a beach indicates fecal contamination either by humans, animals or the accumulation of trash, in addition to geographical and environmental characteristics which could influence the development of the parasites. The aim of this study was to determine the presence of parasites pathogenic to humans as indicators of fecal contamination on a beach in Puerto Cabello. A total of 110 sand samples were collected over one year at different geographic locations selected to cover the entire bay area. Samples were registered as collected during the pre or post-holiday seasons, and the relative moisture of the sand, geographic location and season were also noted. Parasites were collected by washing with 0.85% saline solution, and tested using the Rugai method modified by Willis. Overall, 25% of samples showed positive for the parasites as follows: Strongyloides spp.: rhabditoid (8.33%) and filarioid (2.08%) larvae, Ancylostoma spp.: eggs (2.08%) and rhabditoid larvae (12.49%), Toxocara spp.: eggs (4.17%) and Isospora belli: oocysts (2.08%), indicating extensive fecal contamination from both human and animal sources. We hope that investigations such as this will lead to the establishment of hygiene standards and programs for monitoring the levels of parasitic contamination on sandy beaches. <![CDATA[<b><i>Clinical and epidemiological aspects of intestinal coccidian infection in Urumaco, Falcon state, Venezuela</i></b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1690-46482014000200006&lng=pt&nrm=iso&tlng=pt Entre septiembre-diciembre de 2013, se realizó un estudio para determinar los aspectos clínicos y epidemiológicos de la infección por coccidios intestinales en 187 habitantes (X ± D.S=25,74 ± 19,73 años) de Urumaco, Estado Falcón, Venezuela. Se hizo evaluación clínica y anamnesis para la búsqueda de signos y síntomas que usualmente se encuentran asociados a las coccidiosis intestinales. El diagnóstico parasitológico se hizo mediante los métodos directo y coloración de Kinyoun. La prevalencia global de parasitosis intestinales fue de 78,07% (146/187), siendo Blastocystis spp. el taxón más frecuentemente observado (57,07%). La prevalencia de coccidiosis intestinal fue de 50,80% (95/187), y la criptosporidiosis la más prevalente con 40,64%. No se encontró una relación estadísticamente significativa entre el sexo o la edad y la presencia de los coccidios intestinales (P>0,05). Hiporexia, epigastralgia y flatulencia fueron las manifestaciones clínicas características significativamente relacionadas con estas infecciones (P ≤ 0,05). Presentar uñas sucias (Odds Ratio [OR]=1,33), el consumo de "comida rápida" (OR= 2,11), andar descalzo (OR=1,16), pertenecer al nivel socioeconómico IV y V (OR= 2,89), que merodeen animales ajenos en las viviendas (OR= 1,01), que no los amarren (OR= 2,79), su alimentación con desperdicios (OR= 1,9) y sin suministro de drogas antiparasitarias (OR= 3,54), aparecen como potenciales factores de riesgo significativamente asociados en la dinámica de transmisión de las coccidiosis intestinales.<hr/>We investigated clinical and epidemiological aspects of intestinal coccidial infections in 187 inhabitants (X ± S.D.=25.74 ± 19.73 years) in the rural community of Urumaco, Falcón state, Venezuela, between September and December 2013. Direct smears and Kinyoun staining were used to perform parasitological diagnosis. The overall prevalence of intestinal parasitoses was 78.07% (146 /187), with Blastocystis spp. the most frequently parasite taxon observed (57.07 %). The prevalence of coccidioses was 50.80 % (95 /187), and among these criptosporidiasis was the most prevalent with 40.64% of patients infected. There were no statistically significant differences in parasite prevalence between sexes or ages (P>0.05). Hyporexia, epigastralgia and flatulence were significantly associated with the infections (P< 0.05). Dirty fingernails (Odds Ratio [OR]=1.33), fast food consumption (OR= 2.11), walking barefoot (OR=1.16), belonging to a lower socioeconomic level (OR= 2.89), animals roaming freely in the houses (OR= 1.01), untied animals (OR= 2.79), feeding animals with food waste (OR= 1.9) and a lack of anti-parasitic treatment (OR= 3.54) were determined as risk factors significantly associated with the transmission dynamics of enteric coccidiosis. <![CDATA[<b><i>A</i></b><b><i>chatina fulica </i></b><b>Bowdich, 1822 (Mollusca, Gastropoda, Achatinidae) carrier of Helminthes, Protozoa and Bacteria in northeast Venezuela</b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1690-46482014000200007&lng=pt&nrm=iso&tlng=pt Por cuanto el molusco Achatina fulica nativo del África es vector de helmintos, pero su relación con protozoarios y bacterias es poco conocida, decidimos estudiar las excretas de 1.200 ejemplares capturados en los estados Anzoátegui, Monagas, Sucre y Nueva Esparta, del noreste de Venezuela. Su moco pedal y heces mostraron infección por los protozoarios Chilomastix spp., Trichomonas spp., Giardia spp., Balantidium spp., Entamoeba spp., Iodamoeba spp., Blastocystis spp. Y por los helmintos de los grupos Ascarioidea, Trichuroidea, Ancylostomatidae y Cestoda. El moco céfalopodal mostró únicamente larvas de Rhabditida. Las bacterias Citrobacter freundii, Escherichia coli, Klebsiella pneumoniae, K. azaenae, Aeromonas hydrophila, Acinetobacter baumannii, Pseudomonas aeruginosa, Campylobacter spp. infectaron a las tres excretas. Los mecanismos de transmisión y la composición de las excretas, como nichos fisiológicamente apropiados para los organismos encontrados, son discutidos en relación con el riesgo epidemiológico que el molusco representa en salud pública y veterinaria.<hr/>The mollusk Achatina fulica, native to Eastern Equatorial Africa, has been incriminated as a carrier or vector of helminthes. Nevertheless, information in the literature as regards its status as a carrier for bacteria is scarce, and we could find no reference at all for its relation to protozoa. We studied microscopically the excreta from 1200 snails captured in Anzoátegui, Monagas, Sucre and Nueva Esparta states, in northeast Venezuela. The pedal mucus and feces were infected by the protozoa Chilomastix spp., Trichomonas spp., Giardia spp., Balantidium spp., Entamoeba spp., Iodamoeba spp., Blastocystis spp., as well as helminthes of Ascarioidea, Trichuroidea, Ancylostomatidae and Cestoda groups. The only helminthes found in the cephalopodal mucus were Rhabditida larvae. The three excreta were also infected by the bacteria: Citrobacter freundii, Escherichia coli, Klebsiella pneumoniae, K. azaenae, Aeromonas hydrophila, Acinetobacter baumannii, Pseudomonas aeruginosa, Campylobacter spp. Risk of infection and transmission mechanisms as well as the composition of the excreta as appropriate physiological niches for the organisms mentioned, are discussed with regard to the epidemiological importance of this snail for in human and veterinary health. <![CDATA[<b><i>Abundance and species diversity of Culicinae (Diptera: Culicidae) from Alto Orinoco, Amazonas, Venezuela</i></b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1690-46482014000200008&lng=pt&nrm=iso&tlng=pt Se realizó un estudio longitudinal en 18 comunidades o shabonos de las localidades Yanomami Ocamo y Mavaca del municipio Alto Orinoco entre Junio 1998 y Diciembre 2000 para caracterizar la fauna de culicinos. Se colocaron trampas CDC dentro de viviendas entre 1900 y 0600 horas a fin de capturar los mosquitos atraídos por personas protegidas por mosquiteros. Se capturaron un total de 4.635 culicinos pertenecientes a nueve géneros y 25 especies. Del total de especies identificadas, 18 especies (72%) constituyen nuevos registros para el estado Amazonas. Las especies más abundantes fueron Mansonia (Mansonia) titillans (48%), Aedes (Ochlerotatus) fulvus (16,9%) y Culex (Melanoconion) spissipes (12%). Se observaron correlaciones negativamente significativas entre precipitación, nivel del río y abundancia de Ma. titillans y Ae. fulvus. La abundancia y diversidad de especies resultó diferente entre comunidades. La mayor diversidad y abundancia se encontró en las comunidades Santa María de los Guaicas y Carlitos. El esfuerzo de captura, disponibilidad de criaderos y diversidad de hábitats en estas comunidades influye en esta diferencia con respecto a las otras comunidades. El presente constituye el primer y único estudio longitudinal realizado en el estado Amazonas para caracterizar la fauna de culicinos, la cual incluye especies de importancia en salud pública.<hr/>A longitudinal study was conducted in 18 communities or "shabonos" in the Yanomami inhabited Ocamo and Mavaca regions in the Alto Orinoco municipality between June 1998 and December 2000. CDC light traps were placed inside dwellings to catch mosquitoes attracted to humans protected by mosquito nets between 1900 and 0600 hours. A total of 4.635 culicines belonging to 9 genera and 25 species were captured. Of the total number of species identified, 18 (72%) are new reports for Amazonas state. The most abundant species were Mansonia (Mansonia) titillans (48%), Aedes (Ochlerotatus) fulvus (16.9%) and Culex (Melanoconion) spissipes (12%). There were significantly negative correlations between rainfall and river levels, and the abundance of Ma. titillans and Ae. fulvus. The abundance and species diversity of culicines differed among communities, with the highest values of both these parameters found in Santa María de los Guiacas and Carlitos. These differences were partly due to sampling effort but could also have been produced by the availability of larval habitats and niche diversity in these communities compared to the others surveyed. Up until, now this is the only longitudinal study to characterize the culicine fauna in Amazonas state, which includes several species of public health importance. <![CDATA[<b><i>Larvicide effect of methanolic extracts from seeds and leaves </i></b><b>Persea americana <i>(Laurales: Lauraceae) (avocado) on </i>Aedes aegypti <i>(Diptera: Culicidae)</i></b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1690-46482014000200009&lng=pt&nrm=iso&tlng=pt Aedes aegypti es el principal vector del dengue en el continente americano, una de las arbovirosis de mayor impacto en Venezuela, debido a su alta morbilidad y mortalidad. La ausencia de una vacuna contra este virus ha llevado al uso indiscriminado de insecticidas provocando la resistencia del vector, daños al medio ambiente y reacciones adversas en personas susceptibles. En un esfuerzo por encontrar formas eficaces y asequibles para el control, surge como una alternativa el uso de extractos vegetales que proporcionen menos riesgo ambiental y bajo costo. El objetivo de esta investigación fue evaluar el efecto larvicida de extractos metanólicos obtenidos a partir de semillas y hojas de Persea americana (aguacate) sobre Ae. aegypti [cepas Rockefeller y Mario Briceño Iragorry (MBI)]. El mayor efecto tóxico se observó con el extracto metanólico obtenido a partir de las semillas con 100% de mortalidad a 25 mg/L para la cepa Rockefeller y 50 mg/L para la cepa MBI, 24 horas post-exposición. Las concentraciones letales (CL50) fueron las siguientes: a) extracto metanólico preparado a partir de las semillas CL50= 5,7mg/L para Rockefeller y CL50=9,9 mg/L para MBI; b) extracto metanólico de las hojas CL50= 22,8mg/L para Rockefeller y para MBI CL50= 26,2 mg/L. Los resultados muestran el efecto tóxico de los extractos metanólicos preparados a partir de la semilla y de la hoja del aguacate, sobre Ae. aegypti y sugieren la potencialidad de estos productos como agentes de control químico.<hr/>Aedes aegypti is the main vector of dengue in the Americas. This disease is one of the most important arboviruses in Venezuela due to the high morbidity and mortality it produces. The absence of a vaccine against this virus has led to the indiscriminate use of insecticides causing vector resistance, environmental damage and adverse reactions in susceptible individuals. The search for effective and inexpensive ways to control dengue has led to the evaluation of plant extracts as a low environmental risk, economical alternative. The aim of this study was to assess the larvicidal effect of methanol extracts obtained from the seeds and leaves of Persea americana (avocado) on Ae. aegypti [Rockefeller and Mario Briceño Iragorry (MBI) strains]. The methanol extract obtained from the seeds proved to be the most toxic for Ae. aegypti, producing 100% mortality 24 hours post-exposure, at concentrations of 25 mg/L (Rockefeller strain) and 50 mg/L (MBI strain). The lethal concentrations (LC50) were: a) methanol seed extract LC50 = 5.7 mg/L (Rockefeller strain) and 9.9 mg/L (MBI strain), b) methanol leaf extract LC50 = 22.8 mg/L (Rockefeller strain) and 26.2 mg/L (MBI strain). These results demonstrate the toxicity of methanol extracts from avocado seeds and leaves for Ae. aegypti and suggest that these products have potential as chemical controls of this vector. <![CDATA[<b><i>Effect of the growth regulator pyriproxyfen on </i></b><b>Aedes aegypti <i>(Linnaeus, 1762) (Diptera: Culicidae) from La Pedrera, Maracay, Aragua, Venezuela</i></b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1690-46482014000200010&lng=pt&nrm=iso&tlng=pt Pyriproxyfen es un análogo sintético de la hormona juvenil ampliamente usado para el control de mosquitos. Esta investigación describe bioensayos realizados en condiciones de laboratorio con una formulación granulada del producto (Sumilarv G-0,5 %), utilizando larvas del IV instar temprano de Aedes aegypti. Se realizaron estos bioensayos con el fin de determinar su eficacia y actividad residual a las dosis de 0,01; 0,02; 0,03; 0,04 y 0,05 ppm sobre larvas Aedes aegypti de las cepas La Pedrera y Rockefeller de referencia; las evaluaciones fueron realizadas a 1, 7, 15, 30, 45, 60, 75 y 90 días después del tratamiento. Los resultados evidenciaron que especímenes tratados, presentaron malformaciones que afectaron su desarrollo y causaron la muerte de los individuos. Fue calculado el porcentaje de mortalidad de larvas, pupas y adultos, asimismo el porcentaje de inhibición de emergencia de adultos. La media aritmética para el porcentaje de mortalidad (% M) en la cepa Rockefeller fue de 76,83 % y para la cepa La Pedrera de 65,37 %. Para el porcentaje de inhibición de la emergencia, la cepa Rockefeller presentó media aritmética de 91,80 %; la cual fue significativamente mayor que la de la cepa La Pedrera, que fue igual a 78,08 %. El producto pyriproxyfen mostró eficacia en la inhibición de la emergencia de adultos con las dosis evaluadas, pero para la dosis de 0,01 ppm fue de menor efecto residual. Los resultados demuestran una mayor eficacia y actividad residual del pyriproxyfen a las dosis de 0,04 y 0,05 ppm hasta los 90 días después del tratamiento; en efecto ambas dosis presentaron elevada eficacia y actividad residual prolongada por 90 días, sugiriendo su uso como alternativa en los programas de control de Aedes aegypti y para el manejo integrado de otros mosquitos vectores en Venezuela, como es el caso de Aedes albopictus.<hr/>Pyriproxyfen is a synthetic juvenile hormone analogue widely used in mosquito control. Bioassays were undertaken to evaluate the efficacy and residual activity of a granular formulation of pyriproxyfen (Sumilarv G- 0.5% in granules) against IV instar Aedes aegypti (La Pedrera and Rockefeller reference strains), at doses of 0.01; 0.02; 0.03; 0.04 and 0.05 ppm. Larvae were examined 1, 7, 15, 30, 45, 60, 75 and 90 days after treatment. The treated specimens showed malformations affecting their development, and higher mortality rates compared to the controls. The percentage mortality of larvae, pupae and adults and the percentage of adult emergence inhibition were calculated. Mortality rates (% M) were 76.83 % (Rockefeller strain) and 65.37 % (La Pedrera strain). The percentage of adult emergence inhibition was significantly higher (91.80 %) in the Rockefeller strain than the La Pedrera strain (78.08 %). Pyriproxyfen produced effective inhibition of adult emergence at all the doses evaluated, but with less residual effect at 0.01 ppm. Doses of 0.04 and 0.05 ppm were the most efficient and persistent over the 90 days of the experiment. The results suggest that pyriproxyfen could be used as an alternative in Aedes aegypti control strategies and integrated mosquito vector management programs in Venezuela. <![CDATA[<b>Proposals for improving the handling of hazardous substances in laboratories at the Department of Biology, Faculty of Science and Technology, Carabobo University</b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1690-46482014000200011&lng=pt&nrm=iso&tlng=pt El presente trabajo de investigación tiene como finalidad proponer el manejo adecuado de sustancias peligrosas en los laboratorios del Departamento de Biología de la Facultad de Ciencia y Tecnología de la Universidad de Carabobo, de acuerdo a la norma ambiental vigente. El tipo de investigación que se implementó fue documental y de campo. El tiempo de recolección de datos fue de cuatro meses y las herramientas utilizadas para la recolección de los mismos fueron la entrevista, la aplicación de encuesta y el muestreo; se trabajó con una muestra de 54 estudiantes y un técnico y un profesor para cada laboratorio. Se caracterizaron los efluentes una vez tomadas las muestras y también luego de la aplicación de los tratamientos fisicoquímicos (oxidación química para remover la DBO y la DQO) según el parámetro en estudio. Luego de la recolección de datos y el análisis de los resultados se determinó que los laboratorios no están tomando en cuenta un plan de manejo de las sustancias peligrosas que se deben manipular de acuerdo a lo establecido por la normativa legal vigente (Decreto 2635) y que algunos de los efluentes no cumple con los límites máximos permisibles para las descarga a redes cloacales según el Decreto 883. Finalmente se propone mejorar el plan de manejo de sustancias peligrosas mediante el acondicionamiento del depósito de desechos, la elaboración de hojas de seguridad y etiquetas para todos los reactivos, la señalización adecuada dentro de las instalaciones, la incineración de los desechos almacenados en el deposito destinado para ello y el reúso del formol y etanol.<hr/>The aim of this study was to propose ways to ensure the correct handling of hazardous substances in laboratories at the Department of Biology, Faculty of Science and Technology, Carabobo University. Both documentary and field data were collected. The survey was completed in four months and consisted of interviews, questionnaires and sampling. A total of 54 students, and one technician and one professor for each laboratory were surveyed. Samples were taken of all effluents and these were characterized before and after physiochemical treatments were implemented (chemical oxidation to remove BDO and CDO) when applicable. The laboratories surveyed did not follow pre-set procedures for the handling of hazardous substances, which should be manipulated according to the provisions of the current legislation (Decree No. 2635). In addition, the concentrations of some of the effluents were higher than the maximum permitted limits for discharge into the sewage system according to Decree No 883. The handling of hazardous substances could be greatly improved by: refurbishing the waste reservoir, drawing up safety data sheets, labelling chemical substances, the use of adequate signage within the facilities, incinerating waste materials stored in the designated warehouse and the reuse of formaldehyde and ethanol. <![CDATA[<b><i>Spatial and temporal evolution of malaria in the municipality of Sifontes, Bolívar State, Venezuela. 1980-2013</i></b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1690-46482014000200012&lng=pt&nrm=iso&tlng=pt La malaria es endemo-epidémica en el estado Bolívar, con brotes epidémicos frecuentes debido a causas naturales y antrópicas. En el municipio Sifontes, ubicado en el noreste del estado, durante el último decenio se ha registrado un aumento considerable de los casos, cerrando 2013 con 46.610 casos, 71% % de la malaria del estado y 61% del país. Este es un foco meso-endémico de malaria inestable donde la transmisión ocurre durante todo el año, con brotes epidémicos estrechamente relacionados con la minería aurífera. A partir de 1983, gran parte de la malaria en Venezuela depende de la generada en el estado Bolívar, y esta a su vez de la generada en el municipio Sifontes. La revisión de la data de malaria del estado en el periodo 1980-2013 reveló la existencia de dos niveles epidemiológicos, uno basal y otro adicional, este último ligado a la minería aurífera; sin embargo, no se encontró correlación con la lluvia, para el nivel basal (R = 0,41, P= 0,78) ni para el nivel epidémico (R = 0,41; P = 0,32). Asimismo, la proyección geográfica de los datos reveló la naturaleza focal de la enfermedad en el municipio, el cual reúne todas las condiciones epidemiológicas, ambientales, climáticas y sociales para ser considerado un foco caliente. Las características espaciales y temporales de la malaria en el municipio Sifontes lo configuran como el foco de malaria más importante en Venezuela, el cual debe ser priorizado en los planes de control de la enfermedad.<hr/>Malaria is endemo-epidemic in Bolívar state with frequent epidemic outbreaks occurring due to both natural and anthropic factors. The Sifontes municipality, located in the northeastern corner of the state, has reported a considerable increase in the number of cases over the last decade; with 46.610 cases registered during 2013. This represents 71% of the number of malaria cases reported for the state and 61% for the country in this same year. Sifontes municipality is a meso-endemic focus of unstable malaria, and transmission occurs throughout the year with epidemic outbreaks tightly linked to gold mining activities. Since 1988 the increase in the number of cases reported in Venezuela has been determined by the number of cases reported in Bolívar state which, in turn, is correlated with the number of cases reported in Sifontes. A review of malaria data between 1980 and 2013 reveals the existence of two epidemiological categories: a base level and an additional level, the latter associated with gold mining; nevertheless, there was not correlation whit the rainfall for basal (R = 0,41, P= 0,78) or additional level (R = 0,41; P = 0,32). In addition, a geographic plotting of the data revealed the focal nature of malaria in the municipality, which to gather all environmental, climatic, social and epidemiological factors that have produced conditions typical of a hot spot. The spatial and temporal characteristics of malaria in the Sifontes municipality demonstrates that this is the most important malaria focus in Venezuela, and must be prioritized when designing strategies for the control of this disease. <![CDATA[<b><i>New records of potential yellow fever and mayaro vectors from Guarico State, Venezuela</i></b>: <b><i>First record of </i></b><b>Haemagogus janthinomys <i>Dyar from the central plains of Venezuela</i></b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1690-46482014000200013&lng=pt&nrm=iso&tlng=pt Se reportan Haemagogus anastasionis Dyar y Hg. equinus Theobald, con base en capturas realizadas en un área rural del municipio Roscio, estado Guárico y se señala por primera vez la presencia de Hg. janthinomys para la subregión de los Llanos Centrales. Este hallazgo, constituye un factor de riesgo potencial en la emergencia y transmisión de arbovirosis, por lo que se recomienda a los organismos de salud del estado realizar la vigilancia entomológica y epidemiológica respectiva en el área.<hr/>Haemagogus anastasionis Dyar and Hg. equinus Theobald were captured from a rural area in the Roscio municipality, Guarico State, Venezuela, and Hg. janthinomys was reported for the first time from the central lowland plains subregion of Venezuela. These findings represent potential risk factors for the emergence of arboviruses and their transmission.
