Flavicarpa (maracuyá). Los frutos fueron obtenidos en el municipio de Rivera-Huila y su análisis fitoquímico inicial dejó ver abundante contenido de taninos, pigmentos antociánicos, al igual que constituyentes de naturaleza terpénica. Al evaluar la funcionalidad antioxidante se encontró que los compuestos de la cáscara y la semilla de cholupa poseen una ligera habilidad para captar radicales libres y bloquear en alguna etapa la cadena de oxidación, habilidad que se incrementó a medida que aumentó la concentración del extracto, alcanzando su máxima capacidad en un tiempo de 6 a 11 minutos. Se detectó una importante actividad antimicrobiana de los extractos etanólicos del fruto la cual fue máxima a la concentración de 10.000 µg·mL-1 y responde a la abundancia de metabolitos secundarios y a otros compuestos bioactivos y muestra a la cholupa con un alto potencial bacteriostático, siendo la semilla la parte de mayor actividad. La bacteria Klebsiella pneumoniae fue la cepa más sensible a la acción de los compuestos presentes en este fruto. Ni aun en sus máximas concentraciones los productos derivados de la cáscara y la semilla de esta pasiflora superaron el 10 % de toxicidad en humanos.<hr/>This study set out to recognize and evaluate the chemical and biological characteristics of the seeds, pulp (aryl) and peel (pericarp) of Passiflora maliformis (cholupa) from the South Colombian Massif, thus contributing to the determination of their bioactive and/or agro-industrial potential, in comparison to Passiflora edulis var. Flavicarpa (passion fruit). The fruits were obtained in the Municipality of Rivera-Huila, and their initial phytochemical analysis showed abundant content of tannins, anthocyanic pigments, and nature terpene constituents. When evaluating the antioxidant functionality it was found that the compounds of the peel and seed of cholupa have a slight ability to scavenge free radicals and block the oxidation chain, ability that increased as extract concentration increased, reaching the maximum capacity in a time of 6 to 11 minutes. It was revealed an important antimicrobial activity of the fruit ethanolic extracts which was maximum at the concentration of 10,000 µg·mL-1, and responds to the abundance of secondary metabolites and other bioactive compounds and shows the cholupa with high bacteriostatic potential, being the seed the part with greater activity. The bacterium Klebsiella pneumoniae was the most sensitive strain to the action of the compounds present in the fruit. Not even at their maximum concentrations, products derived from peel and seed of P. maliformis exceeded 10 % toxicity in humans. <![CDATA[Fractions of soil phosphorus under long term rice culture. I. Inorganic phosphorus]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612016000100002&lng=pt&nrm=iso&tlng=pt Los suelos cultivados con arroz, en el sistema de riego del Río Guárico en Venezuela, han sido fertilizados con fosfatos por muchos años. El conocimiento del estado actual de las formas de fósforo en los suelos, así como sus transformaciones en diferentes fracciones, podría permitir mejorar los métodos de diagnóstico de P actualmente en uso. Se tomaron muestras compuestas de suelo de 0 a 20 cm de profundidad, en 43 parcelas, y fueron llevadas al laboratorio para determinar las diferentes fracciones de P. El fósforo inorgánico (Pi) resina más Pi lábil, considerados disponibles, representaron sólo 2,7 a 5,5 % del Pi total (PiT); esto representa una pequeña parte del P aplicado al suelo. En cambio, la suma del moderadamente lábil (PiML) más el ligeramente lábil (PiLL) mostró una tendencia a disminuir con el tiempo y fue 7,1 a 9,3 veces mayor que las dos formas disponibles anteriores. Las formas no disponibles fueron las más abundantes, entre 64,2 y 72,8 % del PiT. Las fracciones Pi ocluida y Pi recalcitrante se incrementaron con el pH del suelo.<hr/>Soils cultivated with rice in the irrigation system of Guárico State, Venezuela, have been fertilized with phosphates for many years. The current state of knowledge of the forms of phosphorus in soils and their transformations into different fractions could lead to better diagnostic methods currently in use. Composite soil samples were collected from 0 to 20 cm deep in 43 farmer’s plots and the inorganic fractions (Pi) were determined in the laboratory. The Pi resin and Pi labile (both fractions considered available) represented only 2.7 to 5.5 % of total Pi (PiT), thus representing just a small portion of the P applied to the soil, whereas the moderately labile and slightly labile Pi were 7.1 to 9.3 times higher than the available fractions and showed a tendency to decrease with time. The unavailable forms were the most abundant (64.2 to 72.8 % of PiT). The occluded and recalcitrant Pi fractions increased with soil pH. <![CDATA[Postharvest behavior of uchuva fruits (<i>Physalis peruviana</i> L.): Effect of different doses and exposure times at 1-Methylcyclopropene]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612016000100003&lng=pt&nrm=iso&tlng=pt El cultivo de uchuva (Physalis peruviana L.) es una alternativa de producción para la economía de algunos países tropicales. El fruto es climatérico y se considera altamente perecedero con corta vida poscosecha. El objetivo fue evaluar el efecto de diferentes dosis y tiempos de exposición al 1-metilciclopropeno (1-MCP) sobre el comportamiento poscosecha de frutos de uchuva. Se utilizó un diseño experimental completamente al azar, con arreglo factorial de tratamientos de 3x3+1, donde el primer factor consistió en las dosis de 1-MCP (0,3; 1,0 y 3,0 µL·L-1) y el segundo factor correspondió al tiempo de tratamiento con 1-MCP (2, 12 y 24 h), más un testigo absoluto. Los frutos fueron almacenados a temperatura ambiente (16 °C) durante 15 días. Los resultados indicaron que a medida que se incrementó la dosis y el tiempo de exposición al 1-MCP, el efecto sobre la maduración de los frutos de uchuva fue mayor. La aplicación de 3 µL×L-1 de 1-MCP durante 24 h generó significativamente una menor producción de etileno, menor concentración interna de etileno, menor pérdida de firmeza, de peso y de acidez total titulable, en comparación con frutos testigo. El mencionado tratamiento también produjo menores cambios en el índice de color y en los sólidos solubles totales. Se concluye que el 1-MCP representa en una herramienta útil con potencial para conservar la calidad de los frutos de uchuva durante la etapa poscosecha.<hr/>The culture of cape gooseberry (Physalis peruviana L.) is a production alternative for the economy in some tropical countries. The fruit is climacteric and highly perishable with short postharvest life. The objective of this work was to evaluate the effect of 1-methylcyclopropene (1-MCP) at different doses and exposure times on the postharvest behavior of the fruit. A completely randomized design with treatments in a factorial arrangement 3x3 + 1 was used, where the first factor was the dose of 1-MCP (0.3, 1.0, and 3.0 µL·L-1) and the second factor the exposure time to the product (2, 12, and 24 h), plus a control (fruits without treatments). The treated fruits were stored at room temperature (16 °C) for 15 days and the results showed that as the dose and duration of exposure to 1-MCP increased the effect on ripening of cape gooseberry fruits was greater. The application of 3 µL·L-1 for 24 h resulted in a significantly decrease of ethylene production and internal ethylene concentration, less loss of firmness and weight, and lower total acidity compared to control fruits. This same treatment also produced minor changes in color index and total soluble solids. We conclude that 1-MCP represents a useful tool with potential for preserving quality of cape gooseberry fruits during postharvest. <![CDATA[Efecto de la aplicación de vermicompost en las propiedades biológicas de un suelo salino-sódico del semiárido venezolano]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612016000100004&lng=pt&nrm=iso&tlng=pt Los suelos con degradación química están ampliamente distribuidos en el mundo, y su recuperación puede basarse en la aplicación de enmiendas. El objetivo de este trabajo fue evaluar el efecto de un vermicompost sobre las propiedades biológicas de un suelo salino-sódico en el sector El Cebollal, Coro, Venezuela. Se realizó un experimento para comparar diferentes dosis de vermicompost empleando los siguientes cuatro tratamientos: un testigo sin vermicompost (T1); 1 % de vermicompost (T2); 5 % de vermicompost (T3) y 10 % de vermicompost (T4), los cuales fueron sometidos a incubación por 28 días. Las mediciones se realizaron a los 7, 14, 21, y 28 días y en cada una se evaluaron las variables de carbono orgánico (CO), carbono de la biomasa microbiana (C-BM), actividad ureásica (AU) e índice de carbono disponible (C-BM/CO*100). El CO aumentó en todos los tratamientos con vermicompost, con los valores más altos en el T4 (6,7 g·kg-1) y los más bajos en el testigo (2,5 g·kg-1). Asimismo, el C-BM resultó con incrementos significativos, presentando los mayores valores en el T4 (612,5 µg·g-1) y los menores en el testigo (36,4 µg·g-1). La AU aumentó significativamente con la adición del vermicompost. A los 28 días de incubación se encontraron valores de 67,8 µg NH4+g-1 para T4, lo cual representa un incremento de seis veces respecto al tratamiento testigo. La adición de vermicompost demostró ser una buena estrategia para la recuperación biológica de suelos salino-sódicos.<hr/>Chemical degradation of soils are widely distributed in the world, and their recovery can be based on application of amendments. The objective of this research was to evaluate the effect of vermicompost on the biological properties of a saline-sodic soil in the locality El Cebollal, in Coro, Venezuela. An experiment was performed to compare different doses of vermicompost using the following four treatments: (T1) a control, without vermicompost; (T2) 1 % vermicompost; (T3) 5 % vermicompost, and (T4) 10 % vermicompost. An incubation period of 28 days was applied, with measurements at 7, 14, 21, and 28 days. In each date the organic carbon (OC), microbial biomass carbon (MBC), urease activity (UA), and the index of available carbon (MBC/OC * 100) were evaluated. The OC increased in all vermicompost treatments, with the highest values ​​in T4 (6.7 g·kg-1) and the lowest in the control (2.5 g·kg-1). Likewise, the MBC resulted in a significant increase, being the highest also ​​in T4 (612.5 µg·g-1) and the lowest in the control (36.4 µg·g-1). The UA increased significantly with the addition of vermicompost. After 28 days of incubation we found values for NH4+​​ of 67.8 mg·g-1 in T4, which represents a six-fold increase as compared to the control treatment. The addition of vermicompost proved to be a good strategy for biological recovery of saline-sodic soils. <![CDATA[In vitro antifungal activity of essential oils of Ocotea quixos (Lam.) Kosterm. and Piper aduncum L.]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612016000100005&lng=pt&nrm=iso&tlng=pt Los hongos fitopatógenos ocasionan relevantes pérdidas de los cultivos agrícolas, tanto en fase de campo como en poscosecha. Se evaluó el uso de aceites esenciales extraídos de plantas amazónicas, en diferentes concentraciones, para el control in vitro de estos hongos. Los aceites esenciales se obtuvieron por destilación en arrastre de vapor a partir de dos especies vegetales (Ocotea quixos y Piper aduncum) colectadas en la provincia amazónica de Pastaza, Ecuador. La actividad antifúngica se evaluó mediante el método de la difusión en agar sobre los hongos fitopatógenos Aspergillus oryzae, Cladosporium cladosporioides, Fusarium solani, Rhyzopus stolonifer, Moniliophthora roreri y Phytophthora sp. El aceite esencial de O. quixos usado en su máxima concentración (500 µL·mL-1) inhibió el crecimiento de todas las cepas alcanzando 94 % de inhibición promedio y registrando un comportamiento similar al aceite esencial de tomillo (Thymus vulgaris), el cual fue utilizado como patrón de comparación. El aceite esencial de P. aduncum, también en su máxima concentración mostró los mayores porcentajes de inhibición frente a F. solani (94 %) y Phytophthora sp. (91 %). Los resultados sugieren el empleo de estos aceites esenciales como agentes de control biológico de hongos fitopatógenos.<hr/>The phytopathogenic fungi cause relevant crop losses, both under field and post-harvest conditions. The use of essential oils extracted from Amazonian plants, at different concentrations, was investigated, in order to evaluate in vitro properties against those fungi. Essential oils were obtained by steam distillation, from two plant species (Ocotea quixos and Piper aduncum) collected in the Amazonian province of Pastaza, Ecuador. The antifungal activity was evaluated through the agar diffusion method against the Aspergillus oryzae, Cladosporium cladosporioides, Fusarium solani, Rhyzopus stolonifer, Moniliophthora roreri and Phytophthora sp. The O. quixos essential oil, in its maximum concentration (500 µL·mL-1), inhibited the growth of all strains, reaching an average of 94 % inhibition rate, similar to the essential oil of thyme (Thymus vulgaris), which was used as standard. The P. aduncum essential oil, also in the maximum concentration, showed the best inhibition percentage against F. solani (94 %) and Phytophthora sp. (91 %). These results suggest the use of both essential oils as agents of biological control of phytopathogenic fungi. <![CDATA[<b>Adaptation</b><b> of RT</b><b>-PCR </b><b>technique</b><b> for the diagnosis of</b><b> potato </b><b>yellow vein virus</b><b> in Venezuela</b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612016000100006&lng=pt&nrm=iso&tlng=pt El virus del amarillamiento de las venas de papa (PYVV) pertenece al género Crinivirus, familia Closteroviridae y es transmitido por la mosca blanca de los invernaderos (Trialeurodes vaporariorum). Es un problema importante para los países andinos debido a que ocasiona pérdidas en la producción, al causar reducción del número y tamaño de los tubérculos. Su diagnóstico, basado en la expresión de síntomas, es impreciso porque el virus puede infectar en forma asintomática a las plantas y el diagnóstico serológico se dificulta por la inexistencia de pruebas comerciales disponibles. El objetivo de este trabajo fue la adaptación de la técnica RT-PCR para el diagnóstico y posterior rastreo viral del PYVV en las zonas productoras de Venezuela. Se colectaron 10 muestras sintomáticas en los estados Táchira y Trujillo. La extracción del ARN se realizó utilizando el kit RNeasy Plant Mini (Qiagen), y se incorporó al proceso la ultracongelación del tejido (-60 °C) y adición de metabisulfito de sodio (0,05 mg·g-1). La retrotranscripción se ejecutó mediante el kit ImPron-II Reverse-Transcription System (Promega). En la RT-PCR se utilizaron iniciadores específicos para los genes de CP y CPm, probándose diferentes temperaturas de hibridación y extensión, así como varias mezclas de amplificación. La adaptación realizada a la RT-PCR permitió la amplificación de fragmentos específicos, con los cuales se confirmó la infección del virus en el 100 % de las muestras. Se demuestra por primera vez su presencia en Venezuela basados en técnicas moleculares. Estos resultados aportan un referencial tecnológico de diagnóstico que puede ser implementado para prevenir la dispersión del virus a partir del tubérculo-semilla.<hr/>Potato yellow vein virus (PYVV) belongs to the genus Crinivirus, family Closteroviridae, and is transmitted by the greenhouse whitefly (Trialeurodes vaporariorum). It is an important problem for Andean countries because it causes loss of production by reducing the number and size of tubers. The diagnosis based on symptom expression is inaccurate because it can infect plants asymptomatically, and serological diagnosis is hampered by the lack of commercial kits available in the country. The objective of this study was the adaptation of the RT-PCR technique for the PYVV diagnosis and subsequent viral tracking in producing areas. Ten symptomatic samples were collected in Táchira and Trujillo States, Venezuela. RNA extraction was performed using the RNeasy Plant Mini kit (Qiagen). Deep freezing of the tissue (-60 °C) and addition of sodium metabisulfite (0,05 mg·g-1) were incorporated as additional steps. The retrotranscripción was executed by using the kit ImPron-II Reverse-Transcription System (Promega). Specific primers to amplify CP and CPm genes were utilized, as well as different anneling and extension temperatures, and various amplification mixtures. Such adaptations allowed the amplification of specific fragments, and confirmed virus infection in 100 % of the samples. It is the first time that PYVV is diagnosed by molecular techniques in Venezuela, thus it represents a diagnostic procedure that can be implemented to prevent the spread of the virus from seed tuber. <![CDATA[<b>Hydraulic evaluation of porous cups</b><b> for suction irrigation systems</b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612016000100007&lng=pt&nrm=iso&tlng=pt La importancia de disminuir la cantidad de agua utilizada en la agricultura bajo riego intensifica la necesidad de implementar el uso de métodos de aplicación localizada del líquido. Con el objetivo de promover métodos para hacer más eficiente el uso del agua se diseñaron cápsulas porosas para riego elaboradas con un suelo arcilloso proveniente de Sanare, estado Lara, Venezuela. A los tratamientos barbotina (testigo T0), barbotina+arena (T1), barbotina+almidón (T2), barbotina+estiércol (T3) y barbotina+poliacrilamida (T4) se les evaluó la porosidad absoluta, conductividad hidráulica, bulbo de humedecimiento y potencial mátrico. La mayor porosidad absoluta se obtuvo en barbotina+almidón con 33,7 %, mientras que la conductividad hidráulica fue mayor en el tratamiento de barbotina+arena con 0,0139 cm·h-1. Las cápsulas mezcla de barbotina+arena presentaron mayor bulbo húmedo tanto horizontal como vertical con 19,3 cm y 18,0 cm respectivamente. El menor potencial mátrico al límite de la cápsula fue de 16,0 cb en barbotina+estiércol. Los materiales utilizados para la elaboración de las cápsulas permitieron obtener propiedades hidráulicas adecuadas como para considerar estos emisores como alternativa promisoria para su adaptación al riego por succión.<hr/>The importance of reducing the amount of water used in irrigated agriculture intensifies the need to implement the use of methods of localized application of the liquid. With the objective of making more efficient the use of irrigation water we designed porous cups made from a clay soil collected in the locality of Sanare, Lara state, Venezuela. Mixes of ceramic slips were prepared as follows: slip alone (control, T0), slip+sand (T1), slip+starch (T2), slip+manure (T3) and slip+polyacrylamide (T4). Porous cups made from each of the previous mixes were evaluated for total porosity, hydraulic conductivity, and humid bulb, and potential matric. The highest porosity was obtained in slip+starch with 33.7 %, whereas the hydraulic conductivity was highest in the slip+sand treatment with 0.0139 cm·h-1. The cups made from the slip+sand mixture had the highest wet bulb, both horizontally and vertically with 19.3 and 18.0 cm, respectively. The lowest matric potential at the cup limit was 16.0 cb in the slip+manure treatment. The materials used to manufacture the cups allowed the obtaining of appropriate hydraulic properties to consider these emitters as a promising alternative for adaptation to the suction irrigation systems. <![CDATA[<b>Incidence of witches’ broom disease (<i>Crinipellis perniciosa)</i> on yields of two agro-ecosystems of cocoa with different management conditions </b>]]> http://ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612016000100008&lng=pt&nrm=iso&tlng=pt El presente estudio evalúa la incidencia de la enfermedad escoba de bruja (Crinipellis perniciosa Stahel, Singer) sobre los rendimientos en dos agroecosistemas cacao (Theobroma cacao L.) en las fincas La Primavera y La Cayena, cada una con diferentes condiciones de manejo en el estado Carabobo, Venezuela. Se utilizó un diseño estadístico completamente al azar y se comparó, de acuerdo a la sintomatología observada, la incidencia de la enfermedad en los árboles seleccionados; se realizaron mediciones del número de frutos sanos por planta, peso del fruto y rendimiento de almendras secas por planta al año; la comparación entre las fincas para estas variables se llevó a cabo por la prueba t de Student. Los resultados evidenciaron que la enfermedad escoba de bruja tiene una mayor incidencia en la finca La Primavera, donde las prácticas agroecológicas se utilizan a menor escala en comparación con la finca La Cayena; además, se detectaron diferencias significativas (P≤0,01) para las variables productivas, a favor de la finca La Cayena. Se obtuvieron 53 y 40 frutos sanos por planta, 1,02 y 0,90 kg para el peso del fruto, y 0,87 y 0,45 kg·año-1 para el rendimiento de almendra seca por planta, para las fincas la Cayena y La Primavera, respectivamente.<hr/>This study evaluated the incidence of the disease known as witches´ broom (Crinipellis perniciosa Stahel, Singer) on the yield of cocoa (Theobroma cacao L.) in two agro-ecosystems (La Primavera and La Cayena), two farms with different conditions of management in Carabobo State, Venezuela. A completely randomized design was used and the incidence of the disease was evaluated according to the observed symptoms on selected trees. The number of healthy fruits per plant, mean fruit weight, and yield of dry almonds per plant were determined; the comparison between farms for these variables was carried out by the t-Student test. The results show that the witches’ broom disease has a higher incidence in La Primavera farm, which had fewer agro-ecological practices and lower technificatión level as compared to La Cayena farm. Also, significant differences (P≤0,01) were found for the productive variables, favoring La Cayena farm.
