It has been established that growth, progression and metastasis of solid tumors are processes angiogenesis-dependent (new blood vessels formation from preexistent capillaries). Several kind of premalignant and malignant lesions in the oral mucosa, has also been used as a model of tumor progression. In the present study we determined the vascular density, as a measure of the angiogenesis response, in groups of lesion with differents grade of dysplasia and squamous carcinomas, through the quantification of blood vessels identified with the von Willebrand Factor VIII by immunohistochemistry techniques. Results revealed an increase in the vascular density with the diagnosis severity. The vascular density (media ± standard deviation) was lower in the group of mild dysplasia (12.70 ± 3.04), with a progressive increase in the moderate dysplasia (21.62 ± 1.99), and furthermore significative in the group of severe dysplasia (59.50 ± 8.73) and squamous carcinoma (60.62 ± 8.74). However, no statistically significant differences (p= 0.05) were found in the vascular density either between mild and moderate dysplasia, or severe dysplasia and squamous carcinoma. These findings suggest that the onset of angiogenesis, possibly occur early in the development of the oral carcinogenesis, mainly in the severe dysplasia and carcinomas.
