Summary By the combination of in vitro culture technique and induction of mutations with rays, genetic variability was induced in regenerated sugarcane plants with the purpose of obtaining resistant genotypes to mosaic virus of sugarcane, Saccharum sp., beginning from varieties susceptible to the virus. Three varieties susceptible to mosaic virus (B6749, B7987 and PR62258) were selected and their calluses irradiated with gamma rays (0, 2, 4, 8 and 12 Kr) using a source of Co60. A total of 3.502 plants were regenerated. Plants were transferred to the greenhouse and inoculated five times with the B race of SCMV, prepared from leaves of a highly susceptible variety, C571. Sorghum variety Rio indicator plants were used as control. Eighty two percent of regenerated plants remained free of symptoms of mosaic and were transplanted to the field for further observation of natural infectation incidences. Of these 1.607 plants, 911 remained without symptoms. In this way 46% out of 82% of plants that survived artificial inoculation remained symptom less after being exposed to field conditions. Under field conditions, rust susceptibility was also observed in plants obtained from disease resistant varieties
