PERDOMO LEIVA, María Cecilia et al. Advances on isoenzyme molecular fingerprinting of rice genotypes with intelectual property right purposes. Agronomía Trop. [online]. 2006, vol.56, n.4, pp.549-554. ISSN 0002-192X.
Variety characterization is usually performed using morphological descriptors. These descriptors are highly influenced by environmental conditions, and they poorly distinguish among closely related lines. As an alternative, biochemical characterization using isozymes polymorphism is a relative low-cost method better suited to characterize rice germplasm, particularly closely related rice lines. The objective of this research was to evaluate isozymes as molecular markers suited for fingerprinting of Venezuelan rice varieties. A set of ten rice varieties was screened. From each, five day-old coleoptiles were collected. The isozyme systems used were EST, ACP y SDH, on discontinued acrylamide gels. From the resulting zymograms, a present/absent matrix was constructed. Grouping analysis was obtained using the UPGMA method, calculated from Jaccard coefficients. The dendrogram obtained only distinguished five of the varieties, indicating that more isozyme systems are required for molecular fingerprinting of Venezuelan rice varieties.
