Legumes in Venezuela cover an area of an approximately 180.000 km2, forming natural pasture savannas that act as a principal source of animal feeding and represents more than 60% of soils all over the country. Stylosanthes capitata Vog., has a high capacity to fasten nitrogen from the atmosphere, and very deep root systems that contribute with the fertility and stability of compact soils. However, the main disadvantage from these plants is their low sexual reproductive ratio, this has led to, the establishment of various in vitro regeneration processes which have been well developed and have permitted the production of big amounts of plants. Despite, all these advances this methodology, has not been evaluated regarding the stability of genetic regenerated plants of in vitro Stylosathes capitata Vog., which can be determinated by the use of molecular markers. In this research, genetic stability of Stylosanthes capitata Vog., was determined via RAPDs comparing DNA banding patterns of in vitro plants with mother plant. DNA extraction was made following Doyle and Doyle protocol, modified by Oropeza in 2001. For amplification, 9 arbitrary primers from the series OPC 15 to 23 (Invitrogen)were used, in a final volume of 25µl reaction. The results obtained demonstrated that all primers tested, produced sharp and reproducible bands originating DNA fragments ranging from 21220pb to 2000pb with a high genetic stability for regenerated plants from in vitro tissue culture.
