PICHARDO, Eglys et al. Detection of major viruses in potato tubers-sprouts and plantlets by DAS-ELISA technique. Agronomía Trop. [online]. 2013, vol.63, n.1-2, pp.49-55. ISSN 0002-192X.
Viral diseases decrease vigor and yield of the plants and reduce the possibilities to use as seed tubers. The aim of this work was to detect by DAS-ELISA, the virus associated to clones and varieties of potato in quality control of basic seed. The experiment was conducted in the plant laboratory of the Instituto Nacional de Investigaciones Agricolas (INIA), where 65 samples of tissue culture plants and tubers were analyzed using a kit from the Centro Internacional de la Papa (CIP). In samples of plantlets of Costanera, Maria Bonita, INIAFRI, Kennebec, Andinita Tibisay, Granola and clones 392634-21 and 392639-1 varieties, PVY, PVM, PVS and PLRV viruses were detected. Variety Granola three viruses (PVY, PLRV and PVS) at Maria Bonita two (PVM and PVY) was detected in one other materials. In the case of tubers of Diacol Capiro Granola and clone 392639-1; 393194-27 and 386328-7, the virus PVY, PVM, PVS and PLRV were detected; and dual infection was observed in clones 393194-27 with PVS and PVY virus and PLRV 386328-7 and PVS. Also infection rates of viruses ranging between 0 and 20% for tubers and 0 to 33.6% for plantlets were obtained. We conclude that at least some of these viruses: PVY, PVM, PVS and PLRV were detected in the different propagation materials, except for the varieties and clones: Hope, Atlantic, 392636-31, 393193-16 and 392636-4 that they did not show positive results.
