Abstract

The technique of cryopreservation of embryos by vitrification has acquired great relevance world-wide because of the easiness of the process, the low cost of the equipment, and the high levels of viability of the embryos after vitrification. Nevertheless, a great variability exists among the reported protocols, especially in relation to the steps of equilibration and cooling. In order to contribute to the design of a standard protocol of vitrification for murine embryos, the following experiments were conducted.  Experiment 1 studied the effect of cooling rate (fast and very fast), and the volume of the solution (2µL and 1µL) on the viability of murine embryos post vitrification. No effect of the volume of the solution (p>0.05) was observed, but  a significant (p<0.05) effect of the cooling rate was shown. Understanding that the best results (81.3% of viable embryos) were obtained when the Fast - 1µL protocol of vitrification was used. Experiment 2 evaluated the influence of the number of steps and the equilibration time on the viability of murine embryos, combining four step protocols of exposition to cryoprotectant (1, 2, 3 and 4 steps) and four times of equilibration (5, 10, 15 and 20 min). The highest rate of viability (p<0.05) was obtained with 2 steps-10min (93.8%). In conclusion, the present work gives details for the optimization of the vitrification protocol of murine embryos.
