RNA interference (RNAi) is a powerful new molecular tool to dowregulate the gene expression at posttranscriptional level. It is being used in reverse genetic to found functions of several genes and as a therapeutic device to knockout or knockdown altered or over-expressed genes, in gene therapy. RNA interference now provides a fast and easy strategy to depleting mRNA by introducing double-stranded RNA homologous to a particular cellular mRNA, leading to its sequence specific removing. In this review we summarized the basic mechanism for RNAi, some of the structural requirements needed in the siRNA to be able to produce an efficient and specific silencing and some effectors systems used to deliver them inside the cells or tissue. This review is written to divulgate some topics in Molecular Biology of actual medical interest.
