In intact microsomes, two flavones purified from Bauhinia megalandra leaves inhibit glucose-6-phosphatase in a concentration dependant way. Both flavones increased the glucose-6-phosphatase KM for glucose-6-phosphate without change in the VMAX. This result allowed to conclude that both flavones competitively inhibit the glucose-6-phosphate transporter (T1) without affecting the catalytic subunit nor the phosphate/pyrophosphate transporter (T2) of the glucose-6-phosphatase system. The flavones strongly inhibit the neoglucogenic capacity of liver slices incubated in Krebs-Ringer bicarbonate buffer supplemented with lactate and oleate saturated albumine. The inhibition of G-6-P transporter (T1) of hepatic microsomal G-6-Pase system, produced by both flavones might explain the decrease in the neoglucogenic capacity of the rat liver slices and, in turn, it could reduced the blood glucose levels in diabetic patients.
