Studies on the long-term regulation of pyruvate dehydrogenase complex (PDC) have shown that the total amount of enzyme changes in response to different diets. In this report, the levels of mRNA for E1a subunit of the complex were estimated by semi-quantitative RT-PCR. A high-sucrose fat-free diet produced a 40% increase in the levels for the mRNA after a week. After 14-21 days of diet the increase was only by 20%, but the addition of polyunsaturated fat from fish oil reduced the levels of mRNA to the values obtained from control animals in only a week. Regulation of the expression of this gene may be due to changes in the rate of transcription or on the stability of the mRNA transcribed, as has been shown for other enzymes related to the control of lipogenesis.
