Abstract

RIVERON ACOSTA, María Beatriz et al. Hydrolytic enzymes (DNAses, lipases and proteases) secreted by Cladosporium cladosporioides isolated from soil and its potential application in biotechnology. Rev. Soc. Ven. Microbiol. [online]. 2017, vol.37, n.2, pp.61-65. ISSN 1315-2556.

Microorganisms inhabit all types of soils and present an enormous metabolic diversity, which makes them an important source of bioproducts, among them, the enzymes. The objective of this work was to isolate from a sample of garden soil, at least one microorganism able to produce hydrolytic enzymes (lipases, proteases and with emphasis DNAses) and use it as a source of enzymes for the formulation of a detergent. A filamentous fungus was found, initially named F6, which produced such enzymes. Studies with extracts of each enzyme on their substrates (olive oil, casein and DNA extracted from different organisms) showed strong activity at 30 °C and pH 7.5, and stability when stored under refrigeration at 8 °C during two months. The F6 fungus by optical microscopic morphology was identified as Cladosporium cladosporioides and by sequencing the ITS region by the Sanger method. The importance of this unprecedented finding is the fact of obtaining the enzymes of interest from the same microorganism, which facilitates and saves culture conditions, incubation temperature and aeration content, among others, to develop an enzymatic detergent in the future.

Keywords : soil; DNAse; lipases; proteases; Cladosporium cladosporioides.
