Resumen
PARDO, Adriana et al. Evaluation of genetic stability using RAPD markers, in Billbergia rosea Hortus Ex Beer buds conserved in vitro. Bioagro [online]. 2008, vol.20, n.2, pp.97-104. ISSN 1316-3361.
Genetic stability in Billbergia rosea buds, obtained from stem tips subjected to five conservation regimes under minimal growth, was evaluated using RAPD techniques. Protocol was standardized to obtain RAPD bands for this species. For this proposal, DNA extraction, polymorphic DNA amplification and printing of generated bands were made. Dendrogram based in Jaccards similarity index and the principal components analysis showed three groups. Group I, buds kept in ¼ MS with or without sucrose were pooled with minimal similarity (0.91) compared to mother plants. Group II, all treatments with distilled water and ½ MS with sucrose were included with dissimilarity of 0.15 compared to previous group. Group III consisted of buds kept in ½ MS with similarity of 0.58 to both groups. Analysis revealed that buds kept in ¼ MS with sucrose, showed a electrophoretic profile similar to mother plant, thus indicating genetic stability. The results can be used as base to plant medium-term conservation strategies for Billbergia rosea.
