Abstract

Since the effectiveness of the different protocols for isolating DNA in sufficient quantity and quality may vary depending on the crop, it is necessary to standardize the extraction methods to ensure its simplicity and effectiveness. In this paper we propose a modification to the method of Risterucci et al. (2000), which was compared with other protocols to extract DNA from leaves of Cattleya spp. to select the most appropriate method to use for analyzing the genetic potential of the species through molecular markers such as RAPD and ISTR. With the proposed method the obtained DNA was similar to that extracted using a commercial kit, since both methods produced DNA in appropriate quantity and quality. The method offers advantages over other protocols, mainly because the low number of steps for obtaining DNA, ease of implementation and especially, high stability, which is favorable in cases where it is required to process many samples, reducing the possibility of accidental contamination.
