PONCE, Loida et al. Effect of montelukast on the concentration of inflammatory mediators in the murine gouty arthritis model. Salus [online]. 2016, vol.20, n.3, pp.12-17. ISSN 1316-7138.
Treatment of gouty arthritis partially controls the disease with significant side effects. Montelukast is a selective leukotriene receptor antagonist LTD4, useful in the treatment of allergic rhinitis and bronchial asthma. Previous studies have reported a potent anti-inflammatory effect in the murine gouty arthritis model. This study investigated the effect of MK on the concentration of cysteinylleukotrienes and leukotriene B4, interleukin 6 (IL-6) and interleukin 12 (IL-12) on inflammatory exudate induced by sodium monourate (SMU) in the air pouch model. Twenty one BALB/c mice were divided into three groups (n: 7), negative control of inflammation (A), positive control of inflammation (B), and experimental group (C). Before injection of MUS (5 mg / mL) into the air pouch, the C and B group received 1 mg / kg of oral montelukast and saline solution, respectively. The A group was treated with saline solution orally and in the air bag. At 12 hours after induction of the inflammatory process, the concentration of CistLT, LTB4, IL-6 and IL-12 in the exudate obtained from the air pouch was determined by ELISA. Results showed that montelukast significantly (p <0.05) decreased concentrations of LTB4, IL-6 and IL-12 in MUS-induced inflammatory exudation, and significantly increased (p <0.05) concentrations of CistLT. This suggests that the MK dose used has a powerful anti-inflammatory effect, and is potentially beneficial in the treatment of gouty arthritis.
