VASQUEZ DE RICCIARDI, Laura et al. Pharmacokinetic disposition of antimony species in dogs after a dose of meglumine antimonate (Glucantime®). Bol Mal Salud Amb [online]. 2008, vol.48, n.1, pp.27-33. ISSN 1690-4648.
The pharmacokinetic parameters species of antimony present in Glucantime® was studied, in four healthy dogs, following subcutaneous administration of single dose 90 mg/kg-1. Blood samples were analyzed by atomic absorption spectroscopy. After administration, the pentavalent antimony showed decay in two phases, half-life was 13.18 h, volume of distribution Vd of 30,65 Lkg-1, area under curve(0-24h) 48.25 μg. h mL-1 y area under curve(0-∞) 63.55 μg.h mL-1. The trivalent antimony showed one phase well distinguished, the half- life was 16.10 h, volume of distribution of 126.5 Lkg-1, area under curve(0-24h) 10.35 μg.h mL-1 and area under curve(0-∞) 17.35 μg.h mL-1. The systemic conversion of pentavalent antimony to trivalent antimony at 24 hours was 23.62%. The present study was limited by its small sample size, whatever the results obtained revealed a difference in the pharmacokinetics of pentavalent and trivalent antimony in dogs, this might explain, in part, the therapeutic efficacy differences found with separated species.
